Phospholipase D antagonist 1-butanol inhibited the mobilization of triacylglycerol during seed germination in Arabidopsis

Storage oil breakdown plays an important role in the life cycle of many plants by providing the carbon skeletons that support seedling growth immediately following germination. 1-Butanol, a specific inhibitor of phospholipase D(PLD)-dependent production of the signalling molecule phosphatidic acid(PA), inhibited Arabidopsis seed germination. N-Acylethanolamines(NAEs), which have been shown to inhibits PLDa1 activity, have no effect on seed germination. However, mobilization profile of triacylglycerols(TAG) that induced by each compound has not been reported. To gain deeper insights into the mode of mobilization of TAG during NAE 12:0 or 1-butanol treatment, we conducted a detailed comparative analysis of the effect of NAE 12:0, DMSO, 1-butanol and tert-butanol on Arabidopsis seed germination and fatty acid composition, tert-butanol and DMSO served as the corresponding controls treatment respectively. Our data show that 1-butanol, but not the inactive tert-butanol isomer, inhibited Arabidopsis seed germination, which is accompanied by a with retardation of the mobilization of triacylglycerols(TAG). In contrast, NAE 12:0 did not affect mobilization of TAG, nor did it significantly delay seed germination as monitored by radicle and cotyledon emergence. 1-Butanol induced RNA degradation in seeds and seedlings. We speculate that the large-scale degradation of RNA under the induction of 1-butanol may lead to abnormal gene expression in genes necessary for seed germination, including the genes needed for the mobilization of oil bodies, and thus cause a delay of seed germination. To the best of our knowledge, we report for the first time that 1-butanol delays the mobilization of TAG.

In the intricate process of natural product biosynthesis, a metabolon can enhance metabolic flux by associating sequential enzymes. A fungal metabolon, comprising of flavin-dependent monooxygenase Spe F and P450 monooxygenase Spe G, is identified in the biosynthesis of spiro polycyclic alkaloids(+)-notoamide B and its diastereomer(+)-versicolamide B. Using notoamide E as a substance, Spe F/Spe G metabolon can control the stereoselectivity of its 2,3-epoxidation, followed by hydrogen atom ion at C-17 to generate reactive epoxide tau-m A with dienyl iminium unit. Subsequently,(+)-notoamide B and(+)-versicolamide B are produced via tandem nonenzymatic inverse-electron-demand Diels-Alder reaction and semipinacol rearrangement. This provides the first example of metabolon in the biosynthesis of spiro-prenylated indole alkaloids.

Preliminary study on Cd accumulation characteristics in Sansevieria trifasciata Prain

Phytoremediation techniques to clean heavy metal pollution soil depend on identifying plant species that can act as *** important approach to screening potential phytoremediators is to evaluate characteristics of heavy metal *** this study,we performed firsthand analysis of Cd tolerance and accumulation characteristics of three Sansevieria trifasciata cultivars by pot *** growth results showed that all three *** cultivars can tolerate 50 mg kg^-1 soil Cd *** growth under 50 mg kg^-1 soil Cd concentration for 4 months,the Cd bioconcentration factors in the shoots of S.‘Trifasciata’,***‘Laurentii’,and ***‘Silver Hahnii’were 1.26,1.30,and 1.19,while those in the roots were 12.53,11.43,and 5.45,*** result reveals the considerably low translocation factors of 0.10,0.12,and 0.22 for S.‘Trifasciata’,***‘Laurentii’,and ***‘Silver Hahnii’,*** results suggest that all three *** cultivars had high Cd absorption capacities but low Cd translocation *** combination with total Cd accumulation distribution and plant growth characteristics,*** can be designed as a phytostabilizer in Cd-contaminated soils in its cultivation ***,the mechanism of high Cd tolerance and accumulation characteristics in the roots of *** should be *** study provides new resources for dealing with Cd-contaminated soils and exploring Cd tolerance and accumulation mechanisms in plants.

