Identification of two novel nodule-specific genes from Astragalus sinicus L. by suppressive subtractive hybridization

To identify the genes involved in nodule formation and to increase usable molecular probes, a cDNA library of Astragalus sinicus genes specifically expressed in infected roots by Mesorhizobium huakuii 7653R is generated using a PCR-based suppressive subtractive hybridization (SSH) technique with two mRNA populations of infected and uninfected control roots. Two nodule-specific genes, AsIIC259 and AsG2511, are identified from infected roots of A. sinicus. The amino acid sequences deduced from the open reading frames (ORFs) reveal that AsIIC259 and AsG2511 encodes a polypeptide with 134 and 58 amino acids, respectively. A signal peptide sequence is predicted with high probability at the N-termini of the AsIIC259 and AsG2511. The motif searches show that the deduced polypeptide of AsIIC259 con- tains two N-glycosylation sites, a cAMP- and cGMP-dependent protein kinase phosphorylation site and a casein kinase II phosphorylation site. BLASTP searches reveal that AsIIC259 putative protein displays a low degree of similarity to a unique nodulin from Lupinus luteus nodules. No significant identity is displayed over the predicted polypeptides of AsG2511 with any published sequences. Virtual Northern blot and semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) analyses indicate that the two genes are expressed exclusively in inoculated roots and that their expression is 2―4 d later than that of the leghaemoglobin (Lb) gene during nodule development.

Insights into the catalytic mechanism of a type I sulfide quinone oxidoreductase(SQR)from Acidithiobacillus caldus

Bacteria as indicators for finding oil and gas reservoirs:A case study of the Bikaner-Nagaur Basin,Rajasthan,India

Geo-microbial prospecting for hydrocarbons is an exploration method based on the seepage of light gaseous hydrocarbons from oil/gas reservoirs to the surface and their utilization by hydrocarbon oxidizing bacteria. These bacteria utilize hydrocarbon gases as their sole source of food and are found to be enriched in the near surface soils/sediments above the oil and gas reservoirs. The detection of anomalous populations of n-pentane and n-hexane oxidizing bacteria in the surface soils can help to evaluate the prospects for hydrocarbon exploration. A geo-microbial survey has been carried out in the Bikaner Nagaur basin to investigate the prospects for hydrocarbon exploration. In the present study, bacterial counts for n-pentane utilizing bacteria range between 2.0× 10^2 and 1.26× 10^6 cfu/gm and n-hexane utilizing bacteria range between 2.0× 10^2 and 1.21 × 10^6 cfu/gm. The bacterial concentration distribution maps show four distinct anomalies in the study area. The possibility of discovering oil or gas reservoirs using the microbiological method is emphasized by the tact that the hydrocarbon-oxidizing bacteria range between 103 and 106 cfu/gm in soil/sediment receiving hydrocarbon micro-seepages. In the present study area of the Bikaner Nagaur basin, n-pentane and n-hexane utilizing bacteria are found between 105 and 106 cfu/gm of soil sample, which is significant and thereby substantiates the seepage of lighter hydrocarbon accumulations from oil and gas reservoirs. Geo-microbial prospecting studies suggest that hydrocarbon micro-seepage of subsurface origin is present in the study area and indicate that the area has positive prospects for petroleum exploration.

Citrus family especially Grapefruit,has attained considerable attention due to the presence of a number of essential components that have cardiovascular and anti-hypertensive *** juice and essential oil extracted from eight recently released grapefruit cultivars were used to study physicochemical and antioxidant *** total soluble solids(TSS),titratable acidity(TA) and pH of juice samples extracted from various grapefruit varieties differed *** refractive index,specific gravity and optical rotation values for the oil varied from 1.473 to 1.396,0.863 to 0.847 and +93 to +86,*** percent 2,2-diphenyl-1-picrylhydrazyl radical activity(% DPPH activity) and ferric reducing antioxidant power(FRAP) values for grapefruit juice and peel oil varied from 24.06 to 18.79,2.91 to 1.44 mmol g^–1 and 84.87 to 74.73,7.76 to 5.73 mmol g^-1,*** were significant differences in physicochemical,antioxidant properties and volatile profiles of extracted juice and *** oil exhibited higher DPPH and FRAP values than the *** different components identified which accounted for over 99% of the volatile fraction,limonene,myrcene,and benzopyran were major components in all oil samples.

