LncRNA pol-lnc78 as a ceRNA regulates antibacterial responses via suppression of pol-miR-n199-3p-mediated SARM down-regulation in Paralichthys olivaceus

Long non-coding RNAs(lncRNAs)function as key modulators in mammalian immunity,particularly due to their involvement in lncRNA-mediated competitive endogenous RNA(ceRNA)*** their recognized significance in mammals,research on lncRNAs in lower vertebrates remains *** the present study,we characterized the first immune-related lncRNA(pol-lnc78)in the teleost Japanese flounder(Paralichthys olivaceus).Results indicated that pol-lnc78 acted as a ceRNA for pol-miR-n199-3p to target the sterile alpha and armadillo motif-containing protein(SARM),the fifth discovered member of the Toll/interleukin 1(IL-1)receptor(TIR)adaptor *** ceRNA network regulated the antibacterial responses of flounder via the Toll-like receptor(TLR)signaling ***,SARM acted as a negative regulator and exacerbated bacterial infection by inhibiting the expression of inflammatory cytokines IL-1βand tumor necrosis factor-α(TNF-α).Pol-miR-n199-3p reduced SARM expression by specifically interacting with the 3’untranslated region(UTR),thereby promoting SARM-dependent inflammatory cytokine expression and protecting the host against bacterial ***,pol-lnc78 sponged pol-miR-n199-3p to ameliorate the inhibition of SARM *** infection,the negative regulators pol-lnc78 and SARM were significantly down-regulated,while pol-miR-n199-3p was significantly up-regulated,thus favoring host antibacterial *** findings provide novel insights into the mechanisms underlying fish immunity and open new horizons to better understand ceRNA crosstalk in lower vertebrates.

Background Culex pipiens pallens andCulex pipiens quinquefasciatus are the dominant species ofCulex mosquitoes in China and important disease vectors. Long-term use of insecticides can cause mutations in the voltage-gated sodium channel (vgsc) gene of mosquitoes, but little is known about the current status and evolutionary origins ofvgsc gene in different geographic populations. Therefore, this study aimed to determine the current status ofvgsc genes inCx. p. pallens andCx. p. quinquefasciatus in China and to investigate the evolutionary inheritance of neighboring downstream introns of thevgsc gene to determine the impact of insecticides on long-term *** Sampling was conducted from July to September 2021 in representative habitats of 22 provincial-level administrative divisions in China. Genomic DNA was extracted from 1308 mosquitoes, the IIS6 fragment of thevgsc gene on the nerve cell membrane was amplified using polymerase chain reaction, and the sequence was used to evaluate allele frequency and knockdown resistance (kdr) frequency. MEGA 11 was used to construct neighborjoining (NJ) tree. PopART was used to build a TCS *** There were 6 alleles and 6 genotypes at the L1014 locus, which included the wild-type alleles TTA/L and CTA/L and the mutant alleles TTT/F, TTC/F, TCT/S and TCA/S. The geographic populations with akdr frequency less than 20.00% were mainly concentrated in the regions north of 38° N, and the geographic populations with akdr frequency greater than 80.00% were concentrated in the regions south of 30° *** frequency increased with decreasing latitude. The World Health Organization (WHO) 2017 classifications for neuroendocrine neoplasms (NENs) subdivided grade 3 pancreatic neuroendocrine neoplasms (pNENs) into G3 well-differentiated pancreatic neuroendocrine tumors (G3 pNETs) and poorly differentiated pancreatic neuroendocrine carcinomas (pNECs), according to the mitotic count, Ki-67 index, and cell differentiation. As a new category, G3 pNETs remain a challenging group of tumors to manage by lacking large randomized trials and consensus to support its clinical practice. Therefore, the Chinese Pancreatic Surgery Association, Chinese Society of Surgery, Chinese Medical Association gathered experts in this field to formulate this consensus for the diagnosis and treatment of G3 pNETs.

