Outcomes of bictegravir/emtricitabine/tenofovir alafenamide versus efavirenz-based regimens in central China:a real-world assessment

To the Editor:Human immunodeficiency virus(HIV)epidemic poses a significant challenge to global public health.As of 2022,approximately 39.0 million people worldwide were living with HIV.[1]With the ongoing evolution of combined antiretroviral therapy(cART),HIV infection has transformed into a manageable chronic disease requiring long-term treatment.Among the various classes of antiretroviral(ARV)drugs,integrase strand transfer inhibitor(INSTI)-based regimens,known for potency and tolerability,have emerged as the predominant treatment modality worldwide.The nextgeneration of INSTI,bictegravir(BIC),received the Food and Drug Administration(FDA)approval in 2018 as a single-tablet regimen(STR)containing BIC(50 mg)/emtricitabine(FTC,200 mg)/tenofovir alafenamide(TAF,25 mg)for once daily.It has been endorsed as the preferred choice for treatment-naive patients by major guidelines,including the 2021 edition of the China Acquired Immune Deficiency Syndrome(AIDS)Diagnosis and Treatment Guidelines.[2]In China,people living with HIV(PLWH)are provided with flexible cART options,including the National Reimbursement Drug List(NRDL),self-pay drugs,and free drugs.Since being included in NRDL in 2021,BIC/FTC/TAF have gained popularity among an increasing number of patients.

Background:Immune checkpoint inhibitors(ICIs)are increasingly used as first-line therapy for patients with advanced non-small cell lung cancer(NSCLC)harboring no actionable mutations;however,data on their efficacy among patients presenting with intracranial lesions are limited.This study aimed to explore the efficacy and safety of ICIs combined with chemotherapy in advanced NSCLC patients with measurable brain metastasis at initial diagnosis.Methods:Our study retrospectively analyzed clinical data of a total of 211 patients diagnosed with driver gene mutation-negative advanced NSCLC with measurable,asymptomatic brain metastasis at baseline from Hunan Cancer hospital between January 1,2019 and September 30,2021.The patients were stratified into two groups according to the first-line treatment regimen received:ICI combined with chemotherapy(n=102)or chemotherapy(n=109).Systemic and intracranial objective response rates(ORRs)and progression-free survival(PFS)were analyzed.Adverse events were also compared between the groups.Results:Compared with the chemotherapy-based regimen,the ICI-containing regimen was associated with a significantly higher intracranial(44.1%[45/102]vs.28.4%[31/109],χ^(2)=5.620,P=0.013)and systemic(49.0%[50/102]vs.33.9%[37/109],χ^(2)=4.942,P=0.019)ORRs and longer intracranial(11.0 months vs.7.0 months,Pbased chemotherapy as a first-line regimen and prolonged intracranial PFS(hazard ratio[HR]=0.52,95%confidence interval[CI]:0.37-0.73,Pfirst-line treatment option for driver gene mutation-negative advanced NSCLC patients who present with brain metastasis at initial diagnosis.Clinical trial registration:https://www.clinica

At present,water pollution has become an important factor affecting and restricting national and regional economic development.Total phosphorus is one of the main sources of water pollution and eutrophication,so the prediction of total phosphorus in water quality has good research significance.This paper selects the total phosphorus and turbidity data for analysis by crawling the data of the water quality monitoring platform.By constructing the attribute object mapping relationship,the correlation between the two indicators was analyzed and used to predict the future data.firstly,the monthly mean and daily mean concentrations of total phosphorus and turbidity outliers were calculated after cleaning,and the correlation between them was analyzed.Secondly,the correlation coefficients of different times and frequencies were used to predict the values for the next five days,and the data trend was predicted by python visualization.Finally,the real value was compared with the predicted value data,and the results showed that the correlation between total phosphorus and turbidity was useful in predicting the water quality.

The oxidative DNA damage response： A review of research undertaken with Tsinghua and Xiangya students at the University of Pittsburgh

Endogenous stress and exogenous toxicants(chemicals and UV light) alter genetic information either directly or indirectly through the production of reactive oxygen species(ROS), thereby driving genomic instability in cells and promoting tumorigenesis. All living cells try to faithfully preserve and transmit their genomic information from one generation to the next using DNA repair mechanisms to repair oxidative DNA damage to prevent cancer or premature aging. Oxidative DNA damage comprises a mixture of DNA lesions including base damage, DNA single strand breaks(SSBs), and DNA double strand breaks(DSBs). This review summarizes some of the studies on DNA damage response at a defined genome locus that are performed by students from the Tsinghua University School of Medicine and the School of Medicine of Central South University(Xiangya hospital) at the University of Pittsburgh School of Medicine. A summary of their work highlights the continuous contribution of the students to a particular research program and exemplifies the achievements of this China-U.S. collaborative training program.
