Retinitis pigmentosa is a group of inherited diseases that lead to retinal degeneration and photoreceptor cell ***,there is no effective treatment for retinitis pigmentosa caused by PDE6B ***-associated virus(AAV)-mediated gene therapy is a promising strategy for treating retinitis *** aim of this study was to explore the molecular mechanisms by which AAV2-PDE6B rescues retinal *** do this,we injected retinal degeneration 10(rd10)mice subretinally with AAV2-PDE6B and assessed the therapeutic effects on retinal function and structure using dark-and light-adapted electroretinogram,optical coherence tomography,and ***-independent acquisition-mass spectrometry-based proteomic analysis was conducted to investigate protein expression levels and pathway enrichment,and the results from this analysis were verified by real-time polymerase chain reaction and western ***2-PDE6B injection significantly upregulated PDE6βexpression,preserved electroretinogram responses,and preserved outer nuclear layer thickness in rd10 *** expressed proteins between wild-type and rd10 mice were closely related to visual perception,and treating rd10 mice with AAV2-PDE6B restored differentially expressed protein expression to levels similar to those seen in wild-type *** Encyclopedia of Genes and Genome analysis showed that the differentially expressed proteins whose expression was most significantly altered by AAV2-PDE6B injection were enriched in phototransduction ***,the phototransductionrelated proteins Pde6α,Rom1,Rho,Aldh1a1,and Rbp1 exhibited opposite expression patterns in rd10 mice with or without AAV2-PDE6B ***,Bax/Bcl-2,p-ERK/ERK,and p-c-Fos/c-Fos expression levels decreased in rd10 mice following AAV2-PDE6B *** data suggest that AAV2-PDE6B-mediated gene therapy promotes phototransduction and inhibits apoptosis by inhibiting the ERK signaling pathway and upr

Cells are the fundamental units of biological systems and exhibit unique development trajectories and molecular features. Our exploration of how the genomes orchestrate the formation and maintenance of each cell, and control the cellular phenotypes of various organismsis, is both captivating and intricate. since the inception of the first single-cell RNA technology, technologies related to single-cell sequencing have experienced rapid advancements in recent years. These technologies have expanded horizontally to include single-cell genome, epigenome,proteome, and metabolome, while vertically, they have progressed to integrate multiple omics data and incorporate additional information such as spatial sc RNA-seq and CRIsPR screening. single-cell omics represent a groundbreaking advancement in the biomedical field,offering profound insights into the understanding of complex diseases, including cancers. Here, we comprehensively summarize recent advances in single-cell omics technologies, with a specific focus on the methodology section. This overview aims to guide researchers in selecting appropriate methods for single-cell sequencing and related data analysis.

A novel mutation of CYP4V2 gene associated with Bietti crystalline dystrophy complicated by choroidal neovascularization

AIM: To investigate the clinical characteristics and genetic features of a Bietti crystalline dystrophy(BCD) proband in a Chinese ***: A Chinese female diagnosed with BCD complicated by bilateral choroidal neovascularization(CNV) and her parents underwent complete ophthalmic examinations, including fundus autofluorescence(AF), fundus photography(FP), fundus fluorescein angiography(FFA), visual field testing, full-field electroretinography(ERG), optical coherence tomography(OCT) and optical coherence tomography angiography(OCTA). The sequencing of the CYP4 V2 gene was performed to the whole ***: Bilateral tiny glittering crystal-like deposits and differing extent of atrophy of the retinal pigment epithelium(RPE) were found in the posterior pole of her fundus. The diffuse hypo-fluorescence shown on AF images and window defects shown on FFA both indicated the atrophy of the RPE and choriocapillaris. OCT showed the thinning of the RPE and choriocapillaris layer, ellipsoid zone(EZ) band defect and CNV in both eyes. OCTA images proofed bilateral type 2 CNV. The visual field test showed central and paracentral scotoma. ERG showed a slightly decreased b-wave in scotopic ERG. Gene sequencing identified three mutations of the CYP4 V2 gene, c.802_807 del, c.810 del T, and c.1388 G>A. The mutation c.1388 G>A was a novel substitution ***: The novel mutation c.1388 G>A may be a possible cause that could induce the clinical phenotype of BCD.

Weighted Zernike defocus coefficient of treatment zone is a meaningful indicator for myopia control efficacy of Ortho‑K lenses

Background:The goal of this study was to reproduce a three-dimensional representation of corneal defocus characteristics after orthokeratology(Ortho-K)treatment via an indicator defined as the weighted Zernike defocus coefficient of the treatment zone(Cweighted defocus).This could be used to predict the effectiveness of Ortho-K treatment quantitatively in a timely manner after the one-month ***:seventy myopic children with axial length(AL)elongation after Ortho-K treatment(group A)and 63 myopic children with AL shortening after Ortho-K treatment(group B)were included in this one-year retrospective *** proposed indicator was calculated by a customized MATLAB *** binomial logistic regression and multivariate linear regression analyses were used to explore the association between AL change and the C_(weighted defocus),age,sex,and other ocular biometric ***:The 12-month AL change,age,pupil diameter,and vertical decentration of the Ortho-K lens were significantly different between the two *** logistic regression analysis showed that a larger Cweighted defocus(≥0.35 D/mm2)(OR:0.224;95%CI:0.078–0.646;P=0.006)was correlated with the emergence of AL shortening after orthokeratology treatment.A multivariate linear regression model showed that a greater Cweighted defocus was associated with slower 12-month AL elongation(β=−0.51,P=0.001).Conclusions:The Cweighted defocus is an effective predictive indicator of myopia control,and a larger Cweighted defocus may lead to slower elongation of *** meaningful indicator may help in the evaluation and adjustment of Ortho-K lens parameters in a timely manner and minimize the cost of clinical trial and error.

