Association of miR-499 rs3746444, miR-149 rs2292832 polymorphisms and their expression levels with helicobacter pylori-related gastric diseases and traditional chinese medicine syndromes

OBJECTIVE: To provide an objective experimental basis for the gastric mucosa pathological evolution and the transformation of different traditional chinese medicine(TCM) syndromes in helicobacter pylori(H. pylori)-related gastric diseases(HPGD) patients, based on the combination of TCM syndrome differentiation, molecular biology and ***: A total of 203 participants were enrolled in this study. The expressions of miR-499/miR-149 and *** infection in the gastric tissues from all participants were detected. The genotyping for miR-499 rs3746444 and miR-149 rs2292832 was ***: In H. pylori positive subjects, the proportion of precancerous gastric lesions(PGL) in liver-stomach disharmony syndrome(LSDS) group was higher than in spleen Qi deficiency syndrome(SQDS) group(P < 0.001);The proportion of gastric cancer(GC) in SQDS group was higher than in spleen-stomach damp-heat syndrome(SSDHS) group and LSDS group(all P < 0.001). We also found C allele of miR-149 rs2292832 was linked to lower risk of gastric atrophy [miR-149 rs2292832 C vs T:adjusted odds ratio = 0.207; 95% confidence interval(0.043-0.989); P = 0.048]. Compared with healthy control(HC) group, the expression of miR-499 was significantly increased in GC group, while the expression of miR-149was significantly decreased in chronic inflammation group,PGL group and GC group(all P < 0.05). Test for trend showed that GC risk was on a rising trend with the increasing expression of miR-499 and decreasing expression of miR-149(both P for trend < 0.05).CONCLUSION: The C allele of miR-149 rs2292832 may be a protective factor for gastric mucosal atrophy. *** may participate in the evolution of benign to malignant gastric mucosa lesions by inducing the overexpression of miR-499 and down regulation of miR-149. In addition, patients with H. pylori infection combined SQDS or LSDS may have higher risk of gastric mucosal malignant lesions.

Clinical efficacy and safety evaluation of Buzhongyiqi pills(补中益气丸) on appetite improvement in patients with colorectal cancer receiving chemotherapy: a pilot randomized cross-over clinical trial

OBJECTIVE: To evaluate the clinical efficacy and safety of Buzhongyiqi pills (BZYQP，补中益气丸) in improving the appetite of patients with colorectal cancer (CRC) receiving *** DESIGN: A pilot， randomized， single-blind cross-over clinical trial was conducted on diagnosed stage II-IV CRC patients receiving ***: Patients were randomly assigned to either the BZYQP-placebo or placebo-BZYQP groups. The BZYQP-placebo group received BZYQP for 1-2 days before the first cycle of chemotherapy and continued until the end of the third cycle. A 7-day washout followed， after which they received a placebo until the end of the sixth cycle. The placebo-BZYQP group followed the opposite treatment order. The oral dose of BZYQP and placebo was ten pills three times daily. A total of 12 visit points were scheduled in this study， with each visit point carried out before and after each of the six cycles of chemotherapy. The Simplified Nutrition Appetite Questionnaire (SNAQ)， the European Organization for Research and Treatment of Cancer Quality of Life Questionnaire (EORTC QLQ-C30， version 3.0)， and the National Cancer institute Common Terminology Criteria for Adverse Events (CTCAE， V5.0) were used to evaluate patient appetite， quality of life， and drug ***: Totally 62 patients completed the study， and baseline characteristics were balanced between the BZYQP-placebo and placebo-BZYQP groups. The primary outcome， as assessed by SNAQ scores， demonstrates a statistically significant difference between the two groups during the first three cycles of chemotherapy， with the mean SNAQ score of the BZYQP-placebo group consistently higher than that of the placebo-BZYQP group from V1 (P < 0.001). After the washout period， the SNAQ score of the BZYQP-placebo group decreased from V7， and the difference in SNAQ scores between the two groups gradually became more significant after the intersection at V9. In the realm of autoimmune and inflammatory diseases,the cyclic GMP-AMP synthase(cGAS)stimulator of interferon genes(STING)signaling pathway has been thoroughly investigated and *** this,the clinical approval of drugs targeting the cGAS-STING pathway has been *** Total glucosides of paeony(TGP)is highly anti-inflammatory and is commonly used in the treatment of rheumatoid arthritis(RA),emerged as a subject of our *** found that the TGP markedly reduced the activation of the cGAS-STING signaling pathway,triggered by various cGAS-STING agonists,in mouse bone marrow-derived macrophages(BMDMs)and Tohoku Hospital Pediatrics-1(THP-1)*** inhibition was noted alongside the suppression of interferon regulatory factor 3(IRF3)phosphorylation and the expression of interferon-beta(IFN-β),C-X-C motif chemokine ligand 10(CXCL10),and inflammatory mediators such as tumor necrosis factor-alpha(TNF-α)and interleukin-6(IL-6).The mechanism of action appeared to involve the TGP’s attenuation of the STING-IRF3 interaction,without affecting STING oligomerization,thereby inhibiting the activation of downstream signaling *** vivo,the TGP hindered the initiation of the cGAS-STING pathway by the STING agonist dimethylxanthenone-4-acetic acid(DMXAA)and exhibited promising therapeutic effects in a model of acute liver injury induced by lipopolysaccharide(LPS)and D-galactosamine(D-GalN).Our findings underscore the potential of the TGP as an effective inhibitor of the cGAS-STING pathway,offering a new treatment avenue for inflammatory and autoimmune diseases mediated by this pathway.
