Proteomic identification of proteins in exosomes of patients with atherosclerosis

AIM: Atherosclerosis primarily involved systemic arteries. Luminal surface,a monolayer of endothelial cells,of artery directly exposes to blood and is susceptible to active substances in the blood. Exosomes contain significantly amount of proteins and RNAs. Exosomes can be good and bad for cells,depending on their component. Thus,exosomes may contribute to atherosclerosis by affecting endothelial cells. This study analyzed the relationship of exosome proteins and atherosclerosis. METHODS: Fifty-six patients and healthy subjects were recruited and divided into two comparisons: healthy subjects vs atherosclerosis( HS vs AS),and hypertension vs hypertension plus atherosclerosis( HT vs HT + AS). Serum exosomes were decoded by protein mass spectrometry. The protein profile and function were analyzed by gene ontology( GO). RESULTS: It was found that five child terms repeatedly appeared in"response to stimulus"and "immune system process"of BP of the two categories( HS vs AS and AS vs HT + AS): "positive regulation of innate immune response","immune response-activating signal transduction","activation of innate immune response","innate immune response-activating signal transduction"and "innate immune response activating cell surface receptor signaling pathway". Two child terms repeatedly showed in "binding"of MF of the two categories: "antigen binding"and "enzyme binding". Two proteins,PSMA6 and PSMA7,were repeatedly shown in the two categories. CONCLUSION: GO analysis was utilized for structure hierarchy"tree"to illustrate these proteins involved in various terms in BP,CC and MF. The PPI analysis supplied proteins which may play potentially important roles in AS process. Innate immune system and blood coagulation pathway contribute to AS formation. The proteins,PSMA6,PSMA7 and Annexin A2,may can be the new target proteins for prevention and treatment of AS.

目的:观察针药结合治疗慢性盆腔炎的临床疗效。方法:将符合纳入标准的88例慢性盆腔炎患者随机分入治疗组或对照组,治疗组45例,采用针刺配合中药治疗;对照组43例,口服妇科千金胶囊治疗。结果:治疗3个月经周期后,治疗组痊愈28例,显效9例,有效4例,无效4例,总有效率91.1%;对照组痊愈7例,显效8例,有效15例,无效13例,总有效率69.8%,两组比较,差异有统计学意义(P<0.05),治疗组疗效优于对照组。半年后随访,治疗组复发率明显低于对照组(P<0.01)。结论:针药结合治疗慢性盆腔炎疗效好,不易复发。

Background No randomized trial has been performed to compare the efficacy of an intracoronary bolus of tirofiban versus urokinase during primary percutaneous coronary intervention （PCI）.We investigated whether the effects of adjunctive therapy with an intracoronary bolus of urokinase was noninferior to the effects of an intracoronary bolus of tirofiban in patients with ST-elevation myocardial infarction （STEMI） undergoing *** A total of 490 patients with acute STEMI undergoing primary PCI were randomized to an intracoronary bolus of tirofiban （10 μg/kg; n=247） or urokinase （250 kU/20 ml; n=243）.Serum levels of P-selectin,von Willebrand factor （vWF）,CD40 ligand （CD40L）,and serum amyloid A （SAA） in the coronary sinus were measured before and after intracoronary drug *** primary endpoint was the rate of complete （＞70％） ST-segment resolution （STR） at 90 minutes after intervention,and the noninferiority margin was set to 15％.Results In the intention-to-treat analysis,complete STR was achieved in 54.4％ of patients treated with an intracoronary bolus of urokinase and in 60.6％ of those treated with an intracoronary bolus of tirofiban （adjusted difference：-7.0％;95％ confidence interval：-15.7％ to 1.8％）.The corrected TIMI frame count of the infarct-related artery was lower,left ventricular ejection fraction was higher,and the 6-month major adverse cardiac event-free survival tended to be better in the intracoronary tirofiban *** intracoronary bolus of tirofiban resulted in lower levels of P-selectin,vWF,CD40L,and SAA in the coronary sinus compared with an intracoronary bolus of urokinase after primary PCI （P＜0.05）.Conclusions An intracoronary bolus of urokinase as an adjunct to primary PCI for acute STEMI is not equally effective to an intracoronary bolus of tirofiban with respect to improvement in myocardial reperfusion assessed by *** may be caused by less reduction in coronary circulatory platelet activation and inflammation.

AIM： To investigate matrix metalloproteinases（MMPs）and tissue inhibitor of metalloproteinases（TIMPs）expression during the progress of fusarium solani（***） keratitis in a rat model.· METHODS： A rat model of *** keratitis was produced using corneal scarification and a hand-made contact lens. MMPs and TIMPs expressiond were explored in this rat model of *** keratitis using real-time polymerase chain reaction（PCR） and ***6001（400 μmol/m L） was used to treat infected corneas. The keratitis duration, amount and area of corneal neovascularization（CNV） were evaluated.·RESULTS： MMP-3 expression was 66.3 times higher in infected corneas compared to normal corneas. MMP-8,-9,and-13 expressions were significantly upregulated in the mid-period of the infection, with infected- to-normal ratios of 4.03, 39.86, and 5.94, respectively. MMP-2 and-7expressions increased in the late period, with the infected-to-normal ratios of 5.94 and 16.22, ***-1 expression was upregulated in the early period,and it was 43.17 times higher in infected compared to normal corneas, but TIMP-2,-3, and-4 expressions were mildly downregulated or unchanged. The results of DIF were consistent with the result of real-time ***6001, a MMPs inhibitor, decreased the duration of *** infection and the amount and area of CNV.·CONCLUSION： MMPs and TIMPs contributed into the progress of *** keratitis.
