Salsolinol as an RNA m~6A methylation inducer mediates dopaminergic neuronal death by regulating YAP1 and autophagy

Salsolinol(1-methyl-6,7-dihydroxy-1,2,3,4-tetrahydroisoquinoline,Sal)is a catechol isoquinoline that causes neurotoxicity and shares structural similarity with 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine,an environmental toxin that causes Parkinson's ***,the mechanism by which Sal mediates dopaminergic neuronal death remains *** this study,we found that Sal significantly enhanced the global level of N~6-methyladenosine(m~6A)RNA methylation in PC12 cells,mainly by inducing the downregulation of the expression of m~6A demethylases fat mass and obesity-associated protein(FTO)and alk B homolog 5(ALKBH5).RNA sequencing analysis showed that Sal downregulated the Hippo signaling *** m~6A reader YTH domain-containing family protein 2(YTHDF2)promoted the degradation of m~6A-containing Yes-associated protein 1(YAP1)mRNA,which is a downstream key effector in the Hippo signaling ***,downregulation of YAP1 promoted autophagy,indicating that the mutual regulation between YAP1 and autophagy can lead to *** findings reveal the role of Sal on m~6A RNA methylation and suggest that Sal may act as an RNA methylation inducer mediating dopaminergic neuronal death through YAP1 and *** results provide greater insights into the neurotoxic effects of catechol isoquinolines compared with other studies and may be a reference for assessing the involvement of RNA methylation in the pathogenesis of Parkinson's disease.

A phylogenetic analysis of members of the family Buccinidae was conducted using 18S rRNA gene, 28S rRNA gene and the mitochondrial cytochrome oxidase Ⅰ gene. We studied 18 species of Buccinidae that belong to eight different genera and inhabit the China coastal seas. We analyzed the patterns of divergence between an outgroup and basal ingroup taxa, the monophyly of the genus Neptunea, and the position of one unnamed species within the Buccinidae. A phylogenetic tree (neighbor-joining (NJ) method) was reconstructed based on the sequences of 18S rRNA, 28S rRNA and COI, with Rapana venosa as outgroup. The NJ tree indicated that the 18 species could be divided into five groups. The genus Buccinum was monophyletic, whereas Neptunea was shown to be paraphyletic since it included Siphonalia subdilatata and Neptunea sp., a new species. This novel species otherwise clustered consistently with Neptunea cumingi in three other phylogenetic trees, showing a low genetic distance and divergence percentage of sequences belonging to the genus Neptunea. A smaller genetic distance and a smaller difference of 18S rRNA, 28S rRNA and COI sequences between Neptunea cumingii and Neptunea arthritica cumingii confirmed them to be the same species.

Background Comorbidity of irritable bowel syndrome(IBS)and psychiatric disorders is common,and the prevalence of at least one psychiatric disorder has been reported as high as 80%among patients with *** To explore the association of anxiety-depressive disorders with IBS and its different subtypes,and to evaluate the associations of lifestyle habits,dietary habits and sleeping quality with *** A comparative cross-sectional study was conducted at the AL-Mahsama family Practice Center,Ismailia,*** was carried out between October 2019 and October *** were categorised into 175 patients with IBS,diagnosed using the Rome IV criteria,and 175 patients without IBS.A semistructured questionnaire was used to collect data on sociodemographic characteristics,lifestyle habits,dietary habits and sleep quality from both *** Hamilton Anxiety Rating Scale was used to assess anxiety symptoms,whereas the Beck Depression Inventory Second Edition was used to assess depression *** There was a high statistically significant difference between both groups with regard to age,education,occupation and socioeconomic status(SES),being a smoker,being physically inactive,having sleep disturbance and having irregular meals;being either obese or overweight was more reported in the IBS *** was a high statistically significant difference in the rate of anxiety and depression between patients with and without ***,moderate and severe anxiety were reported in 37.1%,42.9%and 20.0%of patients with IBS while most(80.0%)of the patients without IBS reported mild *** depression,mild,moderate and severe depression were reported in 60.0%,14.3%and 25.7%of the patients with IBS while most(82.9%)of the non-IBS participants reported mild *** The study shows a significant association between anxiety-depressive disorders and IBS,but no significant associations between anxiety-depressive disorders and IBS subtypes.

In Arabidopsis, the CLAVATA3/EMBRYO SURROUNDING REGION-RELATED （CLE） peptides play important roles in regulating proliferation and differentiation of plant-specific stem cells. Although receptors of CLEs are reported to be leucine-rich repeat receptor kinases, the mechanisms underlying CLE-induced receptor activation remain largely unknown. Here we show that SOMATIC EMBRYOGENESIS RECEPTOR KINASEs （SERKs） serve as co-receptors in CLE41/TDIF-PXY signaling to regulate plant vascular development. TDIF induces interaction of its receptor PXY with SERKs in vitro and in vivo. Furthermore, the serk1-1 serk2-1 bakl-5 mutant plants are less sensitive to TDIF, phenocopying the pxy mutant with a compromised promotion of procambial cell proliferation. Crystal structure of the PXY-TDIF-SERK2 complex reveals that the last amino acid of TDIF conserved among CLEs and other evolutionary-related peptides is important for the interaction between SERK2 and PXY. Taken together, our current study identifies SERKs as signaling components of the TDIF-PXY pathway and suggests a conserved activation mechanism of CLE receptors.
