Fibroblast growth factor 1(FGF1) regulates vertebrate cell growth, proliferation and differentiation, and energy metabolism. In this study, we cloned rainbow trout(Oncorhynchus mykiss) fgf1 and fgf1a, prepared their recombinant proteins(rFGF1 and rFGF1a), and described the molecular mechanisms by which they improve glycolipid metabolism in carnivorous fish. A 31-d feeding trial was conducted to investigate whether they could enhance glycolipid metabolism in rainbow trout on high-carbohydrate diets(HCD). A total of 720 rainbow trout(8.9 ± 0.5 g) were equally divided into 4 groups: the chow diet(CD) group injected with PBS, the HCD group injected with PBS, the HCD group injected with rFGF1(400 ng/g body weight), and the HCD group injected with rFGF1a(400 ng/g body weight). The results showed that short-term HCD had a significant positive effect on the specific growth rate(SGR) of rainbow trout(P < 0.05). However, it led to an increase in crude fat, serum triglyceride(TG) and glucose content, as well as serum glutamic pyruvic transaminase(GPT) and glutamic oxalacetic transaminase(GOT) contents(P < 0.05), suggesting a negative health effect of HCD. Nevertheless, rFGF1 and rFGF1a showed beneficial therapeutic effects. They significantly reduced the crude fat content of the liver, serum TG, GOT, and GPT contents caused by HCD(P < 0.05). The upregulation in atgl, hsl, and acc2 m RNAs implied the promotion of TG catabolism. Moreover, rFGF1 and rFGF1a contributed to promoting lipolysis by activating the AMPK pathway and reducing lipid accumulation in the liver caused by HCD. In addition,the rFGF1 and r FGF1a-treated groups significantly reduced serum glucose levels and elevated hepatic glycogen content under HCD, and increased glucose uptake by hepatocytes. We observed a decrease in mRNA levels for pepck, g6pase, and pygl, along with an increase in mRNA levels for gys, glut2, and gk in the liver. Association of complement components with risk of colorectal cancer:A systematic review and meta-analysis

Microphysical Insights into a Tornadic Supercell from Dual-Polarization Radar Observations in Jiangsu, China on 14 May 2021

Vascular etiology is the second most prevalent cause of cognitive impairment ***-1,which is produced and secreted by endothelial cells and astrocytes,is implicated in the pathogenesis of ***,the way in which changes in astrocytic endothelin-1 lead to poststroke cognitive deficits following transient middle cerebral artery occlusion is not well ***,using mice in which astrocytic endothelin-1 was overexpressed,we found that the selective overexpression of endothelin-1 by astrocytic cells led to ischemic stroke-related dementia(1 hour of ischemia;7 days,28 days,or 3 months of reperfusion).We also revealed that astrocytic endothelin-1 overexpression contributed to the role of neural stem cell proliferation but impaired neurogenesis in the dentate gyrus of the hippocampus after middle cerebral artery *** proteome profiles and western blot analysis confirmed that levels of glial fibrillary acidic protein and peroxiredoxin 6,which were differentially expressed in the brain,were significantly increased in mice with astrocytic endothelin-1 overexpression in comparison with wild-type mice 28 days after ischemic ***,the levels of the enriched differentially expressed proteins were closely related to lipid metabolism,as indicated by Kyoto Encyclopedia of Genes and Genomes pathway *** chromatography-mass spectrometry nontargeted metabolite profiling of brain tissues showed that astrocytic endothelin-1 overexpression altered lipid metabolism products such as glycerol phosphatidylcholine,sphingomyelin,and phosphatidic ***,this study demonstrates that astrocytic endothelin-1 overexpression can impair hippocampal neurogenesis and that it is correlated with lipid metabolism in poststroke cognitive dysfunction.

Objective Studies have shown that electroacupuncture(EA)can alleviate cognitive impairments from Alzheimer’s disease(AD)by regulating the expression of adenosine monophosphate-activated protein kinase(AMPK),but the specific mechanism involved remains to be ***,this study explores the potential mechanism by which EA improves cognitive function from the perspective of mitochondrial *** The four-month-old transgenic mice with amyloid precursor protein(APP)/presenilin 1(PS1)and AMPKα1-subunit conditional knockout(AMPKα1-cKO)were used for *** evaluate the effects of EA treatment on cognitive function,the T-maze and Morris water maze were *** addition,chemical exchange saturation transfer,thioflavin staining,transmission electron microscopy,mitochondrial membrane potential,and Western blotting were used to examine the potential mechanisms underlying the effects of EA on APP/PS1 *** Both APP/PS1 mice and AMPKα1-cKO mice exhibited dysfunction in mitochondrial dynamics accompanied by learning and memory *** of the AMPK/peroxisome proliferator-activated receptor-γcoactivator-1α(PGC-1α)pathway increased pathological amyloid-β(Aβ)deposition and aggravated the dysfunction in mitochondrial *** addition,EA rescued learning and memory deficits in APP/PS1 mice by activating the AMPK/PGC-1αpathway,specifically by reducing pathological Aβdeposition,normalizing energy metabolism,protecting the structure and function of mitochondria,increasing the levels of mitochondrial fusion proteins,and downregulating the expression of fission ***,the therapeutic effect of EA on cognition in APP/PS1 mice was hindered by AMPKα1 *** The regulation of hippocampal mitochondrial dynamics and reduction in Aβdeposition via the AMPK/PGC-1αpathway are critical for the ability of EA to ameliorate cognitive impairment in APP/PS1 mice.

Aircraft Observation and Simulation of the Supercooled Liquid Water Layer in a Warm Conveyor Belt over North China

This paper studied a snow event over North China on 21 February 2017,using aircraft in-situ data,a Lagrangian analysis tool,and WRF simulations with different microphysical schemes to investigate the supercooled layer of warm conveyor belts(WCBs).Based on the aircraft data,we found a fine vertical structure within clouds in the WCB and highlighted a 1-2 km thin supercooled liquid water layer with a maximum Liquid Water Content(LWC) exceeding0.5 g kg^(-1) during the vertical aircraft *** the main features of thermodynamic profiles were essentially captured by both modeling schemes,the microphysical quantities exhibited large diversity with different microphysics *** conventional Morrison two-moment scheme showed remarkable agreement with in-situ observations,both in terms of the thermodynamic structure and the supercooled liquid water ***,the microphysical structure of the WCB clouds,in terms of LWC and IWC,was not apparent in HUJI fast bin *** reduce such uncertainty,future work may focus on improving the representation of microphysics in bin schemes with in-situ data and using similar assumptions for all schemes to isolate the impact of physics.
