Objective To evaluate the effect of diisononyl phthalate（DINP） exposure during gestation and lactation on allergic response in pups and to explore the role of phosphoinositide 3-kinase/Akt pathway on it. Methods Female Wistar rats were treated with DINP at different dosages（0, 5, 50, and 500 mg/kg of body weight per day）. The pups were sensitized and challenged by ovalbumin（OVA）. The airway response was assessed; the airway histological studies were performed by hematoxylin and eosin（HE） staining; and the relative cytokines in phosphoinositide 3-kinase（PI3K）/Akt pathway were measured by enzyme-linked immunosorbent assay（ELISA） and western blot analysis. Results There was no significant difference in DINP＇s effect on airway hyperresponsiveness（AHR） between male pups and female pups. In the 50 mg/（kg·d） DINP-treated group, airway response to OVA significantly increased and pups showed dramatically enhanced pulmonary resistance（RI） compared with those from controls（P〈0.05）. Enhanced Akt phosphorylation and NF-κB translocation, and Th2 cytokines expression were observed in pups of 50 mg/（kg·d） DINP-treated group. However, in the 5 and 500 mg/（kg·d） DINP-treated pups, no significant effects were observed. Conclusion There was an adjuvant effect of DINP on allergic airway inflammation in pups. Maternal DINP exposure could promote OVA-induced allergic airway response in pups in part by upregulation of PI3K/Akt pathway.

Association between Phthalate Exposure and the Use of Plastic Containers in Shanghai Adults

Objective Consuming phthalates may be due to the presence of food contact materials, such as plastic containers. In this study, we investigated the association between plastic container use and phthalate exposure in 2,140 Shanghai adults. Methods Participants completed a questionnaire on the frequency of using plastic containers in different scenarios in the previous year （e.g., daily, weekly） and on the consumption of plastic-packaged foods in the previous three days （yes or no）. Urinary phthalate metabolites were used to assess the association between phthalate exposure and the use of plastic containers. Results The metabolites of di-（2-ethylhexyl） phthalate （DEHP） were the most frequently detected in urine. The results revealed that phthalate exposure was associated with consumption of plastic-packaged breakfast or processed food items in the previous three days. The consumption of these two food items had strong synergistic effects on increasing urinary concentrations of most phthalate metabolites. Conclusion Our results of plastic-packaged breakfast and processed food may be explained by the use of flexible plastic containers, indicating the importance of risk assessment for the application of flexible plastic containers.

Phenome-Wide Association Analysis Reveals Novel Links Between Genetically Determined Levels of Liver Enzymes and Disease Phenotypes

Serum liver enzymes(alanine aminotransferase[ALT],aspartate aminotransferase[AST],λ-glutamyl transferase[GGT]and alkaline phosphatase[ALP])are the leading biomarkers to measure liver injury,and they have been reported to be associated with several intrahepatic and extrahepatic diseases in observational studies.We conducted a phenome-wide association study(PheWAS)to identify disease phenotypes associated with genetically predicted liver enzymes based on the UK Biobank cohort.Univariable and multivariable Mendelian randomization(MR)analyses were performed to obtain the causal esti-mates of associations that detected in PheWAS.Our PheWAS identified 40 out of 1,376 pairs(16,17,three and four pairs for ALT,AST,GGT and ALP,respectively)of genotype-phenotype associations reaching statistical significance at the 5%false discovery rate threshold.A total of 34 links were further validated in Mendelian randomization analyses.Most of the disease phenotypes that associated with genetically determined ALT level were liver-related,including primary liver cancer and alcoholic liver damage.The disease outcomes associated with genetically determined AST involved a wide range of phenotypic categories including endocrine/metabolic diseases,digestive diseases,and neurological disorder.Genetically predicted GGT level was associated with the risk of other chronic non-alcoholic liver disease,abnormal results of function study of liver,and cholelithiasis.Genetically determined ALP level was associated with pulmonary heart disease,phlebitis and thrombophlebitis of lower extremities,and hypercholesterolemia.Our findings reveal novel links between liver enzymes and disease phenotypes providing insights into the full understanding of the biological roles of liver enzymes.