Gut microbial dysbiosis has been linked to many noncommunicable ***,little is known about specific gut microbiota composition and its correlated metabolites associated with molecular signatures underlying host response to ***,we describe the construction of a proteomic risk score based on 20 blood proteomic biomarkers,which have recently been identified as molecular signatures predicting the progression of the *** demonstrate that in our cohort of 990 healthy individuals without infection,this proteomic risk score is positively associated with proinflammatory cytokines mainly among older,but not younger,*** further discover that a core set of gut microbiota can accurately predict the above proteomic biomarkers among 301 individuals using a machine learning model and that these gut microbiota features are highly correlated with proinflammatory cytokines in another independent set of 366 *** metabolomics analysis suggests potential amino acid-related pathways linking gut microbiota to host metabolism and ***,our multi-omics analyses suggest that gut microbiota composition and function are closely related to inflammation and molecular signatures of host response to infection among healthy *** results may provide novel insights into the cross-talk between gut microbiota and host immune system.

Combinatorial mutagenesis of Bacillus amyloliquefaciens for efficient production of protease

As an important industrial enzyme,protease is widely used in feed,food and other *** present,the insufficient protease activity obtained from microorganisms cannot meet the purpose of industrial *** this study,Bacillus amyloliquefaciens with high protease production was screened from animal feces by plate transparent circle *** improve the production of protease,atmospheric room temperature plasma(ARTP)mutagenesis was used in the first round,protease activity reached 315.0 U/***,to enhance production of protease,^(60)Co-γ irradiation was used for combined mutagenesis,leading to protease activity of *** FMME ZK003 up to 355.0 U/***,to realize the efficient production of protease,after optimization of fermentation conditions,protease activity was increased to 456.9 U/***,protease activity of *** FMME ZK003 reached 823.0 U/mL in a 5 L *** results indicate that *** can efficiently produce protease,which provides a good foundation for the industrial production of protease.

Selenopeptides may be a valuable bioactive compound to promote gut microbiota-targeted therapeutic methods for intestinal disease and ***,limited information is available on the utilization of selenopeptides by gut microbiota,especially Selenium(Se)*** this purpose,the present study aimed to investigate the protective effect of selenopeptide(RYNA(Se)MNDYT,Se-P2,purity of≥95%)and its original peptide(RYNAMNDYT,P2,purity of≥95%)in vivo by the microbiota-metabolite axis and further analyze the potential contribution of Se biofortification to Se-P2 *** results showed that Se-P2 exhibits a higher protective effect on lipopolysaccharide(LPS)-induced inflammation than P2,including pathology of the colon and liver,which suggested that the bioactivity of P2 was promoted by the organic combination of ***,gut microbiota composition tended to be a healthy structure by Se-P2 pretreatment in LPS-injured mice,which had a positive effect on LPS-induced gut microbiota ***,only Se-P2 promoted an increase in the relative abundance of Lactobacillus,Alistipes,and Roseburia and a decrease in the relative abundance of Akkermansia,Erysipelatoclostridium,and Bacteroides in LPS-injured *** changes in gut microbiota were obviously correlated with the changes in metabolites and affected the metabolic pathways of valine,leucine,isoleucine,phenylalanine,tyrosine,and tryptophan biosynthesis and phenylalanine *** may be one of the key reasons for Se-P2 to exert bioactivity through the microbiota-metabolite ***,Se-biofortification in Se-enriched Cordyceps militaris affected the parental proteins of Se-P2 to modulate mitogen-activated protein kinase,GPI anchored protein,and carbohydrate metabolism,translation,folding,sorting and degradation,which may contribute to the bioactivity of *** study provides information on the effect of Se on selenopeptides in vivo,which further promotes the prospective

Background:The Huashanshen(HSS)dripping pill has been widely used in asthma for a long time in ***,the relaxant mechanism of HSS is not well ***:In this report,high performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry was used to identify the constituents in rat plasma after oral administration of ***-sensitized allergic asthma and isolated trachea were studied for the anti-asthmatic mechanism of ***:D-anisodamine,L-anisodamine,scopolamine and atropine were detected in the rat plasma containing *** was clear that the HSS inhibited the release of inflammatory mediators,regulated the balance of T-helper 1 and T-helper 2 to reduce the airway inflammation,and relaxed the tracheal smooth muscle by controlling the KCa channel,Ca^(2+)influx and release to reduce the airway ***:Atropine,anisodamine and scopolamine might be active compounds of HSS which inhibited the release of inflammatory mediators,regulated the balance of Th1/Th2,and relaxed the tracheal smooth muscle to reduce airway hyperresponsiveness.
