Cortical Organoid-on-a-Chip with Physiological Hypoxia for Investigating Tanshinone IIA-Induced Neural Differentiation

Cortical organoids represent cutting-edge models for mimic human brain development during the early and even middle stage of pregnancy, while they often fail to recreate the complex microenvironmental factors, such as physiological hypoxia. Herein, to recapitulate fetal brain development, we propose a novel cortical organoid-on-a-chip with physiological hypoxia and further explore the effects of tanshinone IIA (Tan IIA) in neural differentiation. The microfluidic chip was designed with a micropillar array for the controlled and efficient generation of cortical organoids. With low oxygen, the generated cortical organoids could recapitulate key aspects of early-gestational human brain development. Compared to organoids in normoxic culturing condition, the promoted neurogenesis, synaptogenesis and neuronal maturation were observed in the present microsystem, suggesting the significance of physiological hypoxia in cortical development. Based on this model, we have found that chinese herbal drug Tan IIA could promote neural differentiation and maturation, indicating its potential therapeutic effects on neurodevelopmental disorders as well as congenital neuropsychiatric diseases. These results indicate that the proposed biomimetic cortical organoid-on-a-chip model with physiological hypoxia can offer a promising platform to simulate prenatal environment, explore brain development, and screen natural neuroactive components.

Anxiety Mouse Model Constructed by Single Intraperitoneal Injection of m-Chlorophenpiperazine

Anxiety disorder is a common mental disorder. It is necessary to establish a rapid, stable and specific anxiety model to provide a theoretical basis for further research on the pathogenesis of anxiety and drug development. A single intraperitoneal injection of m-chlorophenylpipera-zine (mCPP) (1, 2, 4 mg/kg) was given to male ICR mice to establish an anxiety model, and the effects of mCPP on anxiety behavior, pain, athletic ability, passive avoidance response ability and depressive behavior of male ICR mice were evaluated. A single intraperitoneal injection of mCPP shortened the time in open arms and decreased the percentage of time in open arms of mice in the elevated plus-maze test. mCPP also shortened center zone distance and reduced the number of entries to the central zone in the open field test. Moreover, mCPP reduced head-dip counts and increased the head-dip latency of mice in the hole-board test. After being administrated with a single intraperitoneal injection of mCPP for 24h, the mice showed no significant difference in the entry into the light side and the percentage of time in the light side of the light-dark box test. A single intraperitoneal injection of mCPP had no effects on tail flick latency, rotating time, number of errors and the step-down latency, the immobility time of mice in the tail-flick test, rotarod test, step-down test and TST respectively. In conclusion, we established a rapid and stable anxiety mouse model by single intraperitoneal injection of mCPP.

Hypomethylation of glycine dehydrogenase promoter in peripheral blood mononuclear cells is a new diagnostic marker of hepatitis B virus-associated hepatocellular carcinoma

Background: Glycine dehydrogenase(GLDC) plays an important role in the initiation and proliferation of several human cancers. In this study, we aimed to detect the methylation status of GLDC promoter and its diagnostic value for hepatitis B virus-associated hepatocellular carcinoma(HBV-HCC). Methods: We enrolled 197 patients, 111 with HBV-HCC, 51 with chronic hepatitis B(CHB), and 35 healthy controls(HCs). The methylation status of GLDC promoter in peripheral mononuclear cells(PBMCs) was identified by methylation specific polymerase chain reaction(MSP). The mRNA expression was examined using real-time quantitative polymerase chain reaction(q PCR). Results: The methylation frequency of the GLDC promoter was significantly lower in HBV-HCC patients(27.0%) compared to that in CHB patients(68.6%) and HCs(74.3%)( P < 0.001). The methylated group had lower alanine aminotransferase level( P = 0.035) and lower rates of tumor node metastasis(TNM) Ⅲ/Ⅳ( P = 0.043) and T3/T4( P = 0.026). TNM stage was identified to be an independent factor for GLDC promoter methylation. GLDC mRNA levels in CHB patients and HCs were significantly lower than those in HBV-HCC patients( P = 0.022 and P < 0.001, respectively). GLDC mRNA levels were significantly higher in HBV-HCC patients with unmethylated GLDC promoters than those with methylated GLDC promoters( P = 0.003). The diagnostic accuracy of alpha-fetoprotein(AFP) combined with GLDC promoter methylation for HBV-HCC was improved compared with that of AFP alone(AUC: 0.782 vs. 0.630, P < 0.001). In addition, GLDC promoter methylation was an independent predictor for overall survival of HBV-HCC patients( P = 0.038). Conclusions: The methylation frequency of GLDC promoter was lower in PBMCs from HBV-HCC patients than that from patients with CHB and HCs. The combination of AFP and GLDC promoter hypomethylation significantly improved the diagnostic accuracy of HBV-HCC.

High-resolution remote thermometry based on Bi^(3+)-doped 0D zinc halide hybrids

Developing highly sensitive optical thermometers is of great significance due to their capability to enable remote and non-contact temperature measurements, rendering them highly applicable in diverse and harsh environments. Herein we report a temperature-dependent phosphor of 0D metal halide hybrid by incorporating Bi^(3+)into the(MePPh_(3))_(2)ZnCl_(4) matrix. Through Bi^(3+)doping,the initially non-luminescent(MePPh_(3))_(2) ZnCl_(4) matrix exhibits a deep-blue emission centered at 453 nm, with a photoluminescence quantum yield(PLQY) of 5.71% and a Stokes shift of 75 nm at room temperature. Experimental characterization demonstrates that exciton-like luminescence of Bi^(3+)is mainly responsible for the blue emission. Single crystals of Bi^(3+)-doped(MePPh_(3))_(2)ZnCl_(4) show an unusual correlation between photoluminescence(PL) lifetime and temperature. Particularly, the dependence of luminescence lifetime on temperature is most remarkable in the temperature range of 80 to 100 K with an exceptional sensitivity up to 0.09 K^(-1), representing one of the best levels for thermometry based on PL decay lifetime. Our work not only provides a viable strategy for designing a novel, environmentally friendly, and stable blue emitter, but also paves the way for precise thermometric application at low temperature.

Paclitaxel-lipid prodrug liposomes for improved drug delivery and breast carcinoma therapy

Pt nanodendrites with a PtIr alloy surface structure exhibit excellent stability toward acidic hydrogen evolution reaction

The development of effective and stable electrocatalysts for the hydrogen evolution reaction(HER)in acidic electrolytes is a significant *** this work,homogeneous Pt nanodendrites(Pt NDs)with a PtIr shell were successfully synthesized by a two-step wet chemical *** open three-dimensional(3D)dendritic structure exhibited exceptional electrocatalytic characteristics,exposing as many active sites as ***,by alloying Ir with Pt on the surface,catalytic activity was greatly enhanced while ensuring extremely high *** surface-enriched platinum nanodendritic catalysts(Pt@PtIr NDs)outperformed the control samples and the commercial *** acidic HER test,Pt@PtIr NDs had a lower overpotential(22 mV)than Pt NDs(26 mV)and commercial Pt/C(31 mV)at 10 mA/cm^(2),and the activity of Pt@PtIr NDs remained consistent even after undergoing a continuous durability test for at least 168 h,which was much superior to the performance of commercial Pt/C(10 h)under identical test *** study revealed that the application of 3D Pt dendritic metal alloys may offer a chance for the development of enhanced electrocatalysts in acidic HER.
