Breeding Potential of Some Exotic Tomato Lines:A Combined Study of Morphological Variability,Genetic Divergence,and Association of Traits

Tomato(Solanum lycopersicum L.)is called‘the poor man’s orange’due to its low price and improved nutritional *** experiment was conducted to study the breeding potential of some exotic tomato lines by assessing various qualitative and quantitative traits conferring yield and quality *** the qualitative traits,greater variability was observed for growth type,stem hairiness,and fruit shape and size.A determinate growth habit was observed in the genotype AVTO9802 while the genotype AVTO0102 produced yellow color fruits.A significant(p≤0.01)variation was also observed for the studied quantitative *** on yield and traits attributed to yield,the genotypes AVTO0314,GPB0107,GPB0120 and AVTO9802 were selected as promising *** differences between the genotypic and phenotypic coefficients of variation(GCV and PCV)of the studied quantitative traits were very *** suggests that the apparent variation was mainly due to the *** higher GCVand PCV values were observed for the number of primary branches plant^(−1)(NPB),number of fruits cluster^(−1)(NFC),individual fruit weight(IFW)and total soluble solids(TSS).High heritability was recorded for all quantitative traits in a broad ***,the individual fruit diameter showed the highest heritability(99.56).The highest(102.75)genetic advance(GA)was observed for the number of fruits plant^(−1)(NFP).High heritability coupled with high GA as percentage of mean were recorded for the traits NFP,NFC,fruit yield plant^(−1)(FYP)and *** showed a significant positive correlation with NFC(0.714^(***))and a negative correlation with days to the first harvest(−0.539^(***))and plant height(−0.492**).Principal component analysis revealed that the first four components explained 78.5%of the total variation among the ***,the promising genotypes(AVTO0314,GPB0107,GPB0120,AVTO9802 and AVTO0102)isolated from this study can be used for developing high-yielding and high-quality tomato va

Identification and fine mapping of quantitative trait loci for the number of vascular bundle in maize stem

Studies that investigated the genetic basis of source and sink related traits have been widely ***, the vascular system that links source and sink received much less attention. When maize was domesticated from its wild ancestor, teosinte, the external morphology has changed dramatically; however, less is known for the internal anatomy changes. In this study, using a large maize-teosinte experimental population, we performed a high-resolution quantitative trait locus（QTL） mapping for the number of vascular bundle in the uppermost internode of maize *** results showed that vascular bundle number is dominated by a large number of small-effect QTLs, in which a total of 16 QTLs that jointly accounts for 52.2% of phenotypic variation were detected, with no single QTL explaining more than 6% of variation. Different from QTLs for typical domestication traits, QTLs for vascular bundle number might not be under directional selection following *** Near Isogenic Lines（NILs） developed from heterogeneous inbred family（HIF）, we further validated the effect of one QTL qVb9-2 on chromosome 9 and fine mapped the QTL to a 1.8-Mb physical region. This study provides important insights for the genetic architecture of vascular bundle number in maize stem and sets basis for cloning of qVb9-2.

Laboratory-and Field-Phenotyping for Drought Stress Tolerance and Diversity Study in Lentil (Lens culinaris Medik.)

Drought susceptibility and low genetic variability are the major constraints of lentil(Lens culinaris Medik.)production *** of an efficient pre-field drought phenotyping technique and identification of diversified drought tolerant lentil genotype(s)are therefore vital and *** separate experiments were conducted using thirty diverse lentil genotypes to isolate drought tolerant genotype(s)as well as to assess their *** both of the experiments,significant(p≤0.01)variation in genotype(G),treatment(T)and G X T was observed for most of the studied *** experiment I,genotypes were examined for drought tolerance at the seedlings stage under hydroponic conditions by assessing root and shoot *** the 30 genotypes studied,BM-1247,BM-1227 and BM-502 were selected as highly tolerant to drought stress as they showed maximum seedling survivability and minimum reduction in growth parameters under drought *** experiment II,the genotypes were assayed for diversity and drought stress tolerance based on morphological traits grown under field *** stress caused a substantial reduction in yield attributing traits,however,the genotypes BM-1247,BM-981,BM-1227 and BM-502 were categorized as drought tolerant genotypes with less than 20%yield *** field screening result of drought stress tolerance was coincided well with the results of laboratory *** divergence study reflected the presence of considerable diversity among the *** laboratory and field screening results,the genotypes,BM-1247,BM-1227,BM-981 and BM-502 were selected as the best drought tolerant *** information can be exploited for further breeding in developing drought tolerance in lentil.

The article studies the activity of the peroxidase enzyme in cereals and barks of some varieties of soy plants. The soy (plant) samples we studied were obtained in the conditions of the Tashkent region at the experimental field of the Institute of genetics and plant experimental biology in the village of Durmen. 11 samples out of 40 examined on electrophoregrams did not show the presence of the studied enzymes. Further research is needed to clarify the results.