A new subtribal classification of the woody bamboo tribe Arundinarieae is proposed based on recent phylogenomic *** subtribes,corresponding to the five major lineages of the ddRAD-seq based phylogenomic trees,are recognised:Arundinariinae(the leptomorph lineage),Ampelocalaminae(the ADH lineage),Gaoligongshaniinae(represented by Gaoligongshania),Hsuehochloinae(represented by Hsuehochloa) and Thamnocalaminae(the pachymorph lineage,i.e.,alpine bamboos).Subtribes Ampelocalaminae,Gaoligongshaniinae and Hsuehochloinae are newly established,while the circumscriptions of subtribes Arundinariinae and Thamnocalaminae differ from the traditional *** Arundinariinae also includes those taxa that were previous members of the subtribe ***,among the five redefined subtribes,Arundinariinae is the most heterogenous in terms of *** Arundinarieae,rhizome type has greater implications for classification than other vegetative and reproductive characters at the subtribal *** addition,the new monotypic genus Ravenochloa is described on the basis of its mo rphological characteristics and geographical distribution to accommodate the unique phylogenetic entity of Indocalamus wilsonii.

A comparison of different methods for preserving plant molecular materials and the effect of degraded DNA on ddRAD sequencing

Obtaining high-quality plant materials for experiments is challenging for many research ***, it is of special importance to determine the best method for preserving biological macromolecules like DNA, which degrade over time. Although some research has demonstrated that DNA degradation has little effect on traditional molecular markers, the effects of DNA degradation on dd RADseq, a popular reduced-representation sequencing technology, have not been adequately investigated. In this study, we first chose six woody bamboo species(Bambusoideae, Poaceae) to explore appropriate methods for preserving molecular materials with two DNA extraction approaches. Then we sequenced twenty-one bamboos and examined the effects of DNA quality on data generation using the dd RAD-seq technique(Midd RAD-seq). Finally, we reconstructed phylogenies of twenty woody bamboo species. We found that the integrity of dry-powdered DNA was preserved longer than that of TE-dissolved DNA,regardless of whether the DNA was extracted by a modified CTAB protocol or DNAsecure plant kit. The dd RAD-seq data were robust, except when DNA was severely degraded. In addition, we resolved the phylogenetic positions of the sampled Phyllostachys spp. Our results suggest that dry-powdered DNA is the most appropriate preservation method for plant molecular materials. Furthermore, a moderate level of DNA degradation has little effect on reduced representation sequencing techniques represented by dd RAD-seq.

Flavonoid scutellarin positively regulates root length through NUTCRACKER

Exploring approaches to regulate meristem is of special importance and broad *** this study,we found that the flavonoid scutellarin,which has a 6-hydroxyl and a 7-glucoside,increased root length through the transcription factor NUTCRACKER(NUC).This root lengthening disappeared in NUCknockout and reappeared in NUC-rescue *** induced NUC expression and promoted the division of cortex/endodermal *** contrast,naringenin,which has same chemical backbone but without 6-hydroxyl and with 7-hydroxyl group,showed the opposite or no *** results demonstrate that scutellarin promotes root length through NUC-mediated regulatory pathways and reveal that flavonoids with and without the 6-hydroxyl and 7-glucoside have positive and negative effects on meristem size,respectively。

Deep genome skimming reveals the hybrid origin of Pseudosasa gracilis (Poaceae: Bambusoideae)

Pseudosasa gracilis(Poaceae:Bambusoideae)is a temperate woody bamboo species endemic to Southcentral China with a narrow *** phylogenetic studies revealed an unexpected,isolated phylogenetic position of *** we conducted phylogenomic analysis by sampling populations of *** and its sympatric species *** and Sinosasa polytricha reflecting different genomic signals,by deep genome *** molecular evidence from chloroplast genes and genome-wide SNPs,we deciphered the phylogenetic relationships of *** plastid and nuclear data indicate that *** is more closely related to Sinosasa,which is discordant with the taxonomic *** further explore this molecular-morphological conflict,we screened 411“perfect-copy”syntenic genes to reconstruct phylogenies using both the concatenation and coalescent *** observed extensive discordance between gene trees and the putative species tree.A significant hybridization event was detected based on 411 genes from the D subgenome,showing *** was a hybrid descendant between Sinosasa longiligulata and ***,with 63.56%and 36.44%inheritance probabilities of each ***,introgression events were detected in the C subgenome between *** and *** in the same distribution *** findings suggest that sympatric hybridization and introgression play a crucial role in the origin of *** providing an empirical example of bamboo of hybrid origin using comprehensive analyses based on genomic data from different inheritance systems and morphological characters,our study represents a step forward in understanding of reticulate evolution of bamboos.

