Urinary tract infection (UTI) is a frequent pathology among HTLV-I+ individuals being capable of severely compromising the kidneys and bladder. Molecular characteristics of uropathogenic Escherichia coli (UpEC) from HTLV-I+ infected individuals are unknown. UpEC isolates from HTVL-I+ individuals, with and without clinical symptoms of myelopathy, were submitted to genetic typing seeking to infer bacterial diversity and potential virulence. 71 bacterial isolates were characterized according to random amplified polymorphic DNA and phylotypes. phylotyping classified E. coli into four phylogenetic groups: A (18.3%), B1 (16.9%), B2 (39.4%), and D (25.3%) and 8 phylotypes according to the presence of the genetic sequences chuA, yjaA and the DNA fragment TSpE4.C2: ﹣﹣﹣ (5.6%) and ﹣+﹣ (12.6%) in phylogroup A, ﹣﹣+ (7.0%) and ﹣++ (9.8%) in B1, +++ (32.3%) and ++﹣ (7.0%) in B2, +﹣﹣ (15.4%) and +﹣+ (9.8%) in D. The B2 phylogroup was the most prevalent among HTLV﹣ associated myelopathy and asymptomatic individuals. RApD-pCR typing revealed a high degree of bacterial polymorphism indicating a non-clonal origin. Genotypes were not found to be distributed according to clinical status or epidemiological features. Our results lead us to suggest that the neurological impairment in HTLV-I+ individuals can be a risk factor for urinary infections due E. coli which are caused by distinct bacterial lineages.

In this paper we pursue the study of the best approximation operator extended from L~Φ to L~φ, where φ denotes the derivative of the function Φ. We get pointwise convergence for the coefficients of the extended best approximation polynomials for a wide class of function f, closely related to the Calder′on–Zygmund class t_m^p(x) which had been introduced in 1961. We also obtain weak and strong type inequalities for a maximal operator related to the extended best polynomial approximation and a norm convergence result for the coefficients is derived. In most of these results, we have to consider Matuszewska–Orlicz indices for the function φ.

Clostridium difficile is a nosocomial enteric pathogen, considered the main etiological agent of antibiotic-associated diarrhea in hospitals. C. difficile is resistant to several antimicrobial agents used in empiric treatment protocols, which confers selective advantages to this species as compared with other members of the intestinal microbiota. The aim of this study was to evaluate and identify possible alterations of the intestinal microbiota population resulting from the use of antimicrobials associated with infections by C. difficile (CDI). denaturing Gradient Gel Electrophoresis (DGGE) was used to evaluate fecal samples from two groups: healthy subjects and patients with C. difficile-associated diarrhea (CDAD). A decrease in bacterial diversity was highlighted by the low number of bands in samples from CDAD patients, compared with healthy subjects. This may indicate that antibiotic treatment would affect bacterial diversity, leading to a significant difference between the intestinal microbiota of these two groups, but further studies are still needed. Firmicutes and Verrucomicrobia phyla were detected mainly in healthy individuals, and these could be related to protection factors against the CDI. Klebsiella variicola/K. pneumoniae were found mostly in samples from CDI patients. This study shows the effects of antimicrobials and the CDI itself on human intestinal microbiota.

The sense of smell is important for human quality of life. This sophisticated sensorial system relies on the detection of odorant molecules that engage receptors expressed in the cilia of dedicated neurons that constitute the olfactory epithelium（OE）. Importantly, the OE is a highly active site of adult neurogenesis where short-lived neurons are efficiently replenished, even after massive neuronal cell loss. It is suggested that the degree of olfactory function recovery after OE injury may depend on the nature of the lesion（traumatic, chemical, infectious or inflammatory）, as well on the velocity of cellular regeneration. Topical steroidal anti-inflammatory drugs, such as glucocorticoids, are routinely prescribed for treating upper airway inflammatory conditions, such as chronic rhinosinusitis. While the therapeutic strategy aims to minimize the inflammatory damage and dysfunction to nasal air conduction, new evidences raise concerns if such drugs may impair neuronal regeneration in the OE. In consequence, new directions are necessary in terms of drug development or prescription, in order to preserve olfactory function through lifelong repeated episodes of chronic inflammation in the upper respiratory tract. Here we discuss mechanisms involved in glucocorticoid deleterious effects to OE regeneration and possible therapeutic alternatives considering relevant side effects.

The strawberry tree （Arbutus unedo L.） is an underutilized, drought tolerant, fire resistant species with a south western distribution in Europe, and with ecological and putative socio-economical impact in portugal and Mediterranean countries. Our aim was to develop an appropriate set of molecular markers to enable genetic diversity to be assessed and to fingerprint Arbutus unedo genotypes for breeding and conservation purposes in portugal. Twenty-seven trees from a broad geographic range were screened with 20 random amplified polymorphic DNA （RApD primers） and 11 microsatellite markers （SSR）. The RApDs generated 124 bands, 57.3% of which were polymorphic, with an expected heterozygosity of 27%. We cross-amplified 11 SSR primers developed for Vaccinium spp., and 5 were found to be polymorphic in A. unedo, with 75% of expected heterozygosity, a number of alleles of 11.6, a null allele frequency of 7.6% and a polymorphic information content of 71%. Although the SSRs were more polymorphic and informative than the RApDs, both markers displayed high genetic variability with the gathered data. No geographic pattern was observed in the genetic variation distribution based on both marker systems, and the lack of correlation between genetic and geographical matrices was confirmed by Mantel tests. Likely, no correlation was found between pairwise SSR and RApD band-sharing matrices. These results and their implications on A. unedo breeding and conservation programs are discussed.

