Initial steroid-free immunosuppression after liver transplantation in recipients with hepatitis c virus related cirrhosis

AIM:Steroids can increase hepatitis C virus(HCV) *** liver transplantation(LTx),steroids are commonly used for immunosuppression and acute rejection is usually treated by high steroid *** can worsen the outcome of recurrent HCV ***, we evaluated the outcome of HCV infected liver recipients receiving initial steroid-free immunosuppression. METHODS:Thirty patients undergoing LTx received initial steroid-free *** for LTx included 7 patients with HCV related *** immunosuppression consisted of tacrolimus 2×0.05mg/kg.d po and mycophenolate mofetil(MMF)2×15mg/kg.d *** tacrolimus dosage was adjusted to trough levels in the target range of 10-15μg/L during the first 3 mo and 5-10μg/L *** of acute rejection were verified histologically. RESULTS:Patient and graft survival of 30 patients receiving initial steroid-free immunosuppression was 86% and 83% at 1 and 2 *** rejection occurred in 8/30 patients, including 1 HCV infected *** HCV-infected patients had HCV genotype Ⅱ(lb).HCV seropositivity occurred within the first 4 mo after *** virus load was not remarkably increased during the first year after ***,grafts had no severe recurrent hepatitis. CONCLUSION:From our experience,initial steroid-free immunosuppression does not increase the risk of acute rejection in HCV infected liver ***,none of the HCV infected patients developed serious chronic liver *** suggests that it may be beneficial to avoid steroids in this particular group of patients after LTx.

Action Mechanisms of Arbuscular Mycorrhizal Fungi in Phosphorus Uptake by Capsicum annuum L.

A pot experiment was conducted to investigate the action mechanisms phorus （P） uptake of Capsicum annuum L. in a sterilized fossil Oxisol of arbuscular mycorrhizal （AM） fungi in phos- Three P levels of 0, 10 and 200 mg kg-1 soil （P0, P10 and P200, respectively） without and with AM fungal inoculation were applied as Ca（H2PO4）2-H20. Shoot dry matter yields and shoot P uptake increased significantly （P 〉 0.05） by the inoculation of AM fungi at P0 and P10. Root length and P concentration in soil solution increased with the inoculation of AM fungi but the root：shoot ratio decreased or remained constant. Around 50% roots of inoculated plants were infected by AM and the external hyphae amounted to 20 m g^-1 soil at P10 and P200. The hyphae surface area of the infected root cylinder amounted to 11 and 2 cm^2 cm^-2 root at P0 and P10, respectively. The increased P uptake of inoculated plants was mainly because of an up to 5 times higher P influx of the infected root. Model calculations showed that the root alone could not have achieved the measured P influx in both infected and non-infected roots. But the P influx for hyphae calculated by the model was even much higher than the measured one. The P uptake capacity of hyphae introduced in the model was too high. Model calculations further showed that the depletion zone around roots or hyphae was very narrow. In the case of the root only 7% of the soil volume would contribute P to the plant, while in the case of hyphae it would be 100%. The results together with the model calculations showed that the increased P uptake of AM inoculated plants could be explained partly by the increased P concentration in the soil solution and by the increased P absorbing surface area coming from the external hyphae.