Floristics and Plant Biogeography in China

In 1998, a revolutionary system of angiosperm classification, the Angiosperm Phylogeny Group system was published. Meanwhile, another new system of classification of angiosperms, the eight-class system was proposed by C.Y. Wu and colleagues based on long term work on the flora of China. The Flora Reipublicae Popularis Sinicae project was initiated in 1959 and completed by 2004. It is the largest Flora so far completed in the world, including 31 228 species of vascular plants, or one-eighth of the global plant diversity. The English-language and updated Flora of China （FOC） is an international joint effort initiated in 1988 and accelerated in 1998. Up to now, 15 of the 24 volumes of the FOC have been published. Based on the floristic data, the composition, characteristics, floristic divisions and affinities of the flora of China have been studied by Wu and colleagues since 1965. In the past 10 years, analyses of the available floristic data have been very productive. The East Asiatic Floristic Kingdom was proposed in 1998. All 346 families of angiosperms in China, according to the eight-class system of classification, were comprehensively discussed by using knowledge of current and historical distribution of seed plants in the world, together with some morphological and molecular data. A scheme of distribution patterns or areal-types of families and genera of seed plants in China was modified and elucidated, together with a proposed scheme of areal-types of the world. Molecular phylogenetic and biogeographical studies of angiosperms in China in the past 10 years also witnessed a progressive development. Integration of morphological and molecular data and fossil evidence revealed some significant results. Eastern Asia, which used to be regarded as an important center of survival during the ice age, is likely an important center of diversification of angiosperms.

Nitrogen Use Efficiency of Rice Under Cadmium Contamination:Influence of Rice Cultivar Versus Soil Type

There is a need for rice cultivars with high yields and nitrogen(N) use efficiency(NUE), but with low cadmium(Cd) accumulation in Cd-contaminated paddy *** determine the relative effects of rice genotype, soil type, and Cd addition on rice grain yield and NUE, a pot experiment consisting of nine rice cultivars was conducted in two types of paddy soils, red soil(RS) and yellow soil(YS),without or with Cd spiked at 0.6 mg kg^(-1).The N supply was from both soil organic N pools and N fertilizers; thus, NUE was defined as the grain yield per unit of total crop-available N in the *** addition decreased grain yield and NUE in most rice cultivars,which was mainly related to reduced N uptake efficiency(NpUE, defined as the percentage of N taken up by the crop per unit of soil available N).However, Cd addition enhanced N assimilation efficiency(NtUE, defined as the grain yield per unit of N taken up by the crop) by 21.9% on average in all rice *** NpUE was mainly affected by soil type, whereas NtUE was affected by rice *** cultivars had higher NUEs than the japonica and indica cultivars because of their greater biomass and higher tolerance to Cd *** of NUE after Cd addition was stronger in RS than in YS, which was related to the lower absorption capacity for Cd in *** correspondence analysis-based variation partitioning showed that cultivar type had the largest effect(34.4%) on NUE, followed by Cd addition(15.2%) and soil type(10.0%).

Plastome characteristics of Cannabaceae

Cannabaceae is an economically important family that includes ten genera and ca. 117 accepted *** explore the structure and size variation of their plastomes, we sequenced ten plastomes representing all ten genera of Cannabaceae. Each plastome possessed the typical angiosperm quadripartite structure and contained a total of 128 genes. The Inverted Repeat(IR) regions in five plastomes had experienced small expansions(330 e983 bp) into the Large Single-Copy(LSC) region. The plastome of Chaetachme aristata has experienced a 942-bp IR contraction and lost rpl22 and rps19 in its IRs. The substitution rates of rps19 and rpl22 decreased after they shifted from the LSC to IR. A 270-bp inversion was detected in the Parasponia rugosa plastome, which might have been mediated by 18-bp inverted repeats. Repeat sequences, simple sequence repeats, and nucleotide substitution rates varied among these *** markers with more than 13% variable sites and 5% parsimony-informative sites were identified, which may be useful for further phylogenetic analysis and species identification. Our results show strong support for a sister relationship between Gironniera and Lozanell(BS ? 100). Celtis, CannabisHumulus, Chaetachme-Pteroceltis, and Trema-Parasponia formed a strongly supported clade, and their relationships were well resolved with strong support(BS ? 100). The availability of these ten plastomes provides valuable genetic information for accurately identifying species, clarifying taxonomy and reconstructing the intergeneric phylogeny of Cannabaceae.