Cardiac resident macrophages: Spatiotemporal distribution, development, physiological functions, and their translational potential on cardiac diseases

Cardiac resident macrophages (CRMs) are the main population of cardiac immune cells. The role of these cells in regeneration, functional remodeling, and repair after cardiac injury is always the focus of research. However, in recent years, their dynamic changes and contributions in physiological states have a significant attention. CRMs have specific phenotypes and functions in different cardiac chambers or locations of the heart and at different stages. They further show specific differentiation and development processes. The present review will summarize the new progress about the spatiotemporal distribution, potential developmental regulation, and their roles in cardiac development and aging as well as the translational potential of CRMs on cardiac diseases. Of course, the research tools for CRMs, their respective advantages and disadvantages, and key issues on CRMs will further be discussed.

Complex networked systems, which range from biological systems in the natural world to infrastructure systems in the human-made world, can exhibit spontaneous recovery after a failure; for example, a brain may spontaneously return to normal after a seizure, and traffic flow can become smooth again after a jam. Previous studies on the spontaneous recovery of dynamical networks have been limited to undirected networks. However, most real-world networks are directed. To fill this gap, we build a model in which nodes may alternately fail and recover, and we develop a theoretical tool to analyze the recovery properties of directed dynamical networks. We find that the tool can accurately predict the final fraction of active nodes, and the prediction accuracy decreases as the fraction of bidirectional links in the network increases, which emphasizes the importance of directionality in network dynamics. Due to different initial states, directed dynamical networks may show alternative stable states under the same control parameter, exhibiting hysteresis behavior. In addition, for networks with finite sizes, the fraction of active nodes may jump back and forth between high and low states, mimicking repetitive failure-recovery processes. These findings could help clarify the system recovery mechanism and enable better design of networked systems with high resilience.

Polyphenol-rich extract from Sanghuangporous vaninii against acetaminophen-induced liver injury by blocking ferroptosis via NRF2/GPX4 pathway

Acetaminophen (APAP) used as an antipyretic and analgesic agent can cause acute liver injury (AILI) under overdose. Sanghuangporous vaninii (S. vaninii) is a edible fungus with abundant metabolites exhibits excellent hepatoprotective activities， but the effect for AILI is not yet fully understood. In this study， the polyphenol-rich extract from S. vaninii (PSV) was prepared， with a total phenolic content of 75.72% and 34 compounds. The data of hepatoprotection indicated that PSV obviously alleviated the hepatocellular injury induced by APAP in vivo and in vitro. The protective mechanism of PSV against APAP-induced AILI might be attributed to the activating NRF2/GPX4 pathway. Based on network pharmacology analysis， the active components of PSV such as caffeic acid， osmundacetone and hispolone played a key role in hepatoprotection of PSV. Consequently， this study highlights the protection of PSV on AILI， which provides new insight into bioactivities of S. vaninii.

Farmers’ Knowledge and Awareness towards the Use of Agrochemicals in Vegetable Cultivation, Fako Cameroon

Agrochemicals are contemporary, omnipresent tool used in vegetable cultivation. Farmers’ knowledge and awareness of the proper usage of agrochemicals are critical for mitigating the negative effects on human health. This cross-sectional study was aimed at assessing the usage knowledge, risk awareness of toxicological and chemical classes, proper handling and use practices for agrochemicals homologated for use in vegetable farming, and the occurrence of health-related symptoms as a result of exposure among these farmers. The study included 93 vegetable growers from agricultural hotspot towns in Fako, southwest Cameroon. The field study, ran from November 2021 to December 2023, using a questionnaire to collect information on farmers demographic, and their knowledge of pesticide classes, and the related risk of associated with the handling of agrochemicals. Results show that all vegetable farmers, particularly those engaged in agribusiness, employ pesticide inputs to maximize production. Six pesticides, two fertilizer types, and one unknown substance were identified. While 23 active compounds were found, the most utilized were abamectin, emamectin (10.46%), dimethoate (9.30%,) and ethoprophos (8.13%). Two active chemicals, dimethoate and methalaxyl, are illegal yet remain in circulation. Toxicological classes I and II, with the greatest harmful effect on human health, were the most commonly utilized (64.27%). Thirty-nine percent of farmers never use personal protection equipment when working with agrochemicals, demonstrating a significant gap in knowledge and awareness of agrochemicals and their various applications and handling procedures in the field. The government should implement an intensive specialized educational program for on-field farmers with incentives in order to promote sustainable agriculture methods that ensure environmental and human safety.
