Accumulating evidence has indicated that long non-coding RNAs(lncRNAs)play critical roles in the development and progression of cancers,including esophageal squamous cell carcinoma(ESCC).However,the mechanisms of lncRNAs in ESCC are still incompletely understood and therapeutic attempts for in vivo targeting cancer-associated lncRNA remain a *** RNA-sequencing analysis,we identified that LLNLR-299G3.1 was a novel ESCC-associated ***-299G3.1 was up-regulated in ESCC tissues and cells and promoted ESCC cell proliferation and *** of LLNLR-299G3.1 with ASO(antisense oligonucleotide)resulted in opposite ***,LLNLR-299G3.1 bound to cancerassociated RNA binding proteins and regulated the expression of cancer-related genes,including OSM,TNFRSF4,HRH3,and ***-seq(chromatin isolation by RNA purification and sequencing)revealed that these genes contained enriched chromatin binding sites for *** experiments confirmed that the effects of LLNLR-299G3.1 on ESCC cell proliferation were dependent on interaction with HRH3 and ***,intravenous delivery of placental chondroitin sulfate A binding peptide-coated nanoparticles containing antisense oligonucleotide(pICSA-BP-ANPs)strongly inhibited ESCC tumor growth and significantly improved animal survival in ***,our results suggest that LLNLR-299G3.1 promotes ESCC malignancy through regulating gene-chromatin interactions and targeting ESCC by pICSA-BP-ANPs may be an effective strategy for the treatment of lncRNA-associated ESCC.

Electrophysiological study on differentiation of rat bone marrow stromal stem cells into neuron-like cells in vitro by edaravone

Objective： To explore the electrophysiological proper- ties of differentiation of rat bone marrow-derived stromal stem cells （rBMSCs） to neuron-like cells in vitro by edaravone, a new type of free radical scavenger. Methods： Stromal stem cells were separated from rat bone marrow with Ficoll-Paque reagent and expanded in different culture medium in vitro, rBMSCs were induced by edaravone containing serum-free L-DMEM. Morphologic observation and Western blot analysis including the expression of Nav1.6, Kvl.2, Kv1.3, Cav1.2 were performed, and whole patch-clamp technique was used. Results： Cyton contraction and long processes were shown in differentiated stromal stem cells. Navl.6, Kvl.2, Kv 1.3 and Cav 1.2 were expressed in both differentiated and undifferentiated ceils. However, the expression of channel proteins in differentiated cells was up-regulated. Consistently, their resting potential and outward currents were also enhanced in the differentiated cells, which was especially significant in the outward rectifier potassium current. Conclusion： In vitro, neuron-like cells derived from rBMSCs, induced by edaravone, possess electrophysiological properties of neurons.

Acupuncture combined with acupoint injection for 25 cases of post-cold cough

Objective To observe the efficacy of acupuncture combined with acupoint injection for post-cold cough. Methods Twenty-five cases of post-cold cough, who visited the department of acupuncture-moxibustion of our hospital from January 1, 2016 to June 30, 2016, were reviewed, and all the patients were treated by acupuncture combined with acupoint injection. Acupoint selection for acupuncture： bilateral Dìngchuǎn（定喘 EX-B 1）, Fēngmén（风门 BL 12）, Lièquē（列缺 LU 7）, Fèishū（肺俞 BL 13） and Shènshū（肾俞 BL 23）; the acupuncture was conducted once a day, with 1 day of interval after 6 days;6 times was considered as 1 course, and 2 consecutive courses were needed. Acupoint injection with medicinal vitamin D2 and calcium levulinate injection was conducted after acupuncture. Acupoint selection for injection： bilateral EX-B 1 and BL 13, and the treatment was conducted once every other day. Results Among the 25 patients with post-cold cough, 8 patients were cured, 16 cases were effective, 1 case was ineffective, and the total effective rate was 96.0%. Conclusion The efficacy of acupuncture combined with acupoint injection in treatment of post-cold cough is good.

Microarray-based screening of common pathophysiological processes in dilated cardiomyopathy and ischemic cardiomyopathy