Background: Cervical cancer is the main cause of cancer deaths in some developing countries. Age-related cervical cancer incidence has been fundamental for understanding the different stages of carcinogenesis concepts. No Brazilian study explored the environmental risk factors involved on cervical cancer, according to age groups taking 70’s sexual revolution in Brazil as reference. Aim: To determine the prevalence of epidemiological and clinical aspects related to cervical cancer development in a sample of three age groups of Brazilian women based on 70’s sexual revolution. Methods: A cross-section study was proceeded in a hospital-based cohort of women with altered pap-test refereed to the National Cancer Institute for Colposcopy and treatment from October 2004 to May 2006. Two register nurses interviewed all patients ascertaining risk factors and clinical characteristics. Biopsy, partial ablation were used for CIN-1, and conization was the treatment for CIN-2/3. Results: From 318 women included in the study, 42.8% were 18 - 30 years old (born after 1975), 43.4% were 31 - 49 years old (born 1955-1975), and 13.8% were 50 - 68 years old (born 1936-1954). pregnancies (OR = 1.16;CI95%: 1.01 - 1.34) and menarche under 12 years old (OR = 1.95;CI95%: 1.17 - 3.25) were independently associated to CIN2- 3/cancer in 18 - 30 age group;menopause age (OR = 1.21;CI95%: 1.04 - 1.41) and current smoking habit (OR = 1.37;CI95%: 1.10 - 1.70) were associated to CIN2- 3/cancer in 31 - 49 age group;no statistical significance was observed for 50 - 68 age group. Conclusion: Brazilian women present distinct risk for cervical cancer according to the generation they belong, when taking the Brazilian sexual revolution on 70’s as a reference.

Arbutoid mycorrhizae were synthesized between adult se- lected clones of Arbutus unedo L. and pisolithus arhizus. Two micro- propagated clones were tested： ALl, in vitro and C1 （acclimatized plants） in nursery and later in a field trial. In vitro, rooted shoots were trans- ferred to test tubes containing the substrate previously inoculated with mycelium cultured on agar. In the nursery, two inoculation treatments were tested （vegetative inocula or dry sporocarps） and compared to con- trol plants. In the field trial, plants from nursery inoculation treatments were compared and an additional control treatment using seedlings was implemented. plant height was evaluated 4 months later in the nursery and 20 months later in the field trial.

Spinal muscular atrophy(SMA)is caused by dysfunction of the alpha motor neurons of the spinal *** is an autosomal recessive disease associated to the SMN1 gene,located in the subtelomeric region of 5q13.A paralog SMN2 gene is located at the centromeric region of the same chromosome,which apparently originated by an ancestral inverted duplication occurring only in *** exon sequence differs in two nucleotides in exon 7 and exon 8,which leads to an SMN2 transcript that lacks exon 7 and results in a truncated ***(10%)of the SMN2 transcripts avoids the splicing of exon 7 but most of the copies are *** a disease scenario,the more SMN2 copies the higher possibility to restore at least partly the effects of SMN1 *** therapeutic approaches are being developed to increase the expression of *** determine the number of SMN1 and SMN2 copies,the methodology must distinguish accurately between both *** this work,we present the results obtained using multiplex ligation-dependent probe amplification(MLpA)in 60 SMA suspected patients/carriers derived from different regions of *** 32 of these DNA samples we found alterations in *** these,16 presented a heterozygous deletion(carrier status)and 14 an homozygous deletion(patient status)in exon 7 and 8 of *** one case,exon 7 was found homozygously deleted but exon 8 presented a single copy,and in another case,exon 7 was found heterozygously deleted while exon 8 was *** half of the patients(7/15)presented a normal diploid number of SMN2 while the other half(8/15)presented an increased *** this work we showed how a probe-based methodology such as MLpA was able to distinguish between the paralog genes and determine the amount of copies in DNA samples from suspected patients/carriers of SMA.

Autophagy is an essential cellular homeostatic mechanism by which intracellular components are delivered into the lysosomes for degradation and *** has been related with a diversity of pathological or physiological dentary processes such as bone remodeling,skeletal aging,osteoclastogenesis,osteoblastogenesis and different types of oral ***-rich plasma(pRp),isolated from autologous blood,is a plasma preparation containing a higher concentration of platelets which contains numerous different growth factors and cytokines that activate several cellular signaling *** purpose of this study is to investigate the effect of pRp on autophagy stimulation in both osteoblast precursor 3T3-L1 and non-related osteoblastic *** results showed that pRp can increase the number of autophagic structures in 3T3-L1 and HeLa(cervical cancer cells)***,we have determined by Western blot a rise in the lipidated form of the autophagic protein LC3(***3-II)upon pRp *** together,our results suggest that pRp is able to induce a strongly autophagy response in osteoblast precursor and,to a lesser extent,in non-related osteoblastic cells,suggesting that pRp could be a potential therapeutic tool for some autophagy-related diseases associated with bone homeostasis.
