Biomimetic Phosphohydrolase Nanozyme Based on Defect-Engineered Metal-Organic Framework

Phosphodiester bonds are the backbone linkages of all the nucleic acids which store and transfer biological *** hydrolysis of phosphodiester bonds is of great importance to the replication,recombination,and damage repair of DNA as well as the maturation and processing of ***,the spontaneous scission of individual phosphodiester linkages is kinetically challenging under physiological conditions,with an estimated half-life of 30 million ***,we discover a defect-engineered Zr-metal-organic framework(Zr-MOF)nanozyme containing cluster-missing reo defects,named UiO-66 REO,that possesses intrinsic phosphodiesterase-like activity that allows for the cleavage of the phosphodiester bond in physiological condition(pH=7.4 and 37℃),outperforming the activityrecording *** atomic-level structure of the UiO-66 REO nanozyme has been directly identified by the synchrotron radiation absorption spectrum and integrated differential phase contrast-scanning transmission electron *** find that both the defective boundary regions and unusual Zr6-clusters formed in the reo phase of the UiO-66 REO nanozyme contribute to the improvement in efficiency of phosphodiesterase-like hydrolysis,which realizes the cleavage of DNA in mild *** work offers a new insight into biomimetic enzymology using crystal and defect engineering and opens up new possibilities for the development of low-cost,structurally stable and sustainable phosphodiesterase nanozymes for different biological applications.

Prostate cancer （PCa） results from a multistep process. This process includes initiation, which occurs through various aging events and multiple insults （such as chronic infection, inflammation and genetic instability through reactive oxygen species causing DNA double-strand breaks）, followed by a multistep process of progression. These steps include several genetic and epigenetic alterations, as well as alterations to the chromatin structure, which occur in response to the carcinogenic stress-related events that sustain proliferative signaling. Events such as evading growth suppressors, resisting cell death, enabling replicative immortality, inducing angiogenesis, and activating invasion and metastasis are readily observed. In addition, in conjunction with these critical drivers of carcinogenesis, other factors related to the etiopathogenesis of PCa, involving energy metabolism and evasion of the immune surveillance system, appear to be involved. In addition, when cancer spread and metastasis occur, the ＇tumor microenvironment＇ in the bone of PCa patients may provide a way to sustain dormancy or senescence and eventually establish a ＇seed and soil＇ site where PCa proliferation and growth may occur over time. When PCa is initiated and progression ensues, significant alterations in nuclear size, shape and heterochromatin （DNA transcription） organization are found, and key nuclear transcriptional and structural proteins, as well as multiple nuclear bodies can lead to precancerous and malignant changes. These series of cellular and tissue-related malignancy-associated events can be quantified to assess disease progression and management.

The monkeypox virus(MPXV)has triggered a current outbreak *** sequencing of MPXV and rapid tracing of genetic variants will benefit disease diagnosis and *** is a significant challenge but necessary to optimize the strategy and application of rapid full-length genome identification and to track variations of MPXV in clinical specimens with low viral loads,as it is one of the DNA viruses with the largest genome and the most AT-biased,and has a significant number of tandem *** we evaluated the performance of metagenomic and amplicon sequencing techniques,and three sequencing platforms in MPXV genome sequencing based on multiple clinical specimens of five mpox cases in Chinese *** rapidly identified the full-length genome of MPXV with the assembly of accurate tandem repeats in multiple clinical *** sequencing enables cost-effective and rapid sequencing of clinical specimens to obtain high-quality MPXV ***-generation sequencing facilitates the assembly of the terminal tandem repeat regions in the monkeypox virus genome and corrects a common misassembly in published ***,several intra-host single nucleotide variations were identified in the first imported mpox *** study offers an evaluation of various strategies aimed at identifying the complete genome of MPXV in clinical *** findings of this study will significantly enhance the surveillance of MPXV.

In this study,we propose an efficient numerical framework to attain the solution of the extended fisher-Kolmogorov(EFK)*** temporal derivative in the EFK equation is approximated by utilizing the Crank-Nicolson *** temporal discretization,the generalized finite difference method(GFDM)with supplementary nodes is utilized to address the nonlinear boundary value problems at each time *** supplementary nodes are distributed along the boundary to match the number of boundary *** incorporating supplementary nodes,the resulting nonlinear algebraic equations can effectively satisfy the governing equation and boundary conditions of the EFK *** demonstrate the efficacy of our approach,we present three numerical examples showcasing its performance in solving this nonlinear problem.

