Alpha-lactalbumin(α-LA)is a major whey protein found in breast milk and plays a crucial role in the growth and development of *** this study,Bacillus subtilis RIK1285 harboring AprE signal peptide(SP)was selected as the original strain for the production ofα-*** was found thatα-LA was identified in the pellet after ultrasonic disruption and centrifugation instead of in the fermentation *** original strain most likely only producedα-LA intracellular,but not *** improve the expression and secretion ofα-LA in RIK1285,a library of 173 homologous SPs from the *** 168 genome was fused with target LALBA gene in the pBE-S vector and expressed extracellularly in *** YjcN was determined to be the best signal *** in supernatant were observed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and purified by nickel column to calculate the highest yield signal *** addition,different promoters(P_(aprE),P_(43),and P_(glv))were compared and *** results indicated that the strain RIK1285-pBE-P_(glv)-YjcN-LALBA had the highestα-LA yield,reaching 122.04μg/*** study demonstrates successful expression and secretion of humanα-LA in *** and establishes a foundation for simulating breast milk for infant formulas and developing bioengineered milk.

Proteomic studies of the effects of processing techniques on properties of abalone muscles: A comprehensive review

Background: With the increasing per capita demand for animal protein， there is a growing interest in the abundant abalone protein resources. Abalone proteins are known for their nutritional and functional properties that contribute to flavor and texture. Scope and approach: We systematically constructed the relationship between abalone protein， processing， and proteomics. This paper reviews the nutritional properties of abalone proteins and evaluates the effects of different thermal processing techniques， non-thermal processing， and freezing on abalone proteins. In addition， we synthesize published abalone proteomics studies and the use of proteomics technology to better elucidate the quality changes of abalone and its products， and as a technical basis for the study of blue food marker proteins. It is important direction to clearly explain the protein composition and meat quality mechanism of abalone in the processing and storage by *** findings and conclusions: During various types of thermal processing， non-thermal processing， and freezing of abalone， the various chemical forces between protein molecules are disrupted， which in turn leads to different degrees of denaturation， aggregation， and gelation， which may have an impact on the organoleptic properties， bioavailability， and digestibility of abalone muscle. Proteomics is used in abalone biology studies to understand developmental biology， physiology， disease/stress， and species identification and can also be a powerful tool to characterize processing methods on abalone quality properties.

Rational protein engineering of thermostable heparinase I from Bacteroides thetaiotaomicron for highly efficient heparin degradation

Heparin， a glycosaminoglycan， is a stable source of carbon that supports the growth of microorganisms in the human intestine. It is also a commonly used anticoagulant drug in clinical practice， with significant therapeutic effects. Low molecular weight heparin （LMWH） is a highly active low molecular weight fragment obtained via enzymatic reaction or the chemical degradation of heparin. LMWH has been applied globally in the prevention and treatment of venous thromboembolism in thrombosis patients. Simultaneously， as a potential prebiotic， because of its low molecular weight， LMWH can be well degraded by the gut microbiota to maintain intestinal balance. Enzymatic heparin degradation has recently emerged as a viable disposal method for LMWH preparation; however， only very few benchmark enzymes have been thoroughly described and subjected to protein engineering to improve their properties over the past few years. The commercialization of enzymes will require the development of robustly engineered enzymes that meet the demands of industrial processes. Herein， we report a rational protein engineering strategy that includes molecular dynamic simulations of flexible amino acid mutations and disulfide bond screening. Several Bt-HepI mutants were obtained and screened for high thermal stability. We obtained the Bt-HepID204C/K208C/H189W/Q198Rvariant， which features a stabilized protein surface structure， with a 1.3-fold increase in kcat/Km， a 2.44-fold increase in thermal stability at 50℃， and a 1.8-fold decrease in the average molecular weight of LMWH produced at 40℃ compared with that seen with Bt-HepIWT. Our study establishes a strategy to engineer thermostable HepI to underpin its industrial applications.

