Objective: To observe the protective effect and mechanism of hydroxyl safflower yellow A(HSYA)from myocardial ischemia-reperfusion injury on human umbilical vein endothelial cells(HUVECs). Methods:HUVECs were treated with oxygen-glucose deprivation reperfusion(OGD/R) to simulate the ischemia reperfusion model, and cell counting kit-8 was used to detect the protective effect of different concentrations(1.25–160 μmol/L)of HSYA on HUVECs after OGD/R. HSYA 80 μmol/L was used for follow-up experiments. The contents of inflammatory cytokines interleukin(IL)-18, IL-1β, monocyte chemotactic protein 1(MCP-1), tumor necrosis factor α(TNF-α) and IL-6 before and after administration were measured by enzyme-linked immunosorbent assay. The protein expressions of toll-like receptor, NOD-like receptor containing pyrin domain 3(NLRP3), gasdermin D(GSDMD) and GSDMD-N-terminal domain(GSDMD-N) before and after administration were detected by Western blot. NLRP3 inflammasome inhibitor cytokine release inhibitory drug 3 sodium salt(CRID3 sodium salt, also known as MCC950) and agonist were added, and the changes of NLRP3, cysteine-aspartic acid protease 1(Caspase-1), GSDMD and GSDMD-N protein expressions were detected by Western blot. Results: HSYA inhibited OGD/R-induced inflammation and significantly decreased the contents of inflammatory cytokines IL-18,IL-1β, MCP-1, TNF-α and IL-6(P<0.01 or P<0.05). At the same time, by inhibiting NLRP3/Caspase-1/GSDMD pathway, HSYA can reduce the occurrence of pyroptosis after OGD/R and reduce the expression of NLRP3,Caspase-1, GSDMD and GSDMD-N proteins(P<0.01). Conclusions: The protective effect of HSYA on HUVECs after OGD/R is related to down-regulating the expression of NLRP3 inflammasome and inhibiting pyroptosis.

Plasma nitrogen fixation(PNF)has been emerging as a promising technology for greenhouse gasfree and renewable energy-based ***,most PNF studies seldom address practical application-specific *** this work,we present the development of a compact and automatic PNF system for on-site agricultural *** system utilized a gliding-arc discharge as the plasma source and employed a dual-loop design to generate NO_(x)from air and water under atmospheric *** results showed that the system with a dualloop design performs well in terms of energy costs and production *** operational parameters for the system were determined through experimentation,resulting in an energy cost of 13.9 MJ mol^(-1)and an energy efficiency of 16 g kWh^(-1)for NO_(3)^(-)production,***,the concentration of exhausted NO_(x)was below the emission *** lettuce cultivation experiments demonstrated that NO_(x)^(-)produced by the PNF system could serve as liquid nitrate nitrogen ***,our work demonstrates the potential of the developed PNF system for on-site application in the production of green-leaf vegetables.

Photocatalytic degradation of volatile organic compounds(VOCs)is a significant applying aspect of *** the modulation of photocatalysts and the rational dispersion of them on supports are key for solar-driven VOC *** batch-type photoreactors have low efficiency while continuous-flow photoreactors suffer from the problem of incomplete removal of ***,aiming for continuous and complete degradation of toluene gas as the target contaminant,continuous-flow pho-tocatalytic degradation reactors were made by adhering the vanadium and nitrogen codoped TiO_(2)on honeycomb ceramics(V/N-TiO_(2)@HC)by a simple sol-gel *** such a reactor,the rich ordered pores in the HC accelerate mass transport of toluene,and the introduction of V/N dopants narrows the bandgap and widens the light absorption range of TiO_(2),together resulting in continuous and nearly-complete pho-tocatalytic degradation of *** unique and stable structure of HC allows the photocatalysts to be *** degradation rate of toluene gas can reach 97.8%,and after 24 rounds of photocatalytic degra-dation,there is still a degradation rate of 96.7%.The impacts of loading times and gaseous flow rate on the photocatalytic performance of V/N-TiO_(2)@HC are studied in *** study provides a practical so-lution for the continuous and complete photocatalytic degradation of VOCs and opens a new application field for HC.

Small molecule drugs play a pivotal role in the arsenal of anticancer pharmacological agents. Nonetheless, their small size poses a challenge when directly visualizing their localization, distribution, mechanism of action (MOA), and target engagement at the subcellular level in real time. We propose a strategy for developing triple-functioning drug beacons that seamlessly integrate therapeutically relevant bioactivity, precise subcellular localization, and direct visualization capabilities within a single molecular entity. As a proof of concept, we have meticulously designed and constructed a boronic acid fluorescence drug beacon using coumarin–hemicyanine (CHB). Our CHB design includes three pivotal features: a boronic acid moiety that binds both adenosine triphosphate (ATP) and adenosine diphosphate (ADP), thus depleting their levels and disrupting the energy supply within mitochondria;a positively charged component that targets the drug beacon to mitochondria;and a sizeable conjugated luminophore that emits fluorescence, facilitating the application of structured illumination microscopy (SIM). Our study indicates the exceptional responsiveness of our proof-of-concept drug beacon to ADP and ATP, its efficacy in inhibiting tumor growth, and its ability to facilitate the tracking of ADP and ATP distribution around the mitochondrial cristae. Furthermore, our investigation reveals that the micro-dynamics of CHB induce mitochondrial dysfunction by causing damage to the mitochondrial cristae and mitochondrial DNA. Altogether, our findings highlight the potential of SIM in conjunction with visual drug design as a potent tool for monitoring the in situ MOA of small molecule anticancer compounds. This approach represents a crucial advancement in addressing a current challenge within the field of small molecule drug discovery and validation.

In the hydrological year 2022/2023,the glaciers in the Qilian Mountains experienced unprecedented mass *** glacier-wide mass balance was-1,188 mm w.e.,in contrast to-350 mm of average mass balance since 1990 over the Bailanghe Glacier No.12 in the middle of Qilian *** temperature during 2022–2023 reached the highest value ever recorded,second only to 2022,while at the same time the precipitation amount was less compared to other year since 2000,which together led to the strongest glacier mass loss during 2022–*** atmospheric circulation analysis shows that the high temperature in the Qilian Mountains in 2023 was jointly caused by the Arctic air mass and East Asian monsoon.

This work presents a generic strategy to create a series of metal mercaptides complexes via coordination selfassembly between transition metals(Mn,Cu,Co,Fe,and Ni)and cysteine(Cys)by forming the sulfur-metal *** strategy involves dissolving metal chlorides and Cys into deep eutectic solvents(DES),followed by the precipitation of metal mercaptides complexes(such as Cys-Mn)by adding water as an antisolvent,where DES serves as the solvent,shape directing,and capping agent,thereby preventing the formation of other metal ***,the prepared complexes possess both laccase and peroxidase-like properties,allowing the design of a technique for the detection of L-3,4-dihydroxyphenylalanine(L-DOPA)and uric acid,*** prepared Cys-Mn can linearly oxidize L-DOPA with its concentrations from 0.1 to 130μM and the detection limit was calculated to be 75.5 n ***,the Cys-Mn can mimic the activity of peroxidase towards oxidization of o-phenylenediamine at neutral p H,allowing single-step and one-pot cascade reactions for visual and fluorometric measurements of uric acid(UA)that could work in the range of 0.2-500μM UA with a detection limit of 0.06μM and 0.054μМ,*** assay was successful in detecting UA in serum and urine samples with relative standard deviation(RSD)ranging from 7.3%to 10.2%and 3.0%-8.5%respectively,suggesting that it may prove useful in medical diagnostic testing.