Quantum fisher information(QFI)associated with local metrology has been used to parameter estimation in open quantum *** this work,we calculated the QFI for a moving Unruh-DeWitt detector coupled with massless scalar fields in n-dimensional spacetime,and analyzed the behavior of QFI with various parameters,such as the dimension of spacetime,evolution time,and Unruh *** discovered that the QFI of state parameter decreases monotonically from 1 to 0 over ***,we noted that the QFI for small evolution times is several orders of magnitude higher than the QFI for long evolution *** also found that the value of QFI decreases at first and then stabilizes as the Unruh temperature *** was observed that the QFI depends on initial state parameterθ,and Fθis the maximum forθ=0 orθ=π,Fφis the maximum forθ=π/*** also obtain that the maximum value of QFI for state parameters varies for different spacetime dimensions with the same evolution time.

We study the local quantum fisher information(LQFI)in the mixed-spin Heisenberg XXZ *** the maximal and minimal LQFI are studied and the former is essential to determine the accuracy of the quantum parameter estimation,the latter can be well used to characterize the discord-type quantum *** investigate the effects of the temperature and the anisotropy parameter on the maximal LQFI and thus on the accuracy of the parameter *** we make use of the minimal LQFI to study the discord-type correlations of different site *** dimensions of the subsystems cause different values of the minimal LQFI which reflects the asymmetry of the discord-type *** addition,the site pairs at different positions of the spin chains have different minimal LQFI,which reveals the influence of the surrounding spins on the bipartite quantum *** results show that the LQFI obtained through a simple calculation process provides a convenient way to investigate the discord-type correlation in high-dimensional systems.

Exploring the associations between skeletal muscle echogenicity and physical function in aging adults:A systematic review with meta-analyses

Background:Assessment and quantification of skeletal muscle within the aging population is vital for diagnosis,treatment,and injury/disease *** clinical availability of assessing muscle quality through diagnostic ultrasound presents an opportunity to be utilized as a screening tool for function-limiting ***,relationships between muscle echogenicity and clinical functional assessments require authoritative ***,we aimed to(a) synthesize the literature to assess the relationships between skeletal muscle echogenicity and physical function in older adults(≥60 years),(b) perform pooled analyses of relationships between skeletal muscle echogenicity and physical function,and(c) perform sub-analyses to determine between-muscle ***:CINAHL,Embase,MEDLINE,PubMed,and Web of Science databases were systematically searched to identify articles relating skeletal muscle echogenicity to physical function in older ***-of-bias assessments were conducted along with funnel plot ***-analyses with and without sub-analyses for individual muscles were performed utilizing fisher's Z transformation for the most common measures of physical ***'s Z was back-transformed to Pearson's r for ***:Fifty-one articles(n=5095,female=~2759,male=~2301,72.5± 5.8 years,mean±SD(1 study did not provide sex descriptors))were extracted for review,with previously unpublished data obtained from the authors of 13 *** rectus femoris(n=34) and isometric knee extension strength(n=22) were the most accessed muscle and physical qualities,*** relationship between quadriceps echogenicity and knee extensor strength was moderate(n=2924,r=-0.36(95% confidence interval:-0.38 to-0.32),p <0.001),with all other meta-analyses(grip strength,walking speed,sit-to-stand,timed up-and-go) resulting in slightly weaker correlations(r:-0.34 to-0.23,all p <0.001).Sub-analyses determined minimal differences in predicti

Holevo bound independent of weight matrices for estimating two parameters of a qubit

Holevo bound plays an important role in quantum metrology as it sets the ultimate limit for multi-parameter estimations,which can be asymptotically *** for some trivial cases,the Holevo bound is implicitly defined and formulated with the help of weight *** we report the first instance of an intrinsic Holevo bound,namely,without any reference to weight matrices,in a nontrivial ***,we prove that the Holevo bound for estimating two parameters of a qubit is equivalent to the joint constraint imposed by two quantum Cramér–Rao bounds corresponding to symmetric and right logarithmic *** weightless form of Holevo bound enables us to determine the precise range of independent entries of the mean-square error matrix,i.e.,two variances and one covariance that quantify the precisions of the estimation,as illustrated by different estimation *** result sheds some new light on the relations between the Holevo bound and quantum Cramer–Rao *** generalizations are discussed.

Arabidopsis Forkhead-Associated Domain Protein 3 negatively regulates peroxisome division

Peroxisomes are ubiquitous and dynamic eukaryotic organelles capable of altering their abun- dance in response to environmental and developmental cues, yet the regulatory mechanism of plant peroxisome division/proliferation is unclear. To identify transcriptional regulators of the peroxisome division factor gene PEX 11b, we performed a nuclear pull-clown experiment and identified Arabidopsis Forkhead- Associated Domain Protein 3 （FHA3） as a novel protein that binds to the promoter of PEX 11b. Our data supported the conclusion that, in contrast to the previously identified HY5 HOMOLOG （HYH） protein that promotes the transcription of PEX 11b, FHA3 is a negative regulator of PEX 11b expression and peroxisome division.