Recent research on the genome of Bifidobacterium bifidum has mainly focused on the isolation sources(intestinal tract niche)recently,but reports on the isolation region are *** study analyzed the differences in the genome of *** isolated from different geographical populations by comparative genomic *** at the genome level indicated that the GC content of American isolates was significantly higher than that of Chinese and Russian *** phylogenetic tree,based on 919 core genes showed that *** might be related to the geographical characteristics of isolation ***,functional annotation analysis demonstrated that copy numbers of carbohydrate-active enzymes(CAZys)involved in the degradation of polysaccharide from plant and host sources in *** were high,and 18 CAZys showed significant differences across different geographical populations,indicating that *** had adapted to the human intestinal environment,especially in the groups with diets rich in *** habits were one of the main reasons for the differences of *** across different geographical ***,*** exhibited high diversity,evident in glycoside hydrolases,the CRISPR-Cas system,and *** study provides a genetic basis for further research and development of ***.

Andrias davidianus(Chinese giant salamander,CGS)is the largest and oldest extant amphibian species in the world and is a source of prospective functional food in ***,the progress of functional peptides mining was slow due to lack of reference genome and protein sequence *** this study,we illustrated full-length transcriptome sequencing to interpret the proteome of CGS meat and obtain 10703 coding DNA *** functional annotation and amino acid composition analysis,we have discovered various genes related to signal transduction,and 16 genes related to *** have also found vast variety of functional peptides through protein coding sequence(CDS)analysis by comparing the data obtained with the functional peptide ***-Pro-Ile predicted by the CDS analysis was released from the CGS meat through enzymatic hydrolysis,suggesting that our approach is *** study suggested that transcriptomic analysis can be used as a reference to guide polypeptide mining in CGS meat,thereby providing a powerful mining strategy for the bioresources with unknown genomic and proteomic sequences.

Dendrimer-induced synthesis of porous organosilica capsules for enzyme encapsulation

Organic matter-induced mineralization is a green and versatile method for synthesizing hybrid nanostructured materials,where the material properties are mainly influenced by the species of natural biomolecules,linear synthetic polymer,or small molecules,limiting their ***,we adopted dendrimer poly(amidoamine)(PAMAM)as the inducer to synthesize organosilica-PAMAM network(OSPN)capsules for mannose isomerase(MIase)encapsulation based on a hard-templating *** structure of OSPN capsules can be precisely regulated by adjusting the molecular weight and concentration of PAMAM,thereby demonstrating a substantial impact on the kinetic behavior of the MIase@OSPN *** MIase@OSPN system was used for catalytic production of mannose from Dfructose.A mannose yield of 22.24% was obtained,which is higher than that of MIase in organosilica network capsules and similar to that of the free *** overall catalytic efficiency(kcat/Km)of the MIase@OSPN system for the substrate D-fructose was up to 0.556 s^(-1)·mmol^(-1)·***,the MIase@OSPN system showed excellent stability and recyclability,maintaining more than 50% of the yield even after 12 cycles.

Conventional filling therapy fails to fundamentally reduce oral cariogenic ***,oral microbiota follow-up intervention after filling would be necessary for improving dental caries *** recruited 9 caries-free individuals,and 89 dental caries subjects(5 dropouts).Eighty-nine patients were randomized into three groups:caries(n=8;no treatment),control(n=40;filling),and postbiotics(n=41;filling and 14-day Probio-Eco®intervention).Salivary samples were collected at 0 day(after filling)and 14 *** results showed that the diversity of dental caries oral microbiota was significantly increased compared with healthy subjects,and filling could restore a healthier oral microbiota partially and *** intervention keeps a low ***-occurrence network analysis showed that a more stable oral microbiota structure after postbiotics *** and functional annotation of the microbiota revealed that postbiotics co-treatment significantly:increased the relative abundance of Pseudomonas and ***,decreased the relative abundance of Prevotella shahii,and enriched the energy metabolism-related ***-predicted phenotypes inferred to an oral microbiota with decreased potential pathogenic bacteria and increased oxidative stress-tolerant bacteria after postbiotics ***,it suggested that postbiotics co-treatment could be a promising strategy that restores the oral microecological balance for dental caries.
