sofosbuvir is one of the new direct-acting antiviral drugs against hepatitis C virus(HCV) infection. This drug has recently been launched into the market, and generic versions of the medication are expected to be produced by local drug producers in some countries. Therefore, new methods are required to control sofosbuvir in pharmaceuticals. In the present study, a new method based on reversed phase(rp)-ultra-high performance liquid chromatography(UHPLC) coupled to diode array detection(DAD) and mass spectrometry(Ms) was developed to facilitate the qualitative and quantitative analysis of sofosbuvir in film coated tablets. A wavelength of 260 nm was selected to perform a cost-effective quantification and the method showed adequate linearity,with an R^2 value of 0.9998, and acceptable values of accuracy(75%–102%) and precision(residual standard deviation s were 0.07 μg/mL and 0.36 μg/mL, ***, the use of high-resolution Ms enabled us to ensure the specificity, check impurities and better sensitivity. Therefore, this methodology promises to be suitable not only for the routine analysis of sofosbuvir in pharmaceutical dosage forms, but also for potential degradants.

In this paper,generalized wavelet-neural network(WNN)based models were developed for estimating reference evapotranspiration(ETo)corresponding to Hargreaves(HG)method for different agro-ecological regions(AERs):semi-arid,arid,sub-humid,and humid in *** input and target to the WNN models are climate data(minimum and maximum air temperature)and ETo(estimated from FAO-56 Penman Monteith method),*** developed WNN models were compared with the various generalized conventional models such as artificial neural networks(ANN),linear regression(LR),wavelet regression(WR),and HG method to test the best performed *** performance indices used for the comparison include root mean squared error(RMsE),Nash-sutcliffe efficiency(NsE),the ratio of average output to the average target ETo values(Rratio),and relative percentage(rp).The WNN and ANN models were performed better as compared to LR,WR and HG ***,the best performed WNN and ANN models were tested on locations,which were not included in training to test their generalizing *** is concluded that the WNN and ANN models were shown good generalizing capability for the tested locations as compared to HG method.

In this study, we developed and validated a fast, specific, sensitive, precise and stability-indicating high performance liquid chromatography（HPLC） method to determine the drug apocynin in bovine serum albumin（BsA） nanoparticles. Chromatographic analyses were performed on an rp C18 column and using a photodiode array detector at a wavelength of 276 nm. Mobile phase consisted of a mixture of acetonitrile and 1% acetic acid（60：40, v/v）, and it was eluted isocratically at a flow rate of 0.8 mL/min. The retention time of apocynin chromatographic peak was 1.65 min. The method was linear, precise, accurate and specific in the range of 5–100 μg/mL. The intra-and inter-day precisions presented relative standard deviation（RsD） values lower than 2%. The method was robust regarding changes in mobile phase proportion, but not for flow rate. Limits of detection and quantitation were 78 ng/mL and 238 ng/mL, respectively. Apocynin was exposed to acid and alkali hydrolysis, oxidation and visible light. The drug suffered mild degradation under acid and oxidation conditions and great degradation under alkali conditions. Light exposure did not degrade the drug. The method was successfully applied to determine the encapsulation efficiency of apocynin in BsA nanoparticles.

In angiosperms, low red (R)/far-red (FR) ratio light increases gibberellin (GA) levels. GA signaling in conifer seedlings requires FR to promote shoot elongation and reduce the inhibition of shoot elongation induced by paclobutrazol (PAC), yet the effects of far-red light in inducing shoot elongation in the presence or absence of PAC is poorly understood. In this study, transcriptomic and proteomic analyses was used to examine the molecular mechanism of FR regulation of shoot elongation in Chinese pine (Pinus tabuliformis Carr.) seedlings in the presence of PAC. Four treatments were compared: white light + water (WW), FR + water (RW), white light + PAC (WP), and FR + PAC (rp), and 1436 differentially expressed genes (DEGs) and 450 differentially expressed proteins (DEPs) were identified in RW_WW (RW contrast WW), and 1862 DEGs and 481 DEPs in rp_WP (rp contrast WP). Metabolic and signal transduction pathway analyses of DEGs and DEPs in RW_WW and rp_WP, indicated that the former required more energy than the latter. Moreover, gibberellic acid, auxin, and brassinolide are equally important in RW_WW and rp_WP for shoot elongation, except for the ethylene pathway. Amino acid metabolism and cell wall organization were significantly enriched in RW_WW and rp_WP, respectively. In summary, RW_WW and rp_WP had different effects in secondary metabolism, energy metabolism, amino acid metabolism, cell wall organization, and hormone response. These results provide an important theoretical and reference basis for studying the regulatory effect of low R/FR and PAC in conifer shoot elongation.

To study the influence of a stress-relaxation plate on the remodeling of callus and cortex under the plate Methods The bilateral tibia diaphysis of New Zealand rabbit were osteotomized and fixed with stress-relaxation plate (srp) and rigid plate (rp), respectively Polarized light microscopy and transmission electron microscopy (TEM) were used to study the remodeling of callus and the cortex under the plate from 4 to 24 weeks postoperatively Results Polarized light microscopy: the structural changes of callus and cortex beneath the plate are similar in the srp and rp groups at the early postoperative stage, manifesting an alignment disorder of collagen fibers with a weak birefringence in the callus and absorption cavities in the cortex under the plate After the twelveth postoperative week, the srp group showed callus starting to transform to lamellar bone and absorption cavities in the cortex under the plate becaming smaller By contrast in the rp group the absorption cavities in the callus and cortex under the plate became larger and the whole layer of cortex was cancellated TEM: the active osteoclasts appeared in both srp and rp groups in the period from 4 to 8 weeks postoperatively In the srp group, many functionally active osteoblasts could be seen on the surface of the bone, while in the rp group, the osteoblasts were not very active By 24 weeks postoperatively, the shape of osteocytes were normal but the number of the osteoclasts were small in the srp group In the rp group, the osteoclasts became more active and osteocytic osteolysis was manifested Conclusions Fixation with srp not only enhanced callus remodeling, but also abated the degree of osteoporosis in the cortex under the plate This approach may lead to an improved osteosynthetic apparatus

OBJECTIVE： The stable quality of Chinese herbal medicines is a critical factor for their reliable clinical efficiency. An improved liquid-liquid extraction procedure and a liquid chromatographic method were developed to simultaneously analyze five anthraquinones （aloe-emodin, rhein, emodin, chrysophanol and physcion） in a Chinese traditional hospital preparation, Fuyankang mixture, in order to quantitatively control its quality in a more effective way. METHODs： A more economical and repeatable extraction procedure based on conventional liquid-liquid extraction technique was developed and used to extract five marker components in Fuyankang mixture. These anthraquinones were separated in less than 20 min on a C18 column with methanol and 0.1% phosphoric acid （88：12, v/v） as mobile phase. The method was validated for specificity, precision, spiked recovery and stability. REsULTs： Compared to conventional liquid-liquid extraction, the improved liquid-liquid extraction was found to be more effective for simultaneous extraction of anthraquinones from an aqueous Chinese herbal preparation, especially for hydrophobic compounds. The improved extraction method was successfully applied to determine the content of five marker components in Fuyankang mixture by the means of reverse phase high-performance liquid chromatography. CONCLUsION： The improved extraction procedure may be suitable for routine quality control of Fuyankanq mixture and other traditional preparations at city-level hospitals in China.
