In this editorial,we discuss the findings reported by Wang et al in the latest issue of the World Journal of gastrointestinal *** research methodologies,including microbiome analysis,assert that the Tzu-Chi Cancer-Antagonizing and Life-Protecting II Decoction of Chinese herbal compounds mitigates inflammatory responses by inhibiting the NF-κB signaling *** action helps maintain the dynamic equilibrium of the intestinal microecology and lessens chemotherapy-induced gastrointestinal *** efficacy of these compounds is intimately linked to the composition of intestinal *** compounds regulate intestinal microecology by virtue of their specific compatibility and effectiveness,thereby enhancing the overall therapeutic outcomes of cancer ***,the exact mechanisms underlying these effects warrant further ***-omics technologies offer a systematic approach to elucidate the mechanisms and effectiveness of Chinese herbal compounds in *** manuscript reviews the application of multi-omics technologies to Chinese herbal compounds and explores their potential role in modulating the gastrointestinal microenvironment following cancer chemotherapy,thus providing a theoretical foundation for their continued use in adjunct cancer treatment.

Objective:To investigate the effects of Alpiniae oxyphyllae Fructus(AOF)on renal lipid deposition in diabetic kidney disease(DKD)and elucidate its molecular ***:The mechanism of AOF in treating DKD was explored by network pharmacological enrichment analysis,molecular docking,and molecular dynamics *** effects of AOF on renal function and lipid deposition were assessed in a mouse model of DKD and high glucose-stressed HK-2 *** viability and lipid accumulation were detected by CCK8 and oil red O *** expressions of PPARαand fatty acid oxidation-related genes(ACOX1 and CPT1A)were detected by quantitative RT-PCR,Western blot,and ***,PPARαknockdown was performed to examine the molecular mechanism of AOF in treating ***:Network pharmacological enrichment analysis,molecular docking,and molecular dynamics simulation showed that the active compounds in AOF targeted PPARαand thus transcriptionally regulated ACOX1 and *** lowered blood glucose,improved dyslipidemia,and attenuated renal injury in DKD ***-containing serum accentuated high glucose-induced decrease in cell viability and ameliorated lipid ***,it significantly upregulated the expression of PPARα,ACOX1,and CPT1A in both in vivo and in vitro experiments,which was reversed by PPARα***:AOF may promote fatty acid oxidation via PPARαto ameliorate renal lipid deposition in DKD.

Brain organoids encompass a large collection of in vitro stem cell–derived 3D culture systems that aim to recapitulate multiple aspects of in vivo brain development and ***,this review provides a brief introduction to the current state-of-the-art for neuroectoderm brain organoid development,emphasizing their biggest advantages in comparison with classical two-dimensional cell cultures and animal ***,despite their usefulness for developmental studies,a major limitation for most brain organoid models is the absence of contributing cell types from endodermal and mesodermal *** such,current research is highly investing towards the incorporation of a functional vasculature and the microglial immune *** this review,we will specifically focus on the development of immune-competent brain *** summarizing the different approaches applied to incorporate microglia,it is highlighted that immune-competent brain organoids are not only important for studying neuronal network formation,but also offer a clear future as a new tool to study inflammatory responses in vitro in 3D in a brainlike ***,our main focus here is to provide a comprehensive overview of assays to measure microglial phenotype and function within brain organoids,with an outlook on how these findings could better understand neuronal network development or restoration,as well as the influence of physical stress on microglia-containing brain ***,we would like to stress that even though the development of immune-competent brain organoids has largely evolved over the past decade,their full potential as a pre-clinical tool to study novel therapeutic approaches to halt or reduce inflammation-mediated neurodegeneration still needs to be explored and validated.

Single-cell and spatial omics:exploring hypothalamic heterogeneity

elucidating the complex dynamic cellular organization in the hypothalamus is critical for understanding its role in coordinating fundamental body functions. Over the past decade, single-cell and spatial omics technologies have significantly evolved, overcoming initial technical challenges in capturing and analyzing individual cells. These high-throughput omics technologies now offer a remarkable opportunity to comprehend the complex spatiotemporal patterns of transcriptional diversity and cell-type characteristics across the entire hypothalamus. Current single-cell and single-nucleus RNA sequencing methods comprehensively quantify gene expression by exploring distinct phenotypes across various subregions of the hypothalamus. However, single-cell/single-nucleus RNA sequencing requires isolating the cell/nuclei from the tissue, potentially resulting in the loss of spatial information concerning neuronal networks. Spatial transcriptomics methods, by bypassing the cell dissociation, can elucidate the intricate spatial organization of neural networks through their imaging and sequencing technologies. In this review, we highlight the applicative value of single-cell and spatial transcriptomics in exploring the complex molecular-genetic diversity of hypothalamic cell types, driven by recent high-throughput achievements.

Research Progress of Raman Spectroscopy and Imaging Techniques for the Pharmaceutical Analysis

Monitoring critical quality attributes in pharmaceutical manufacturing ensures that the final product meets specified requirements. Process analysis allows for appropriate modifications and customization of pharmaceutical design from formulation to production. The laborious sample presentation and throughput have stimulated the development of in-process spectroscopy methods for pharmaceutical analysis. Raman spectroscopy has demonstrated the unique advantages of rapid and nondestructive analysis, good interpretability, and effective moisture interference avoidance. The Raman imaging technique can obtain spatial and chemical information that is useful for characterizing drug quality. This article describes the applications of Raman spectroscopy and imaging techniques in pharmaceutical analysis. It covers the monitoring of crystal forms and particle size distribution, quantification of active pharmaceutical ingredients, evaluation of contents uniformity, prediction of dissolution profiles in solid preparations, identification of counterfeit drugs, and applications in reverse engineering and process analytical technology. Moreover, the challenges and prospects are proposed to facilitate the continued development of Raman spectroscopy and imaging techniques, providing necessary references for further research in this field.

Objective: To examine the effectiveness of Chinese medicine(CM) Lianhua Qingwen granule(LHQW) and Jingyin gubiao Prescription(JYgB) in asymptomatic or mild patients with Omicron infection in the shelter hospital. Methods: This single-center retrospective cohort study was conducted in the largest shelter hospital in Shanghai, China, from April 10, 2022 to May 30, 2022. A total of 56,244 asymptomatic and mild Omicron cases were included and divided into 4 groups, i.e., non-administration group(23,702 cases), LHQW group(11,576 cases), JYgB group(12,112 cases), and dual combination of LHQW and JYgB group(8,854 cases). The length of stay(LOS) in the hospital was used to assess the effectiveness of LHQW and JYgB treatment on Omicron infection. Results: Patients aged 41–60 years, with nadir cycle threshold(CT) value of N gene e fully vaccinated preferred to receive CM therapy. Before or after propensity score matching(PSM), the multiple linear regression showed that LHQW and JYgB treatment were independent influence factors of LOS(both Per PSM, there were significant differences in LOS between the LHQW/JYgB combination and the other groups(Pe results of factorial design ANOVA proved that the LHQW/JYgB combination therapy synergistically shortened LOS(P=0.032). Conclusions: Patients with a nadir CT value ere more likely to accept CM. The LHQW/JYgB combination therapy could shorten the LOS of Omicron-infected individuals in an isolated environment.
