Objective:Benzylisoquinoline alkaloids(BIAs) have pharmacological functions and clinical *** are mainly distributed in plant species across the order Ranunculales and the genus Phellodendron from *** BIA biosynthesis has been intensively investigated in Ranunculales ***,the accumulation mechanism of BIAs in Phellodendron is largely *** aim of this study is to unravel the biosynthetic pathways of BIAs in Phellodendron ***:The transcriptome and metabolome data from 18 different tissues of *** were meticulously sequenced and subsequently subjected to a thorough *** gene co-expression network analysis(WGCNA),a powerful systems biology approach that facilitates the construction and subsequent analysis of co-expression networks,was utilized to identify candidate genes involved in BIAs *** this,recombinant plasmids containing candidate genes were expressed in Escherichia coli,a widely used prokaryotic expression *** purpose of this genetic engineering endeavor was to express the candidate genes within the bacteria,thereby enabling the assessment of the resultant enzyme ***:The synonymous substitutions per synonymous site for paralogs indicated that at least one whole genome duplication event has *** potential BIA biosynthetic pathway of *** was proposed,and two PR10/Bet v1 members,14 CYP450s,and 33 methyltransferases were selected as related to BIA *** PR10/Bet v1 was identified as norcoclaurine synthase,which could catalyze dopamine and 4-hydroxyphenylacetaldehyde into(S)-***:Our studies provide important insights into the biosynthesis and evolution of BIAs in nonRanunculales species.

Research Progress of Raman Spectroscopy and Imaging Techniques for the pharmaceutical Analysis

Monitoring critical quality attributes in pharmaceutical manufacturing ensures that the final product meets specified requirements. Process analysis allows for appropriate modifications and customization of pharmaceutical design from formulation to production. The laborious sample presentation and throughput have stimulated the development of in-process spectroscopy methods for pharmaceutical analysis. Raman spectroscopy has demonstrated the unique advantages of rapid and nondestructive analysis, good interpretability, and effective moisture interference avoidance. The Raman imaging technique can obtain spatial and chemical information that is useful for characterizing drug quality. This article describes the applications of Raman spectroscopy and imaging techniques in pharmaceutical analysis. It covers the monitoring of crystal forms and particle size distribution, quantification of active pharmaceutical ingredients, evaluation of contents uniformity, prediction of dissolution profiles in solid preparations, identification of counterfeit drugs, and applications in reverse engineering and process analytical technology. Moreover, the challenges and prospects are proposed to facilitate the continued development of Raman spectroscopy and imaging techniques, providing necessary references for further research in this field.

Extensive use of avermectin(AVM)in agriculture can seriously contaminate fish in *** acid(FA)is a strong antioxidant sourced from plants and is widely applied in food and medicine,but its protective function on the immunological damage caused by AVM is still *** this study,carp were chronically exposed to(1/1096 hr LC_(50))AVM for 30 day(s),with a dietary supplement of FA(400 mg/kg)to assess its effects on carp *** experimental groups(n=10)included:control,AVM-exposed(2.404μg/L),FA+AVM co-treated,and FA ***-term AVM exposure altered spleen tissue,reducing serum complement C3(p<0.01)and immunoglobulin M levels(p<0.001),and increasing proinflammatory tnf-α(p<0.001),il-6(p<0.001),il-1β(p<0.001),and inos mRNA levels,whilst down-regulating the anti-inflammatory tgf-β(p<0.05).Additionally,it disrupted the balance of oxidative stress indicators such asMDA(p<0.001),T-AOC,GSH,and CAT,leading to spleen tissue apoptosis(42.4%).However,the addition of FA reversed these conditions,elevated the anti-inflammatory factor,and improved spleen immune function following chronic exposure to AVMin ***,the ability to restore oxidative homeostasis in carp by adjusting the Nrf2/NQO-1 axis protected the health of spleen *** discovery also supports the development of fish feed for aquaculture.

A visible light-promoted fast photochemical Wolff rearrangement was developed toward synthesis of α-substituted amides in continuous flow with the use of a photochemical oscillatory flow reactor(POFR).The control experiment indicates that a fast process of the Wolff rearrangement(<40 s) is involved. Notably, this protocol does not require excess use of any reactants, and the resulting α-substituted amides could be isolated by recrystallization in good to excellent yields.

