food allergy is a growing health issue worldwide and the demand for sensitive,robust and high-throughput analytical methods is *** recent years,mass spectrometry-based methods have been established for multiple food allergen *** the present study,a novel method was developed for the simultaneous detection of almond,cashew,peanut,and walnut allergens in bakery foods using liquid chromatography–mass *** unique to these four ingredients were extracted,followed by trypsin digestion,quadrupole time-of-flight(Q-TOF)mass spectrometry and bioinformatics *** raw data were processed by de-novo sequencing module plus PEAKS DB(database search)module of the PEAKS software to screen peptides specific to each nut *** thermal stability and uniqueness of these candidate peptides were further verified using triple quadrupole mass spectrometry(QQQ-MS)in multiple reaction monitoring(MRM)*** nut species was represented by four peptides,all of which were validated for label-free quantification(LFQ).Calibration curves were constructed with good linearity and correlation coefficient(r 2)greater than *** limits of detection(LODs)were determined to range from 0.11 to 0.31 mg/kg,and were compared with the reference doses proposed by Voluntary Incidental Trace Allergen Labelling(VITAL).The recoveries of the developed method in incurred bakery food matrices ranged from 72.5%to 92.1%with relative standard deviations(RSD)offood samples confirmed the presence of these *** conclusion,this method provides insight into the qualitative and quantitative detection of trace levels of nut allergens in bakery foods.

Multifunctional Ce-MOF@PdNPs with colorimetric fluorescent electrochemical activity for ultrasensitive and accurate detection of diethylstilbestrol

The development of biosensors is gaining tremendous attention in various fields due to their extraordinary advantages, however, their sensitivity and accuracy are still challenging. Herein, we proposed a novel multifunctional nanocomposite Ce-MOF@PdNPs (MOF = metal-organic framework, PdNPs = Pd nanoparticles)-mediated triple-readout aptasensor for accurate and reliable detection of diethylstilbestrol (DES), in which Ce-MOF@PdNPs exhibited excellent peroxidase (POD)-like activity, fluormetric, and electro conductive properties. In addition, enzymes-assisted target recycling amplification was utilized to improve the sensitivity, that is the specific binding of aptamer and DES triggered an Exo III enzyme-assisted recycling reaction. The generated F-DNA was captured by the H3 strand linked to Ce-MOF@PdNPs immobilized on the electrode, exposing cleavage sites and activating the *** enzyme-assisted recycling reaction, leading to the dissociation of Ce-MOF@PdNPs and a significant reduced electrochemical signal. The collected Ce-MOF@PdNPs solution also induced a proportional change in the color and fluorescence, achieving a colorimetric and fluormetric detection functionality. The detection limit under colorimetric mode was 0.16 and 0.76 ng/mL under fluorescence mode, and 0.87 pg/mL under electrochemical mode. This triple-readout aptasensor exhibits high sensitivity, selectivity and accuracy, providing a new idea for designing novel biosensing platforms for veterinary drug residue detection.

Preparation of lactic acid bacteria compound starter cultures based on pasting properties and its improvement of glutinous rice flour and dough

The effects of 5 lactic acid bacteria(LAB)fermentation on the pasting properties of glutinous rice flour were compared,and suitable fermentation strains were selected based on the changes of viscosity,setback value,and breakdown value to prepare LAB compound starter *** results revealed that Latilactobacillus sakei HSD004 and Lacticaseibacillus rhamnosus HSD005 had apparent advantages in increasing the viscosity and reducing the setback and breakdown values of glutinous rice *** particular,the compound starter created using the two abovementioned LAB in the ratio of 3:1 had better performance than that using a single LAB in improving the pasting properties and increasing the water and oil absorption capacity of glutinous rice ***,the gelatinization enthalpy of the fermented samples increased *** frozen glutinous rice dough stored for 28 days,the viscoelasticity of frozen dough prepared by compound starter was better than that of control dough,and the freezable water content was lower than that of control *** results indicate that compound LAB fermentation is a promising technology in the glutinous rice-based food processing industry,which has significance for its application.

The loading direction-dependent shear behavior of single-layer chiral graphene sheets at different temperatures is studied by molecular dynamics （MD） simulations. Our results show that the shear properties （such as shear stress-strain curves, buckling strains, and failure strains） of chiral graphene sheets strongly depend on the loading direction due to the structural asymmetry. The maximum values of both the critical buckling shear strain and the failure strain under positive shear deformation can be around 1.4 times higher than those under negative shear deformation. For a given chiral graphene sheet, both its failure strain and failure stress decrease with increasing temperature. In particular, the amplitude to wavelength ratio of wrinkles for different chiral graphene sheets under shear deformation using present MD simulations agrees well with that from the existing theory. These findings provide physical insights into the origins of the loading direction-dependent shear behavior of chiral graphene sheets and their potential applications in nanodevices.