Treatment of genotype 2 and 3 chronic hepatitis C virus-infected patients

AIM： Before pegylated interferon alpha （IFN） was introduced for the therapy of chronic hepatitis C virus （HCV）-induced hepatitis, conventional thrice weekly IFN therapy was supplemented by ribavirin. Also, at that time, higher and more frequent doses of IFN were expected to be more effective than the standard regimen of 3 MU thrice weekly. As ribavirin significantly increases side effects and negatively influences the quality of life particularly in young patients, we started a prospective non-randomized study with a daily IFN-2a monotherapy as an initial treatment for chronic hepatitis C. METHODS： Forty-six consecutive chronic HCV-infected patients received 3 MU IFN-2a per day as an initial treatment. Patients with genotype 2 or 3 （n = 12） were treated for 24 wk, and patients with genotypes other than 2 or 3 （n = 34） for 48 wk. Treatment outcome was followed up for 48 wk after the end of treatment （EOT）. Virological response was defined as the absence of detectable serum HCV-RNA. Patients without virological response at 12 wk after the start of treatment received low-dose ribavirin （10 mg（kg·d）） additionally. RESULTS： During treatment, three genotype 3 patients were excluded from the study due to incompliance. The remaining patients （n = 9） infected with genotype 2 or 3 showed an initial virological response rate of 100%. Six patients （66.7%） were still found to be virus-free at the end of follow-up period. In these patients, initial virological response was evident already after 2 wk of treatment. In contrast, initial virological response occurred first after 4 wk of treatment in the three patients who relapsed （33.3%）. In comparison, patients infected with genotypes other than 2 or 3 （n = 34） showed an initial virological response rate of only 23.5% （n = 8）, and even in combination with ribavirin a sustained virological response （SVR） rate of only 11.8% （n = 4） could be achieved. Phosphoinositide and Inositolpolyphosphate Signalling in Defense Responses of Arabidopsis thaliana Challenged by Mechanical Wounding

Various biochemical signals are implicated in Arabidopsis wound signalling, including jasmonic acid （JA）, salicylic acid, auxin, and Ca^2＋. Here, we report on cross-talk of phytohormones with phosphoinositide signals not previously implicated in plant wound responses. Within 30 min of mechanical wounding of Arabidopsis rosette-leaves, the levels of the lipid-derived soluble inositolpolyphosphate, inositol 1,4,5-trisphosphate （InsP3）, increased four to five-fold. Concom- itantly, the precursor lipids, phosphatidylinositol 4,5-bisphosphate, phosphatidylinositol 4-phosphate and phosphatidylinositol transiently depleted, followed by re-synthesis after 30-60 min of stimulation. Increased InsP3 levels with wounding coincided with JA increases over the first hours of stimulation. In dde2-2-mutant plants deficient in JA biosynthesis, no InsP3 increase was observed upon wounding, indicating that JA was required for InsP3 formation, and InsP3 levels increased in wild-type plants challenged with sorbitol, increasing endogenous JA levels. In InsP 5-ptase plants with attenuated phosphoinositide signalling, the induction of wounding-inducible genes was diminished compared with wildtype plants, suggesting a role for phosphoinositide signalling in mediating plant wound responses. The gene-expression patterns suggest that phosphoinositides contribute to both JA-dependent and JA-independent aspects of wound signalling. Weight gain of Plutella xylostella caterpillars feeding on InsP 5-ptase plants was increased compared with that of caterpillars feeding on wild-type plants. The ecophysiological relevance of phosphoinositide signals in plant defense responses to herbivory is discussed in light of recent findings of inositolpolyphosphate involvement in phytohormone-receptor function.

Background Public interest in the way food is produced on the farm and processed along the food-production chain is *** analysis of isotopic signatures(^(13)C)in cow tail hair provides a method to reconstruct the dietary proportion of maize in cow *** on this,we further investigated whether there is a relationship between isotopic signatures in cow tail hair or farm slurry and the proportion of maize of the total utilized agricultural area per farm[%].We did an on-farm survey on 17 dairy farms in coastal Northwest germany and collected cow tail hair from dairy cows and slurry samples on each *** farms differed in their feeding regime(C_(3) vs.C_(4) plants),their site conditions(sandy soil=‘geestland’;organic soil=‘Peatland’;clayey soil=‘Marshland’),and in the area cultivated with maize as a proportion of the total utilized agricultural area per *** We found a positive relationship between δ^(13)C values in both cow tail hair and slurry and the annual dietary proportion of maize(R^(2)=0.67;and R^(2)=0.63).Furthermore,we confirmed that there was a relationship betweenδ^(13)C values in cow tail hair and area of maize as a proportion of the total utilized agricultural area per farm(R^(2)=0.69).Conclusion Our findings suggest a general applicability of using isotopic signatures(^(13)C)along a wide gradient of site conditions and productions systems in practice.