Summary What is already known about this topic?Limited evidence on healthy longevity was provided in the world,and no studies investigated the fractions of healthy longevity attributed to modifiable factors.What is added by this report?Incidences of longevity and healthy longevity in China are provided.It reveals that the total weighted population attributable fractions for lifestyles and all modifiable factors were 32.8% and 83.7% for longevity,respectively,and 30.4% and 73.4% for healthy longevity,respectively.What are the implications for public health practice?China has a high potential for longevity and healthy longevity.Strategies may be targeted at education and residence in early life as well as healthy lifestyles,disease prevention,and functional optimization in late life.

Objective The study is to investigate the associations between visibility, major air pollutants and daily counts of hospital admission in Shanghai, China. Methods Daily data on hospital admission, visibility, and air pollution during 2005‐2008 were obtained from the Shanghai Insurance Bureau （SHIB）, Shanghai Meteorological Bureau, and Shanghai Environmental Monitoring Center, respectively. The generalized additive model （GAM） with penalized splines was used to examine the associations between daily visibility and hospital admission. Results Among various pollutants, PM 2.5 showed strongest correlation with visibility. Decreased visibility was significantly associated with increased risk of hospital admission in Shanghai. An inter‐quartile range decrease in the 2‐day （L01） moving average of visibility corresponded to 3.66% （95%CI： 1.02%, 6.31%）, 4.06% （95%CI： 0.84%, 7.27%）, and 4.32% （95%CI： 1.67%, 6.97%） increase of total, cardiovascular, and respiratory hospitalizations, respectively. Conclusion Our analyses provide the first piece of evidence in China, demonstrating that decreased visibility has an effect on hospital admission, and this finding strengthens the rationale for further limiting air pollution levels in Shanghai.

Epidemiological studies have found that individuals with diabetes mellitus（DM） display an increased susceptibility for adverse cardiovascular outcomes when exposed to air pollution.This study was conducted to explore the potential mechanism linking ambient fine particles（PM2.5） and heart injury in a Type 2 DM（T2DM） animal model. The KKay mouse, an animal model of T2DM, was exposed to concentrated ambient PM2.5 or filtered air for 8 weeks via a versatile aerosol exposure and concentrator system. Simultaneously, an inhibitor of IκB kinase-2（IKK-a）（IMD-0354）, which is a blocker of nuclear factor κB（NF-κB）nuclear translocation, was administrated by intracerebroventricular injection（ICV） to regulate the NF-êB pathway. The results showed that ambient PM2.5 induced the increase of, NF-êB, cyclooxygenase-2（COX-2） and mitogen activated protein kinase（MAPK） expression in cardiac tissue, and that IMD-0354 could alleviate the inflammatory injury. The results suggested that the NF-êB pathway plays an important role in mediating the PM2.5-induced cardiovascular injury in the T2DM model. Inhibiting NFκB may be a therapeutic option in air-pollution-exacerbated cardiovascular injury in diabetes mellitus.

Diabetes,marked by an elevated concentration of blood glucose,presents a substantial global public health challenge.It is estimated that around 537 million adults(20-79 years)globally suffer from diabetes,primarily type 2 diabetes in 2021.This figure is projected to increase to 783 million by 2045.As of 2021,the countries with the greatest number of adults living with diabetes are China,India,and Pakistan(1).According to data obtained from the China Chronic Disease and Risk Factors Surveillance,the estimated prevalence of diabetes was 12.4%in 2018(2).Diabetes is acknowledged as a multifaceted cardiorenal-metabolic ailment associated with diverse metabolic and homeostatic disturbances that evolve over time(3).Although genetics contribute to the susceptibility to diabetes,increasing evidence suggests that modifiable risk factors,such as lifestyle(including nutrition and physical activity)and socioeconomic status(SES),play a pivotal role in both preventing and managing the disease.
