Based on the recently published whole-genome sequence of cultivated strawberry ’Camarosa’, in this study, 222FaWRKY genes were identified in the ’Camarosa’ genome. Phylogenetic analysis showed that the 222 FaWRKY candidate genes were classified into three groups, of which 41 were in group Ⅰ, 142 were in group Ⅱ, and 39 were in group Ⅲ. The 222 FaWRKY genes were evenly distributed among the seven chromosomes. The exon–intron structures and motifs of the WRKY genes had evolutionary diversity in different cultivated strawberry genomes. Regarding differential expression, the expression of FaWRKY133 was relatively high in leaves, while FaWRKY63 was specifically expressed in roots. FaWRKY207, 59, 46, 182, 156, 58, 39, 62 and 115 were up-regulated during achene development from the green to red fruit transition. FaWRK181, 166 and 211 were highly expressed in receptacles at the ripe fruit stage. One interesting finding was that Fa WRKY179 and 205 were significantly repressed after Colletotrichum fructicola inoculation in both ’Benihoppe’ and ’Sweet Charlie’ compared with Mock. The data reported here provide a foundation for further comparative genomics and analyses of the distinct expression patterns of FaWRKY genes in various tissues and in response to C. fructicola inoculation.

Through analysis of trap catches and fruit decay rate of different pear varieties,it was found that there were significant differences in parasitization of Grapholitha molesta（ Busck） among different pear varieties. The trap catches of Huangguan pear was the highest; the fruit decay rate of Xueqing pear was extremely higher than those of other varieties; the trap catches of early red comice were significantly lower than those of other varieties,and no injured fruits were found.

Bud dormancy facilitates the survival of meristems under harsh environmental conditions.To elucidate how molecular responses to chilling accumulation controlling dormancy in peach buds,chromatin immunoprecipitation sequencing to identify the H3K27me3 modifications and RNA sequencing of two peach cultivars with pronounced differences in chilling requirement were carried out,the results showed that genes associated with abscisic acid and gibberellic acid signal pathways play key roles in dormancy regulation.The results demonstrated that peach flower bud differentiation occurred continuously in both cultivars during chilling accumulation,which was correlated with the transcript abundance of key genes involved in phytohormone metabolism and flower bud development under adverse conditions.The more increased strength in high chillingrequirement cultivar along with the chilling accumulation at the genome-wide level.The function of the dormancy-associated MADS-box gene PpDAM6 was identified,which is involved in leaf bud break in peach and flower development in transgenic Nicotiana tabacum(NC89).In addition,PpDAM6 was positively regulated by PpCBF,and the genes of putative dormancy-related and associated with metabolic pathways were proposed.Taken together,these results constituted a theoretical basis for elucidating the regulation of peach bud dormancy transition.

Optimization of Texture Determination of ‘Yali' by Texture Analyzer

[Objectives] This study was carried out to determine texture parameters for texture profile analysis （TPA）, and optimize the texture determination of ‘Yali’ by texture analyzer.[Methods] The traditional varieties of ‘Yali’ were taken as the materials, and texture parameters were determined at different compression rates and deformations at target.[Results] In the process of the TPA, the deformation at target had an extremely significant influence on 8 TPA texture parameters, namely, the hardness, cohesiveness, springiness, adhesiveness, gumminess, resilience, fracturability, and chewiness （ P ≤0.01）, while the compression rate had significant influence on the hardness and gumminess （ P ≤0.05）, had an extremely significant influence on fracturability （ P ≤0.01）, and had no significant influence on other 5 TPA parameters.[Conclusions] Taking the compression rate of 1 mm/s and 20% deformation at target as the experimental conditions for TPA could avoid the impact load of high speed on the tissue and objectively reflect the textural characteristics of ‘Yali’ pulp tissue.