Comparison of 24×20 mm^(2) swept-source OCTA and fluorescein angiography for the evaluation of lesions in diabetic retinopathy

AIM: To compare ultra-widefield(24×20 mm^(2)) sweptsource optical coherence tomography angiography(ssOCTA) and fluorescein angiography(FA) in the evaluation of diabetic retinopathy(DR) lesions. METHODs: Forty-six eyes of 23 patients with treatmentna?ve DR were included at peking university people’s hospital from september 1, 2021, until December 31, 2021, as well as 23 age and gender matched healthy controls. Quantitative assessments of DR lesions on FA and ss-OCTA(superficial capillary plexus, sCP, 24×20 mm^(2)) were ***: Area of fovea avascular zone(FAZ) was larger in DR cases than controls(0.34±0.069 mm^(2) vs 0.287±0.108 mm^(2), P=0.006). In DR eyes, the mean FAZ area was 0.34±0.069 and 0.334±0.087 mm^(2) on ss-OCTA and FA, respectively(P=0.428), while the median FAZ perimeter was 2.382(IQR, 2.201-2.59) and 2.333(IQR, 2.138-2.6) mm on ss-OCTA and FA images(P=0.733). There was no significant difference in the size of the non-perfusion area(NPA) between the images on ss-OCTA and FA(12.389, IQR 4.96-28.3 and 11.125, IQR 5-28.31 mm^(2), P=0.197). The median total microaneurysm(MA) count was 35(IQR, 19-46) and 73(IQR, 43-93) on ss-OCTA and FA(Pspectively. No significant difference in intra-retinal microvascular abnormality(IRMA) and neovascularization (NV) count was found between the two techniques. The intraclass coefficient(ICCs) of all the parameters above indicated stable ***: Ultra-widefield ss-OCTA represents a reliable, noninvasive, and quantitative imaging technique in the assessment of microvasculature in DR, which offers a potential substitute for FA in DR evaluation.

AIM:To evaluate the efficacy of ruthenium-106 plaque brachytherapy for the treatment of diffuse choroidal hemangioma(DCH)in sturge-Weber syndrome(sWs).METHODs:A total of 8 patients with DCH in sWs managed with plaque brachytherapy were retrospectively *** were treated with ruthenium-106 plaque therapy(median apex dose:83 Gy)at the thickest tumor *** follow-up,we recorded the tumor thickness,the best-corrected visual acuity(BCVA),subretinal fluid(sRF)status,and complications following ***:At a median follow-up of 43 mo,tumor regression was observed in all cases,with a complete resolution of sRF and reduction in *** radiation complications were recorded during the follow up ***:Ruthenium-106 plaque therapy to the thickest portion of the tumor seems to be a useful treatment in patients with DCH in sWs.

The process of retinal vascularization in retinopathy of prematurity after ranibizumab treatment in China

AIM: To explore the process of retinal vascularization and risk factors for retinopathy of prematurity(ROP) treated with intravitreal ranibizumab(IVR) as ***: Infants with type 1 ROP who received IVR as primary treatment from August 2014 to October 2016 at peking university people’s hospital’s Ophthalmology Department were included in the study. All eyes received 0.25 mg ranibizumab at initial treatment. Retinal vascularization was evaluated clinically. Potential risk factors were also recorded and ***: Retinal vascularization was completed in 126 eyes(62.7%), and retinal vascularization terminated in zone II and zone III with 16 eyes(7.9%) and 44 eyes(21.9%), respectively, after more than 1-year follow-up. In multivariate regression analysis, lower birth weight(BW), severity of ROP and repeated injections were found to be risk factors for peripheral avascular area(PsION: In our retrospective study, 29.8% of the ROP eyes treated with ranibizumab have peripheral avascular area at the last follow-up. Lighter BW and the severity of ROP are risk factors. Furthermore, repeated injections also increase the risk of retinal peripheral avascular area remaining in ROP patients.

AIM: To evaluate the outcomes of intravitreal antivascular endothelial growth factor(anti-VEGF) agents for patients with retinal vein occlusion(RVO) related-macular edema(ME) in ***: A retrospective, observational, single-center study. The demographic and clinical data of 90 RVO Tibetan patients(93 eyes) treated with either ranibizumab or conbercept in Tibet Autonomous Region people’s hospital from Jan 2018 to December 2019 were ***: The mean patient age was 56.8±10.6y, 45(50%) of them were female. The mean living altitude was 3867.8±567.9 m. At the last visit, the best-corrected visual acuity(BCVA) significantly increased(52.2±21.8 letters) in comparison with the baseline(38.2±24.1 letters, Pss(CRT) significantly reduced(245.5±147.6 μm) in comparison with the baseline(504.1±165.2 μm, Peyes gained≥15 letters, 60.2% of the eyes gained ≥10 letters, and 78.5% of the eyes gained ≥5 letters. No vision loss was noted in 92.5% of the eyes, 4 eyes lost more than 10 letters during follow-up period. The mean number of injections was 2.4±1.8. No severe ocular or systemic adverse events related to either the drug or injection were ***: Anti-VEGF therapy is effective and safe in Tibetan patients for the treatment of RVO related ME.