Using 3-amino-4-cyanopyrazole, salicylaldehyde and dialkylphosphite as materials, a series of a-aminophosphonates containing pyrazole and 2-hydroxybenzyl units were synthesized under microwave irradiation without solvents and catalysts. The structures of the compounds were verified by IR, 1H NMR, J3C NMR and elemental analysis. The crystal structure of diisobutyl {a-[3- （4-cyano-lH-pyrazol）amino）]-N-（2-hydroxylbenzyl）}phosphonate （4d, C19H27N404P） was deter- mined by single-crystal X-ray diffraction. Compound 4d crystallizes in the orthorhombic system, space group Pbcn with a = 17.329（4）, b = 20.091（5）, c = 12.433（3）/k, V = 4328.7（17） A3, M,. = 406.42, Dc = 1.247 g/cm3, Z = 8, F（000） = 1728,μ = 0.158 mm-1, MoKa radiation （2 = 0.71073 A）, the final R = 0.064 and wR = 0.0169 for observed reflections with I 〉 2σ（I）. X-ray diffraction analysis reveals that two planes lie in 4d, and the dihedral angle is 85.76°. Intermolecular O（1）-H（1B）-O（2）, N（1）-H（1）-N（4）, N（3）-H（3）.-.N（2） and N（3）-H（3）...O（1） hydrogen bonds are found in the structure. All the compounds were evaluated for their antiviral and antitumor activities respectively. Among them, 4d and 4e showed moderate anti-TMV activities at 500 gg/mL, and 4e possessed excellent antitumor activity against PC3 cells at 10 gmol/L.

Copper (Cu) is a kind of micronutrient element that is essential for human ***,it is also considered as an environmental pollutant which is toxic to organisms at a high concentration ***,regarded as beneficial microorganisms for promoting human health,have functions of antioxidant capacity,immune-enhancing properties,intestinal barrier protection and *** studies have reported that probiotics show positive effects on alleviating and intervening heavy metals ***,evidence for relieving copper-induced toxicity by probiotics is still *** this study,we firstly conducted a zebrafish larvae model to screen out microorganisms which are helpful for CuSO_(4)toxicity resistance and one novel strain named as Bacillus coagulans XY2 was discovered with the best protective *** XY2 significantly reduced the mortality of zebrafish larvae exposed to 10μmol/L CuSO_(4)for 96 hr,as well as alleviated the neutrophils infiltration in the larvae lateral line under a 2 hr *** XY2 exhibited a high in vitro antioxidant activity and against CuSO_(4)-induced oxidative stress in zebrafish larvae by up-regulating sod1,gstp1 and cat gene transcriptional levels and relevant enzymatic ***_(4)stimulated the inflammation process resulting in obvious increases of gene il-1βand il-10 transcription,which were suppressed by *** XY2 ***,our results underline the bio-function of *** XY2 on protecting zebrafish larvae from copper toxicity,suggesting the potential application values of probiotics in copper toxicity alleviation on human and the environment.

Highly efficient synthesis of(R)-1,3-butanediol via anti-Prelog reduction of 4-hydroxy-2-butanone with absolute stereoselectivity by a newly isolated Pichia kudriavzevii

(R)-1,3-butanediol is an important pharmaceutical intermediate, and the synthesis of(R)-1,3-butanediol using green biological methods has recently been of interest for industrial application. Here, a novel strain QC-1 that efficiently transforms 4-hydroxy-2-butanone to(R)-1,3-butanediol was isolated from soil samples. Based on morphological, physiological, and biochemical tests and 5.8 S-internal transcribed spacer sequencing, the strain was identified as Pichia kudriavzevii QC-1. The reaction conditions were optimized to 35 ℃, pH 8.0, rotation speed 200 rpm, and 6:5 mass ratio of glucose to 4-hydroxy-2-butanone. Evaluation of the effects of 4-hydroxy-2-butanone concentrations on yield and cell survival rate showed that 85.60 g·L^-1 product accumulated, with an enantiomeric excess of more than 99%, when 30 g·L^-14-hydroxy-2-butanone was added at 0, 10, and 30 h in a 3-L bioreactor. Thus, strain QC-1 showed excellent catalytic activity and stereoselectivity for the synthesis of(R)-1,3-butanediol from 4-hydroxy-2-butanone.