[Objectives]This study was conducted to provide a theoretical basis for production and application of chicken manure compost.[Methods]With dry manure as a material,the variations of the major nutrients during simple composting process of dry chicken manure were studied,in order to get the best fermentation method.[Results]Chicken manure should not be preserved wet,but should be preserved after air-drying and fermented before use.Necessary moisture,simple sealing treatment and a certain amount of N element were conducive to simple fermentation of chicken manure.The treatments in which the chicken manure was wrapped around and compacted,added with an appropriate amount of water(50%-60%)and naturally fermented,or was wrapped around and compacted and added with an appropriate amount of water(50%-60%)and 0.2%of urea,were more appropriate for the production directly.[Conclusions]This study provides a theoretical basis for the actual production and application of chicken manure compost.

Seedless citrus has become one of the breeding goals due to its high edible ratio and convenience in fresh consumption and processing.In this study, the ‘Hongjiangcheng' sweet orange(WT) and its seedless mutant(MT) after cobalt-60 radiation were selected to study the formation metabolism of citrus seedless phenotype. Compared with WT, the MT had altered primary metabolite contents, as indicated by GC-MS analysis. The mature pollen of the MT was mostly distorted and shrunken, and the orange mutant exhibited significantly lower fertility than the WT. Through pollination experiments and paraffin sectioning of the MT, we observed self-compatibility during pollen tube germination in situ, in combination with the absence of natural parthenocarpy and arrested zygotic embryo development at the fourth week after pollination. From transcriptomic analyses of ovules in the fourth week, 815 differentially expressed genes(DEGs) were identified.Furthermore, according to the annotation of gene function and qRT-PCR analysis, Cs4g10930, Cs5g21900 and orange1.1t02243 were identified as candidate genes that may govern the mechanism of seedlessness. Finally, Agrobacterium-mediated transformation verified that the overexpression of Cs4g10930 and Cs5g21900 in Newhall navel orange calli inhibited embryoid production. This study provides a better understanding of seedless formation in citrus and two key genes that may play an important role in the early selection of seedless lines in citrus breeding programs.

Auxin has been regarded as the main signal molecule coordinating the growth and ripening of fruits in strawberry, the reference genomic system for Rosaceae. The mechanisms regulating auxin biosynthesis in strawberry are largely elusive. Recently, we demonstrated that two YUCCA genes are involved in flower and fruit development in cultivated strawberry. Here, we show that the woodland strawberry （Fragaria vesca L.） genome harbors nine loci for YUCCA genes and eight of them encode functional proteins. Transcription pattern in different plant organs was different for all eight FvYUCs. Functionality of the FvYUC6 gene was studied in transgenic strawberry over- expressing FvYUC6, which showed typical high-auxin pheno- types. Overexpression of FvYUC6 also delayed flowering and led to complete male sterility in F. vesta. Additionally, specific repression of FvYUC6 expression by RNA interference signifi- cantly inhibited vegetative growth and reduced plant fertility. The development of leaves, roots, flowers, and fruits was greatly affected in FvYUC6-repressed plants. Expression of a subset of auxin-responsive genes was well correlated with the changes of FvYUC6 transcript levels and free indole-3-acetic acid levels in transgenic strawberry. These observations are consistent with an important role of FvYUC6 in auxin synthesis, and support a main role of the gene product in vegetative and reproductive development in woodland strawberry.