Curvularin derivatives from hydrothermal vent sediment fungus Penicillium ***-50 guided by molecular networking and their antiinflammatory activity

Guided by molecular networking, nine novel curvularin derivatives（1-9） and 16 known analogs（10-25） were isolated from the hydrothermal vent sediment fungus Penicillium sp. HL-50. Notably, compounds 5-7 represented a hybrid of curvularin and purine. The structures and absolute configurations of compounds 1-9 were elucidated via nuclear magnetic resonance（NMR） spectroscopy, X-ray diffraction, electronic circular dichroism（ECD） calculations,13C NMR calculation, modified Mosher's method, and chemical derivatization. Investigation of anti-inflammatory activities revealed that compounds 7-9, 11, 12, 14, 15, and 18 exhibited significant suppressive effects against lipopolysaccharide（LPS）-induced nitric oxide（NO）production in murine macrophage RAW264.7 cells, with IC50values ranging from 0.44 to 4.40μmol·L-1. Furthermore, these bioactive compounds were found to suppress the expression of inflammation-related proteins, including inducible NO synthase（i NOS）, cyclooxygenase-2（COX-2）, NLR family pyrin domain-containing protein 3（NLRP3）, and nuclear factor kappa-B（NF-κB）. Additional studies demonstrated that the novel compound 7 possessed potent antiinflammatory activity by inhibiting the transcription of inflammation-related genes, downregulating the expression of inflammation-related proteins, and inhibiting the release of inflammatory cytokines, indicating its potential application in the treatment of inflammatory diseases.

Enhanced Performance of Co-doped Antimony Electrode for pH Measurement in Marine Environments

The antimony（Sb） electrode, despite its prevalent use for pH detection in complex environments, suffers from severe potential drift due to poor corrosion resistance, thereby impeding long-term or online applications. Intermetallic compounds show excellent resistance to corrosion and oxidation, presenting a promising avenue for improving the stability of the Sb electrode. Therefore, we fabricate an intermetallic compound-doped Sb electrode by adding various cobalt（Co） contents to improve the stability of the Sb electrode without compromising the inherent pH response. It's observed that the alloy electrode exhibits a significant reduction in surface roughness, corrosion product Sb2O3content, cracks, and Sb exfoliation compared to the pure Sb electrode after the electrochemical tests, and the effect becomes more pronounced as the Co content increases. The electrochemical tests further reveal that the alloy electrode exhibits better corrosion resistance than the pure Sb in various solutions, with the alloy electrode containing 80 at% Sb showing the highest corrosion resistance. Moreover,the alloy electrode containing 80 at% Sb also shows excellent sensitivity and anti-interference ability, high accuracy and reproducibility, low hysteresis, and better long-term stability, especially in seawater. These findings suggest that inexpensive Co doping significantly enhances corrosion resistance, thereby reducing electrode surface alterations and eventually improving the potential stability of pure Sb electrodes without affecting the inherent pH response. Furthermore, the obtained results provide an experimental basis for the long-term operation of the alloy pH electrode in complex environments such as seawater without frequent maintenance.

Astragalus membranaceus possesses the function of enhancing immunity,protecting the liver,diuretic,anti-aging,anti-stress,anti-hypertensive,and more extensive antibacterial ***,one kind of the major active ingredients of ***,are considered to be responsible for their versatile ***,a systematic summary of research progress and prospects of polysaccharides from *** polysaccharides(AMPs) is necessary to facilitate their further study and *** this review,the optimal extraction methods,structural features,biological activities,and applications of AMPs were *** structure-activity relationships are also analyzed and ***,ultrasonic,microwave,enzyme-assisted,ultra-high pressure,and combined methods have been used to extract *** them,solvent extraction is the most commonly used method because it is simple and easy to operate,but the efficiency needs to be improved *** ultra-high pressure method is the most efficient but has a low economic *** exhibited various bioactivities,including immunomodulation,antitumor,and *** structure-activity relationships revealed that different structure configurations,chain conformations,and physical properties would have different ***,the new method for purification of certain polysaccharides,detailed structure-activity relationships(SAR),mechanisms of bioactivities,and quality control of AMPs need to be extensively investigated.
