Nitric Oxide and Brassinosteroids Mediated Fungal Endophyte-Induced Volatile Oil Production Through Protein Phosphorylation Pathways in Atractylodes lancea Plantlets

Fungal endophytes have been isolated from almost every plant, infecting their hosts without causing visible disease symptoms, and yet have still proved to be involved in plant secondary metabolites accumulation. To decipher the possible physiological mechanisms of the endophytic fungus-host interaction, the role of protein phosphorylation and the relationship between endophytic fungus-induced kinase activity and nitric oxide （NO） and brassinolide （BL） in endophyte-enhanced volatile oil accumulation in Atractylodes lancea plantlets were investigated using pharmacological and biochemical approaches. Inoculation with the endophytic fungus Gilmaniella sp. ALl2 enhanced the activities of total protein phosphorylation, Ca2^-dependent protein kinase, and volatile oil accumulation in A. lancea plantlets. The upregulation of protein kinase activity could be blocked by the BL inhibitor brassinazole. Furthermore, pretreatments with the NO-specific scavenger cPTIO significantly reduced the increased activities of protein kinases in A. lancea plantlets inoculated with endophytic fungus. Pretreatments with different protein kinase inhibitors also reduced fungus-induced NO production and volatile oil accumulation, but had barely no effect on the BL level. These data suggest that protein phosphorylation is required for endophyte- induced volatile oil production in A. lancea plantlets, and that crosstalk between protein phosphorylation and the NO pathway may occur and act as a downstream signaling event of the BL pathway.

A practical soil management to improve soil quality by applying mineral organic fertilizer

Heavy use of chemical fertilizer causes increasing soil and environmental crisis, and the use of organic fertilizer increases obvious in recent years. In this study,mineral organic fertilizer(MOF) and compound fertilizer(CF) were applied in amaranth culture to explore the effects of these two kinds of fertilizers on soil quality and the potential function for CO_2 fixation. Some soil parameters were tested, e.g. p H value, organic carbon content, microbial biomass, urease activity, and available potassium content. In addition, some parameters of soil infiltration water were also determined, such as p H and HCO_3^- concentration. Experimental results showed that MOF improved soil quality and amaranth biomass and increased possible soil carbon *** the contrary, the utilization of CF worsened soil quality and made the soil acidize. These results suggested that MOF can partially replace CF to improve plant growth, soil quality and possible CO_2 sink.

The severe acute respiratory syndrome coronavirus 2(SARS‐CoV‐2)pandemic resulted in significant societal ***,an in‐depth understanding of SARS‐CoV‐2 virus mutation and its evolution will help determine the direction of the COVID‐19 *** this study,we identified 296,728 de novo mutations in more than 2,800,000 high‐quality SARS‐CoV‐2 *** possible factors affecting the mutation frequency of SARS‐CoV‐2 in human hosts were analyzed,including zinc finger antiviral proteins,sequence context,amino acid change,and translation *** a result,we proposed that when adenine(A)and tyrosine(T)bases are in the context of AM(M stands for adenine or cytosine)or TA motif,A or T base has lower mutation ***,we hypothesized that translation efficiency can affect the mutation frequency of the third position of the codon by the selection,which explains why SARS‐CoV‐2 prefers AT3 codons *** addition,we found a host‐specific asymmetric dinucleotide mutation frequency in the SARS‐CoV‐2 genome,which provides a new basis for determining the origin of the SARS‐CoV‐***,we summarize all possible factors affecting mutation frequency and provide insights into the mutation characteristics and evolutionary trends of SARS‐CoV‐2.

The emerging of high-throughput and high-resolution genomic technologies led to the detection of submicroscopic variants ranging from 1 kb to 3 Mb in the human *** variants include copy number variations(CNVs),inversions,insertions,deletions and other complex rearrangements of DNA *** paper briefly reviews the commonly used technologies to discover both genomic structural variants and their potential ***,we highlight the array-based,PCR-based and sequencing-based assays,including array-based comparative genomic hybridization(aCGH),representational oligonucleotide microarray analysis(ROMA),multiplex amplifiable probe hybridization(MAPH),multiplex ligation-dependent probe amplification(MLPA),paired-end mapping(PEM),and next-generation DNA sequencing ***,we discuss the limitations and challenges of current assays and give advices on how to make the database of genomic variations more reliable.

MicroRNAs as potential biomarkers for gastric cancer

Gastric cancer is the fourth most common cancer in the world and the second leading cause of cancer-related death. More than 80% of diagnoses occur at the middle to late stage of the disease, highlighting an urgent need for novel biomarkers detectable at earlier stages. Recently, aberrantly expressed microRNAs (miRNAs) have received a great deal of attention as potential sensitive and accurate biomarkers for cancer diagnosis and prognosis. This review summarizes the current knowledge about potential miRNA biomarkers for gastric cancer that have been reported in the publicly available literature between 2008 and 2013. Available evidence indicates that aberrantly expressed miRNAs in gastric cancer correlate with tumorigenesis, tumor proliferation, distant metastasis and invasion. Furthermore, tissue and cancer types can be classified using miRNA expression profiles and next-generation sequencing. As miRNAs in plasma/serum are well protected from RNases, they remain stable under harsh conditions. Thus, potential functions of these circulating miRNAs can be deduced and may implicate their diagnostic value in cancer detection. Circulating miRNAs, as well as tissue miRNAs, may allow for the detection of gastric cancer at an early stage, prediction of prognosis, and monitoring of recurrence and/or lymph node metastasis. Taken together, the data suggest that the participation of miRNAs in biomarker development will enhance the sensitivity and specificity of diagnostic and prognostic tests for gastric cancer. Food waste is the most difficult waste to manage because the high treatment costs and the risk of environmental *** straw burning has become an increasingly discussed topic in recent years because the air pollution caused by ***,food waste and wheat straw contain high amounts of nutrient elements,efficient utilization of these waste can improve the environment by lessening airborne pollutant emission and the amount of waste that must be *** is well known that use of low cost and abundant waste materials in microbial fermentations can reduce product *** aimed to use these resources while improving laccase production by the endophytic fungus Phomopsis liquidambari *** cultured *** B3 in medium containing food waste and wheat straw as the main nitrogen and carbon sources,*** optimized the fermentation conditions by response surface methodology(RSM),using a Box-Behnken design for RSM I and a central composite design for RSM Ⅱ.Optimization resulted in an 11.07-fold(1.98-fold RSM I;5.59-fold RSM Ⅱ) increase in laccase yield compared with that before *** model was validated by mathematical evaluations and by comparisons between predicted and experimental *** optimized conditions,53.76%of lignin in wheat straw was *** optimizing fermentation conditions and using multiple bioresources,laccase production by this fungus was *** results provide the foundation for future research and for scaled-up laccase production.