Background Heart failure is the end stage of various heart diseases,which has different underlying mechanisms but somehow has similar features among different etiologies. Dilated cardiomyopathy(DCM) and ischemic cardiomyopathy(ICM)are two major subtypes. Unlike widely explored mechanisms on these heart diseases,their shared gene expression alterations in heart failure have been rarely investigated. Methods We first obtained genetic profiles of DCM and ICM as well as their non-failing(NF)control from two datasets in GEO database,GSE3585 and GSE1869. Comparing to NF,the differentially expressed genes(DEGs)of DCM and ICM were screened out and used for Gene Ontology(GO)analysis by DAVID. The two sets of DEGs were compared and the overlap was selected out. Protein-protein interaction(PPI)network of the encoded proteins of shared DEGs was constructed and visualized via STRING and Cytoscape respectively. Gene annotation interactions were analyzed using Bi NGO. The closely linked genes were sorted out as hub genes and accounted for common processes of DCM and ICM. Results There were 99 DEGs in DCM vs. NF,while 1317 DEGs in ICM vs. NF.29 DEGs were shared in two comparisons,which were mainly enriched in regulation of protein catabolic process,Golgi to endosome transport,protein targeting to Golgi,inactivation of MAPK activity,Golgi to plasma membrane protein transport,cytosol,and protein binding. There were 13 genes linked in the PPI network and thus they were selected as hub genes. Functions of these hub genes were mainly focused on energetic metabolism disorder,apoptosis and cardiac fibrosis. Conclusion Energetic metabolism disorder,apoptosis and cardiac fibrosis are likely the shared pathophysiological processes for dilated cardiomyopathy and ischemic cardiomyopathy.[S Chin J Cardiol 2019;20(3):190-197]

BACKGROUND The frequency of acute hypertriglyceridemic pancreatitis(AHTGP) is increasing worldwide. AHTGP may be associated with a more severe clinical course and greater mortality than pancreatitis caused by other causes. Early identification of patients with severe inclination is essential for clinical decision-making and improving prognosis. Therefore, we first developed and validated a risk prediction score for the severity of AHTGP in Chinese *** To develop and validate a risk prediction score for the severity of AHTGP in Chinese *** We performed a retrospective study including 243 patients with AHTGP. Patients were randomly divided into a development cohort(n = 170) and a validation cohort(n = 73). Least absolute shrinkage and selection operator and logistic regression were used to screen 42 potential predictive variables to construct a risk score for the severity of AHTGP. We evaluated the performance of the nomogram and compared it with existing scoring systems. Last, we used the best cutoff value(88.16) for severe acute pancreatitis(SAP) to determine the risk stratification *** Age, the reduction in apolipoprotein A1 and the presence of pleural effusion were independent risk factors for SAP and were used to construct the nomogram(risk prediction score referred to as AAP). The concordance index of the nomogram in the development and validation groups was 0.930 and 0.928, respectively. Calibration plots demonstrate excellent agreement between the predicted and actual probabilities in SAP patients. The area under the curve of the nomogram(0.929) was better than those of the Bedside Index of Severity in AP(BISAP), Ranson, Acute Physiology and Chronic Health Evaluation(APACHE II), modified computed tomography severity index(MCTSI), and early achievable severity index scores(0.852, 0.825, 0.807, 0.831 and 0.807, respectively). The effects of Xuezhikang on vascular remodeling and aortic fibers in spontaneously hypertensive rats

Background Xuezhikang is a traditional Chinese medicine for the therapy of patients with cardiovascular diseases. However, the effect of Xuezhikang on vascular remodeling and aortic fibers in spontaneously hypertensive rats remains unclear. Methods Thirty spontaneously hypertensive rats（SHRs） aged 8 weeks were randomized into three groups： SHRs control group（SHRs group, n = 10）, group treated with low dose Xuezhikang（XZK-L,20 mg/kg/d, n = 10） and group treated with high dose Xuezhikang（XZK-H, 200 mg/kg/d, n = 10）. The normal group was comprised of ten Wistar-Kyoto（WKY） rats of the same age. While under pentobarbital anesthesia, in vivo aortic stiffness was determined by a pulse wave velocity（PWV） technique and measured locally in the descending thoracic aorta by Doppler ultrasound echocardiography. Fixed tissues were sectioned and stained with hematoxylin and eosin, masson trichrome, aldehyde-fuchsin and Gordon-Sweets stain. Results The PWV, thicknesses of the aortic wall, wall-to-lumen（W/L） ratio, elastic fibers thicknesses and collagen volume fraction were measured. XZK-L and XZK-H groups were observed to be significantly greater than those in the normal group（P 〈 0.01）. The levels of PWV, W/L ratio and collagen fibers were observed to be significantly reduced in the XZK-H group when compared with the control group（P 〈 0.05）. There was no significance in reticular fiber distributing between the XZK-treated group and SHR group. The reticular fibers in SHR rats were obviously more than those of WKY rats. Conclusion The hypertensive vascular remodeling is associated with changes of elastic fibers and collagen and reticular fibers. Xuezhikang improves hypertensive vascular remodeling by repairing the elastic fibers and inhibiting the levels of collagen.

This study aims to discuss and analyze the perioperative risk factors for postoperative hepatic insufficiency after type A dissection surgery.