Sugar from plant photosynthesis is a basic requirement for life activities.Sugar transporters are the proteins that mediate sugar allocation among or within source/sink organs.The transporters of the major facilitator superfamily(MFS)targeting carbohydrates represent the largest family of sugar transporters in many plants.Strawberry(Fragaria×ananassa Duchesne)is an important crop appreciated worldwide for its unique fruit flavor.The involvement of MFS sugar transporters(STs)in cultivated strawberry fruit sugar accumulation is largely unknown.In this work,we characterized the genetic variation associated with fruit soluble sugars in a collection including 154 varieties.Then,a total of 67 ST genes were identified in the v4.0 genome integrated with the v4.0.a2 protein database of F.vesca,the dominant subgenome provider for modern cultivated strawberry.Phylogenetic analysis updated the nomenclature of strawberry ST homoeologs.Both the chromosomal distribution and structural characteristics of the ST family were improved.Semi-RT-PCR analysis in nine tissues from cv.Benihoppe screened 34 highly expressed ST genes in fruits.In three varieties with dramatically differing fruit sugar levels,qPCR integrated with correlation analysis between ST transcript abundance and sugar content identified 13 sugar-correlated genes.The correlations were re-evaluated across 19 varieties,including major commercial cultivars grown in China.Finally,a model of the contribution of the sugar transporter system to subcellular sugar allocation in strawberry fruits was proposed.Our work highlights the involvement of STs in controlling strawberry fruit soluble sugars and provides candidates for the future functional study of STs in strawberry development and responses and a new approach for strawberry genetic engineering and molecular breeding.

Unlike traditional ridging, mulching broad ridges with a woven polypropylene fabric (WPF) can reduce soil evaporation during the drought season and avoid long saturation time in the root zone of pear trees during the rainy season. In this study, field experiments were conducted from 2017 to 2020 in a pear orchard in the North China Plain to investigate the effects of mulching broad ridges (0.3 m in height and 2 m in width) with WPF on soil temperature and moisture, nitrogen leaching, vegetative and reproductive growth of young pear trees(Pyrus bretschneideri Rehd.‘Yuluxiang’). The experiments involved two treatments, namely, control (traditional no-ridge planting without mulching) and mulching broad ridges with WPF (RM treatment). The results showed that the RM treatment increased soil moisture and temperature and decreased nitrogen leaching, resulting in vigorous growth of the young pear trees. Moreover, the RM treatment increased the tree trunk cross-sectional area and height of the young pear trees by 37%and 8%in 2020, respectively. The nitrate nitrogen content at the soil layer depth of 0-30 cm was significantly higher in the RM than that in control. Furthermore, the RM treatment significantly increased the fruit yield due to larger tree size. In addition, compared with control, significantly higher fruit soluble solid content of RM treatment was detected in 2020. High precipitation (423 mm) occurred during fruit enlargement stage in 2020, RM treatment decreased the rainfall infiltration in the ridge and the soil moisture in root region, resulting in the improvement of fruit quality, compared with control.Therefore, mulching broad ridges with WPF can be implemented to increase soil moisture during drought season, soil temperature, and nitrate nitrogen content, thereby improving the growth and fruit yield of young pear trees. Additionally, it can reduce soil moisture in the root zone during the rainy season and improve the fruit quality of the trees. Finally, it can reduce nitrate nitrogen leaching, thereby re

Infl uence of altitude and tree class on climate-growth relationships in a larch plantation in subtropical China

Precise quantifi cation of climate-growth relationships can make a major contribution to scientifi c forest management.However,whether diff erences in the response of growth to climate at diff erent altitudes remains unclear.To answer this,264 trees of Larix kaempferi from 88 plots,representing diff erent altitudinal ranges(1000-2100 m)and tree classes were sampled and used to develop tree-ring chronologies.tree-ring growth(TRG)was either positively(dominant)or negatively(intermediate and suppressed)correlated with climate in diff erent tree classes at diff erent altitudes.TRG was strongly correlated with growing season at low altitudes,but was less sensitive to climate at middle altitudes.It was mainly limited by precipitation and was highly sensitive to climate at low altitudes.Climate-growth relationships at high altitudes were opposite compared to those at low altitudes.TRG of dominant trees was more sensitive to climate change compared to intermediate and suppressed trees.Climate factors(annual temperatures;moisture,the number of frost-free days)had diff erent eff ects on tree-ring growth of diff erent tree classes along altitudinal gradients.It was concluded that the increase in summer temperatures decreased water availability,resulting in a signifi cant decline in growth rates after 2005 at lower altitudes.L.kaempferi is suitable for planting in middle altitudes and dominant trees were the best sampling choice for accurately assessing climate-growth relationships.
