Background: Post-craniotomy pain has been reported to be moderate to severe. Management of post-craniotomy pain is often inadequate, yet limited by the side effects of opioids. We aim to find out the efficacy of oral oxycodone as compared to oral codeine for the treatment of post-craniotomy pain in our institution. Methods: A randomized, double-blinded controlled trial was used to evaluate the efficacy of oral oxycodone versus oral codeine. 40 patients were randomized to the control group of codeine (n = 20) or the experimental group receiving oxycodone (n = 20) in addition to regular oral paracetamol for both groups of patients. Results: There was no difference in the visual analogue scale scores at 24 hours (2.78 versus 1.85, p = 0.11) or side effects in the oxycodone group compared with the codeine group. Conclusions: Oral oxycodone had similar efficacy as oral codeine in the management of post-craniotomy pain.

The rapidly growing field of functional, molecular and structural bio-imaging is providing an extraordinary new opportunity to overcome the limits of invasive liver biopsy and introduce a "digital biopsy" for in vivo study of liver pathophysiology. To foster the application of bio-imaging in clinical and translational research, there is a need to standardize the methods of both acquisition and the storage of the bio-images of the liver. It can be hoped that the combination of digital, liquid and histologic liver biopsies will provide an innovative synergistic tri-dimensional approach to identifying new aetiologies, diagnostic and prognostic biomarkers and therapeutic targets for the optimization of personalized therapy of liver diseases and liver cancer. A group of experts of different disciplines(Special Interest Group for Personalized Hepatology of the Italian Association for the Study of the Liver, Institute for Biostructures and Bio-imaging of the National Research Council and Bio-banking and Biomolecular Resources Research Infrastructure) discussed criteria, methods and guidelines for facilitating the requisite application of data collection. This manuscript provides a multi-Author review of the issue with special focus on fatty liver.

Social environment - demographics Italian population is quickly becoming older due to a very low birth rate and thanks to the longest life expectancy in Europe (83 years on avarage). Nevertheless, focusing on the quality of that long expected life span, we come at the bottom of this European league: 65 years old Italians can expect on average to live only another 7.5 years of life in good health. There are over 20 million Italians with a sedentary lifestyle, and more than 50% of those over 65 who do not practice any kind of physical activity [2].

AIM:To investigate the inhibitory effect of arsenic trioxide on human glioma cell line U251 growth,change of microRNA125 b and it’s target gene ***:MTT method was used to observe the growth inhibition *** cycle and apoptosis were measured by flow cytometry in U251 cells treated with different doses of As *** invasive activity of the cells was evaluated by transwell chamber *** migration of the cells was determined by scratch *** real-time fluorescent quantitative PCR(qRT-PCR) was used to detect miR-125 b,BAK1,apoptosis associated protein Bcl-2,Bax and cell cycle associated protein CKD6 expression with As *** protein expression of Bcl-2,Bax and caspase-3 with As O were detected by western ***:As O inhibited the growth of U251 cells dramatically and reduced cells of migration and *** were obvious dosage-effect(P < 0.05).As O inhibited miR-125 b expression and increased BAK1 protein of miR-125 b target gene expression levels,decreased Bcl-2/Bax ratio and CDK6 protein expression in U251 cells(P < 0.05).The cell cycle was arrested in G1/S *** occurred in U251 cells treated with As O by Annexin V & Dead Cell Kit ***:As O inhibits the growth of U251 cells in vitro dramatically and induces *** mechanism is probably associated with the decrease of miR-125 b expression and CDK6,increase in BAK1 protein expression and cell cycle arrest.

