The aim of this cross-sectional survey was to analyze the semen parameters of volunteers from the Human Sperm Bank of fudan university,as well as the related factors influencing these *** January 2019 to December 2020,semen parameters from a total of 5214 men were included in this *** Kruskal-Wallis test was used to detect differences associated with several independent variables.A total of 5214 volunteers were *** volunteers were registered in 33 provinces,autonomous regions,municipalities(including Macao and Taiwan)and 294 prefecture-level *** average age of volunteers was 27.40 ***,76.50%of the volunteers had a college education or *** with BMI values of 18.5-23.9 kg/m2 accounted for 60.70%of *** parameters were significantly different according to season,education level,duration of abstinence,age group and *** basic male fertility phenotypes(semen parameters)showed new trends in the study period,and accurate long-term tracking of male semen parameters will help researchers to better understand the changes in male fertility phenotypes(semen).

Particle induced X- ray emission (PIXE) and Rutherford backscattering spectrometry (RBS) have been used extensively for analytical purpose because of their quantitative accuracy, reliability, simplicity and capability of non- destructive and multielement analysis. When these techniques are combined with a scanning microbeam system and a data acquisition system, three dimensional distribution of elemental composition can be displayed. Samples analyzed so far at fudan university include a microelectronic circuit and some biological and archaeological samples. The PIXE and RBS spectra and the secondary electron images have been measured.

For a long time, researchers have sought artificial intelligence(AI) that matches or exceeds human *** agents, which are artificial entities capable of sensing the environment, making decisions, and taking actions, are seen as a means to achieve this goal. Extensive efforts have been made to develop AI agents, with a primary focus on refining algorithms or training strategies to enhance specific skills or particular task performance. The field, however, lacks a sufficiently general and powerful model to serve as a foundation for building general agents adaptable to diverse scenarios. With their versatile capabilities, large language models(LLMs) pave a promising path for the development of general AI agents, and substantial progress has been made in the realm of LLM-based agents. In this article, we conduct a comprehensive survey on LLM-based agents, covering their construction frameworks, application scenarios, and the exploration of societies built upon LLM-based agents. We also conclude some potential future directions and open problems in this flourishing field.

Tumor suppressor p53-mediated cell cycle arrest and DNA damage repair may exert cytoprotective effects against cancer therapies,including WEE1 *** that p53 activation can also lead to multiple types of cell death,the role of this tumor suppressor in WEE1 inhibitor-based therapies remains *** this study,we reported that nucleolar stress-mediated p53 activation enhanced the WEE1 inhibitor AZD1775-induced ferroptosis to suppress lung cancer *** findings showed that AZD1775 promoted ferroptosis by blocking cystine uptake,an action similar to that of ***,inhibition of WEE1 by the WEE1 inhibitors or siRNAs induced compensatory upregulation of SLC7A11,which conferred resistance to ***,AZD1775 prevented the enrichment of H3K9me3,a histone marker of transcriptional repression,on the SLC7A11 promoter by repressing the expression of the histone methyltransferase SETDB1,thereby enhancing NRF2-mediated SLC7A11 *** finding was also validated using the H3K9me3 inhibitor ***,we found that the nucleolar stress-inducing agent Actinomycin D(Act.D)inhibited SLC7A11 expression by activating p53,thus augmenting AZD1775-induced ***,the combination of AZD1775 and Act.D synergistically suppressed wild-type p53-harboring lung cancer cell growth both in vitro and in ***,our study demonstrates that AZD1775 promotes ferroptosis by targeting cystine uptake but also mediates the adaptive activation of SLC7A11 through the WEE1-SETDB1 cascade and NRF2-induced transcription,and inhibition of SLC7A11 by Act.D boosts the anti-tumor efficacy of AZD1775 by enhancing ferroptosis in cancers with wild-type p53.

Data Assimilation(DA) plays a crucial role in modern numerical forecasting(Kalnay, 2002). It has been demonstrated that incorporating real-time observational data into forecast models significantly enhances the accuracy of both shortterm weather forecasts(Bauer et al., 2015;Bannister, 2017)and long-term climate predictions(Karspeck et al., 2013;Mochizuki et al., 2016).

Neurodegenerative diseases cause great medical and economic burdens for both patients and society;however, the complex molecular mechanisms thereof are not yet well understood. With the development of high-coverage sequencing technology, researchers have started to notice that genomic repeat regions, previously neglected in search of disease culprits, are active contributors to multiple neurodegenerative diseases. In this review, we describe the association between repeat element variants and multiple degenerative diseases through genome-wide association studies and targeted sequencing. We discuss the identification of disease-relevant repeat element variants, further powered by the advancement of long-read sequencing technologies and their related tools, and summarize recent findings in the molecular mechanisms of repeat element variants in brain degeneration, such as those causing transcriptional silencing or RNA-mediated gain of toxic function. Furthermore, we describe how in silico predictions using innovative computational models, such as deep learning language models, could enhance and accelerate our understanding of the functional impact of repeat element variants. Finally, we discuss future directions to advance current findings for a better understanding of neurodegenerative diseases and the clinical applications of genomic repeat elements.

A dual-protease-triggered chemiluminescent probe for precise tumor imaging

Imaging detection of interlinked dual proteases is imperative for precise tumor imaging, which remains challenging due to limited modification position of specific substrate and possible steric ***, we have developed a unimolecular chemiluminescent probe(LGP-CL) tandemly activated by two proteases interlinked with liver cancer to achieve precise tumor imaging. Probe LGP-CL consists of a phenoxy-dioxetane scaffold caged by a tripeptide substrate(LGP, leucine-glycine-proline) as the sensing layer, which can be cleaved sequentially by aminopeptidase N(APN) and dipeptidyl peptidase IV(DPPIV)to turn on a strong chemiluminescent signal, and silenced by specific inhibitor of each enzyme, which accounts for an integrated logic gate(AND, OR and INHIBIT). The successful cleavage of dual proteases on the metabolic site depends on the proper structure of the tripeptide substrate, as confirmed by two probes design. Probe LGP-CL(LGP as the substrate) enables the excellent “dual-lock-dual-key” fit with a382-fold enhancement of chemiluminescent emission while no obvious signal is observed by using GPLCL(GPL as the substrate). By virtue of its rapid response(several minutes), high sensitivity and good cell viability, probe LGP-CL has been utilized to evaluate upregulated levels of proteases in vitro and in living systems, especially to distinguish liver tumor cells(Hep G2) from others(LO2, MCF-7, MCF-10a and RAW264.7). Overall, the newly developed CL probe may facilitate rapid investigation into the role played by proteases in liver diseases, enabling timely selection appropriate treatment. Therefore, our work not only sheds light on the rational design of optical probes for dual protease imaging, but provides a promising tool for clinical diagnosis and even drug discovery.