Objective Mutations in CUL4 B gene are one of the most common causes of X-linked mental retardation. Although the E3 ubiquitin ligase complex formed by CUL4 B(CRL4 B) is known to target substrates for proteolysis or to coordinate with PRC2 complex in promoting H3 K27 trimethylation and transcriptional repression, how CUL4 B dysfunction causes mental retardation remains unclear. To gain insights into the mechanisms by which CUL4 B regulates neural development, we employed three lines of conditional knockout mice and observed the learning and memory abilities of these mice. Method We crossed Cul4 b floxed mice with Nestin-Cre transgenic mice to generate nervous system specific Cul4 b conditional knock-out mice(referred to as Cul4 bNestin-Cre hereinafter). We further crossed Cul4 b floxed mice to Nse-Cre or Gfap-Cre transgenic mice to generate neuronspecific Cul4 b knockout mice(Cul4 bNse-Cre) or astrocyte-specific Cul4 b knockout mice(Cul4 bGfapCre), respectively. Result Here we report that mice lacking expression of Cul4 b in nervous system(Cul4 bNestin-Cre) are impaired in spatial learning and memory. To further determine the cell type responsible for the impaired spatial learning and memory in Cul4 bNestin-Cre mice, we characterized two lines of mice that lack expression of Cul4 b in neurons(Cul4 bNse-Cre) and in astrocytes(Cul4 bGfap-Cre), respectively. We observed that learning and memory were impaired in Cul4 bNse-Cre mice, but not in the Cul4 bGfap-Cre line, suggesting that the impairment in spatial learning and memory in Cul4 bNestin-Cre mice was due to the lack of Cul4 b in neurons but not in astrocytes. Conclusion Together, our results demonstrate a critical role of CUL4 B in neuronal function and provide a potential model for human X-linked mental retardation.

Enhancement of naked FIX minigene expression by chloroquine in mice

AIM: To study the effect of chloroquine on the expression of human clotting factor IX (hFIX) in mice. METHODS:Hydrodynamics-based naked DNA plasmid administration was performed by tail vein injection of 10 g of pCMV-hFIX and chloroquine (0, 100, 200, and 500 mol/L) in 2.2 mL of Ringer solution within 6-7 s, the level andstability of hFIX expression, liver damage and toxicity were then examined. RESULTS: The maximum expression ofhFIX level was 4.41.8 mg/L at 8 h after injection, 9.71.6 mg/L at 24 h only existed in 200 mol/L chloroquine-treated animals, which is 3-4 fold higher than that of control (P0.05). CONCLUSION: Chloroquine can enhance and sustain exogenous geneexpression in vivo without side effect under our experimental conditions.

Changes in the Photosynthetic Pigment Contents and Transcription Levels of Phycoerythrin-Related Genes in Three Gracilariopsis lemaneiformis Strains Under Different Light Intensities

Three Gracilariopsis lemaneiformis strains,including wild type and high-temperature-resistant cultivars 981 and 2007,were studied with the changes in their photosynthetic pigment contents and related gene transcription levels under different light intensities(10,60,100,and 200μmolm^(−2)s^(−1)).The three *** strains had the following photosynthetic pigments with high-to-low contents:phycoerythrin(PE),phycocyanin(PC),allophycocyanin(APC),and chlorophyll a(Chl a).Among the three strains,cultivar 981 had the highest PE content,followed by cultivar *** PC and APC contents were similar among the three strains,but they were higher in cultivars 981 and 2007 than in the wild *** Chl a contents in the three *** strains were equal.A low light intensity(10μmolm^(−2)s^(−1))promoted photosynthetic pigment accumulation in *** and improved the relative PE gene transcription(peA and peB)in a short period(≤6 d).A high light intensity decreased the PE *** and PebB,which catalyzed phycoerythrobilin synthesis,showed no compensatory upregulation at a low light intensity among the strains except for the wild *** a high light intensity,transcription levels of pebA and pebB in the three strains were *** study provided an experimental basis for elucidating the photosynthesis of *** key genes of algal growth,photo-synthesis-related genes served as useful gene markers for screening elite varieties with good traits in *** 2007 was superior to cultivar 981 in terms of maintaining high pigment levels in a wide range of light intensities,which is the most suitable for aquaculture.

Wide hybridization:engineering the next leap in wheat yield

Wide hybridization and chromosome engineering constitute an active and important area of plant biology research. The insights obtained in this field have contributed significantly to the understanding of the structure and function of plant chromosomes and genomes, and to the genetic improvement of many agricultural crops. Owing to well defined genomic relationships, clearly visible metaphase chromosomes, and relatively high interspecific and intergeneric crossability, wheat and related Triticeae species form ideal experimental systems for conducting wide hybridization and chromosome engineering studies （Sears, 1972, 1981; Singh and Jauhar, 2006）. Pioneering investigations have demonstrated that the entire genome or chromatin from Triticeae relatives can be transferred into wheat genome through chromosome engineering （re- viewed by Qi et al., 2007）.

Genetic Diversity of Italian Ryegrass Diploid Cultivars, Revealed by Electrophoretically Detected Genotypes in Phosphoglucose Isomerase （PGI）

Nine diploid cultivars of Italian ryegrass （Lolium multiflorum Lain.） from France （Fortyl, Vertyl and Jericho）, Germany （Ligrande）, United Kingdom （Aber Epic and Aber Mario）, Denmark （Cordelia）, Netherlands （Alamo） and Poland （Tur） were tested with horizontal gel electrophoresis according to one locus （with four alleles） of the PGI enzyme system. One of them, named P4 is typical for the species, therefore can serve as a good marker for hybrids identification. Each cultivar was characterized by frequencies of different phenotypes. They were highly polymorphic （Pg = 0.58 - 0.78） and showed differences in heterozygosity level. The variability within populations （GST = 0.055） was higher than among populations （DST = 0.032）.