Delayed bone defect repairs lead to severe health and socioeconomic impacts on patients. Hence, there are increasing demands for medical interventions to promote bone defect healing. Recombinant proteins such as BMP-2 have been recognized as one of the powerful osteogenic substances that promote mesenchymal stem cells (MSCs) to osteoblast differentiation and are widely applied clinically for bone defect repairs. However, recent reports show that BMP-2 treatment has been associated with clinical adverse side effects such as ectopic bone formation, osteolysis and stimulation of inflammation. Here, we have identified one new osteogenic protein, named ‘HKUOT-S2’ protein, from Dioscorea opposita Thunb. Using the bone defect model, we have shown that the HKUOT-S2 protein can accelerate bone defect repair by activating the mTOR signaling axis of MSCs-derived osteoblasts and increasing osteoblastic biomineralization. The HKUOT-S2 protein can also modulate the transcriptomic changes of macrophages, stem cells, and osteoblasts, thereby enhancing the crosstalk between the polarized macrophages and MSCs-osteoblast differentiation to facilitate osteogenesis. Furthermore, this protein had no toxic effects in vivo. We have also identified HKUOT-S2 peptide sequence TKSSLPGQTK as a functional osteogenic unit that can promote osteoblast differentiation in vitro. The HKUOT-S2 protein with robust osteogenic activity could be a potential alternative osteoanabolic agent for promoting osteogenesis and bone defect repairs. We believe that the HKUOT-S2 protein may potentially be applied clinically as a new class of osteogenic agent for bone defect healing.

The selection of process parameters for obtaining optimal tensile properties in the pulsed current gas tungsten arc welding is presented. The tensile properties include ultimate tensile strength, yield strength and notch tensile strength. All these characteristics are considered together in the selection of process parameters by modified taguchi method to analyse the effect of each welding process parameter on tensile properties. experimental results are furnished to illustrate the approach.

Erectile dysfunction （ED） commonly results from endothelial dysfunction of the systemic vasculature. Although phosphodiesterase type 5 （PDE-5） inhibitors are effective at treating most cases of ED, they must be taken routinely and are ineffectual for a meaningful number of men. In recent years gene and stem cell-based therapies targeted at the penile endothelium have been gaining momentum in preclinical studies. These early studies reveal that gene and stem cell-based therapies may be both enduring and efficacious, and may eventually lead to a cure for ED. The following review will highlight our current understanding of endothelial-specific gene and stem cell-based therapies performed to date in a number of experimental animal models.

Mechanical strength and its long-term stability of bioceramic scaffolds is still a problem to treat the osteonecrosis of the femoral *** the long-term stability of diopside(DIO)ceramic but poor mechanical strength,we developed the DIO-based porous bioceramic composites via dilute magnesium substituted wollastonite reinforcing and three-dimensional(3D)*** experimental results showed that the secondary phase(i.e.10%magnesium substituting calcium silicate;CSM10)could readily improve the sintering property of the bioceramic composites(DIO/CSM10-x,x=0-30)with increasing the CSM10 content from 0%to 30%,and the presence of the CSM10 also improved the biomimetic apatite mineralization ability in the pore struts of the ***,the flexible strength(12.5 -30 MPa)and compressive strength(14-37 MPa)of the 3D printed porous bioceramics remarkably increased with increasing CSM10 content,and the compressive strength of DIO/CSM10-30 showed a limited decay(from 37 MPa to 29 MPa)in the Tris buffer solution for a long time stage(8 weeks).These findings suggest that the new CSM10-reinforced diopside porous constructs possess excellent mechanical properties and can potentially be used to the clinic,especially for the treatment of osteonecrosis of the femoral head work as a bioceramic rod.

Multiple sclerosis(MS)is a leading cause of chronic neurological disability in young to middle-aged adults,affecting~2.5 million people ***,most therapeutics for MS are systemic immunosuppressive or immunomodulatory drugs,but these drugs are unable to halt or reverse the disease and have the potential to cause serious adverse ***,there is an urgent need for the development of next-generation treatments that,alone or in combination,stop the undesired autoimmune response and contribute to the restoration of *** review analyzes current MS treatments as well as different cell-based therapies that have been proposed to restore homeostasis in MS patients(tolerogenic dendritic cells,regulatory T cells,mesenchymal stem cells,and vaccination with T cells).Data collected from preclinical studies performed in the experimental autoimmune encephalomyelitis(EAE)model of MS in animals,in vitro cultures of cells from MS patients and the initial results of phase I/II clinical trials are analyzed to better understand which parameters are relevant for obtaining an efficient cell-based therapy for MS.
