Cloning and functional characterization of two cDNAs encoding NADPH-dependent 3-ketoacyl-CoA reductased from developing cotton fibers

Genes encoding enzymes involved in biosynthesis of very long chain fatty acids were significantly up-regulatedduring early cotton fiber development. Two cDNAs, GhKCR1 and GhKCR2 encoding putative cotton 3-ketoacyl-CoAreductases that catalyze the second step in fatty acid elongation, were isolated from developing cotton fibers. GhKCR1and 2 contain open reading frames of 963 bp and 924 bp encoding proteins of 320 and 307 amino acid residues,respectively. Quantatitive RT-PCR analysis showed that both these genes were highly preferentially expressed duringthe cotton fiber elongation period with much lower levels recovered from roots, stems and leaves. GhKCR1 and 2showed 30%-32% identity to Saccharomyces cerevisiae Ybr159p at the deduced amino acid level. These cotton cDNAswere cloned and expressed in yeast haploid ybr159w? mutant that was deficient in 3-ketoacyl-CoA reductase ***-type growth rate was restored in ybr159w? cells that expressed either GhKCR1 or 2. Further analysis showed thatGhKCR1 and 2 were co-sedimented within the membranous pellet fraction after high-speed centrifugation, similar to theyeast endoplasmic reticulum marker ScKar2p. Both GhKCR(s) showed NADPH-dependent 3-ketoacyl-CoA reductaseactivity in an in vitro assay system using palmitoyl-CoA and malonyl-CoA as substrates. Our results suggest thatGhKCR1 and 2 are functional orthologues of ScYbr159p.

The analysis of numerous experimental equations published in the literature reveals awide scatter in the predictions for the static recrystallization kinetics of steels. Thepowers of the deformation variables, strain and strain rate, similarly as the powerof the grain size vary in these equations. These differences are highlighted and thetypical values are compared between torsion and compression tests. Potential errorsin physical simulation testing are discussed.

In this paper we shall study the solvability of discontinuous functional equations, and apply the so-obtained results to discontinuous implicit initial value problems in ordered Banach spaces. The proofs are based on fixed point results in ordered spaces proved recently by the author. A concrete example is solved to demonstrate the obtained results.

Similarities and differences between the 2 main kinds of compound eye （apposition and superposition） are briefly explained before several promising topics for research on compound eyes are being introduced. Research on the embryology and molecular control of the development of the insect clear-zone eye with superposition optics is one of the suggestions, because almost all of the developmental work on insect eyes in the past has focused on eyes with apposition optics. Age- and habitat-related ultrastructural studies of the retinal organization are another suggestion and the deer cad Lipoptena cervi, which has an aerial phase during which it is winged followed by a several months long parasitic phase during which it is wingless, is mentioned as a candidate species. Sexual dimorphism expressing itself in many species as a difference in eye structure and function provides another promising field for compound eye researchers and so is a focus on compound eye miniaturization in very small insects, especially those that are aquatic and belong to species, in which clear-zone eyes are diagnostic or are tiny insects that are not aquatic, but belong to taxa like the Diptera for instance, in which open rather than closed rhabdoms are the rule. Structures like interommatidial hairs and glands as well as corneal microridges are yet another field that could yield interesting results and in the past has received insufficient consideration. finally, the dearth of information on distance vision and depth perception is mentioned and a plea is made to examine the photic environment inside the foam shelters of spittle bugs, chrysales of pupae and other structures shielding insects and crustaceans.

Catalysis over metal nanoparticles is essential for the growth of carbon nanotubes and all the properties of the resulting nanotube,such as diameter and chirality,are affected by the metal ***,it is very important to understand the carbon chemistry taking place on nanometer size metal *** we present the fi rst ab initio computational study of chemical reactions on a nanosized iron *** clusters have reaction sites,such as edges and vertexes between the facets,Which have not been studied *** principles electronic structure calculations,fully incorporating the effects of spin polarization and non-collinear magnetic moments,have been used to investigate CO disproportionation on an isolated Fe55 *** CO dissociation,O atoms remain on the surface while C atoms move into the cluster,presumably as the initial step toward carbide *** we show that the lowest CO dissociation barrier found on the cluster(0.77 eV)is lower than on most previously studied Fe *** dissociation occurs on a vertex between the *** possible paths for CO2 formation were identifi *** calculated lowest reaction barrier is 1.08 eV,which is comparable to the barrier of 0.65 eV obtained by experiment.
