MicroR-9c-5p and novel-mir50 co-target Akt to regulate Lasioderma serricorne reproduction

High fecundity is a common characteristic of insect pests which increases the difficulty of population ***/threonine kinase Akt is an indispensable component of the insulin signaling *** of LsAkt severely hinders reproduction in Lasioderma serricorne,a stored product insect ***,the post-transcriptional pathway of LsAkt in *** remains *** study identified 2 binding sites of miR-9c-5p and novel-mir50 in the coding sequences of *** expression profiles of 2 microRNAs(miRNAs)and LsAkt displayed an opposite pattern during the adult *** reporter assay showed that novel-mir50 and miR-9c-5p could downregulate the expression of *** of miR-9c-5p and novel-mir50 by injection of mimics inhibited the expression of LsAkt and reduced oviposition,decreased egg hatchability,and blocked ovarian *** also decreased the expression of genes involved in ovarian development(LsVg and LsVgR)and the nutritional signaling pathway(LsTOR,LsS6K,and Ls4EBP),and reduced the phosphorylation of ***,injection of miR-9c-5p and novel-mir50 inhibitors induced the expressions of LsAkt,LsVg,LsVgR,LsTOR,LsS6K,and Ls4EBP,enhanced Akt phosphorylation level,and accelerated ovarian *** of bovine insulin downregulated the expression of miR-9c-5p and novel-mir50 and upregulated the LsAkt *** also rescued the reproductive development defects associated with miR-9c-5p/novel-mir50 overexpression,forming a positive regulatory loop of insulin *** results indicate that miR-9c-5p/novel-mir50 regulates the female reproduction of *** by targeting Akt in response to insulin *** data also demonstrate the effects of the insulin/miRNA/Akt regulatory axis in insect reproduction.

Nutritional regulation of skeletal muscle energy metabolism, lipid accumulation and meat quality in pigs

The quality of pork determines consumers’ purchase intention, which directly affects the economic value of pork. Minimizing the proportion of inferior pork and producing high quality pork are the ultimate goals of the pig industry. Muscle energy metabolism, serving as a regulative hub in organism energy expenditure and storage as a fat deposit, is compatible with myofiber type composition, affecting meat color, intramuscular fat content, tenderness, p H values and drip loss. Increasing data illustrate that dietary nutrients and bioactive ingredients affect muscle energy metabolism, white adipose browning and fat distribution, and myofiber type composition in humans, and rodents. Recently, some studies have shown that modulating muscle energy metabolism and lipid accumulation through nutritional approaches could effectively improve meat quality. This article reviews the progress and development in this field, and specifically discusses the impacts of dietary supply of amino acids, lipids, and gut microbiota as well as maternal nutrition on skeletal muscle energy metabolism, lipid accumulation and meat quality of pigs, so as to provide comprehensive overview with respect to effective avenues for improving meat quality.

Expression of Porcine Interleukin-2 and Porcine Interleukin-6 and Their Adjuvant Effects on Gene Deleted Vaccine of Pseudorabies Virus(TK^-/gG^-/LacZ^+)

Porcine interleukin-2 and porcine interleukin-6 cDNA sequences were cloned into the expressing vectors pET-28a and pGEX-KG respectively. They were expressed in E. coli BL21(DE3)with high-level production. The gene deleted vaccine of pseudorabies virus Ea strain(TK-/gG-/LacZ+)was mixed with the two different purified recombinant proteins each, or both, with the doses of 2, 5 or 10 μg ml-1. Ten groups of pseudorabies negative antibody swines were immuned twice with tested vaccines with different doses, or control vaccine, respectively. The antibody liters of the test groups were detected by neutralization test, and the daily weight gains of swines were calculated and analyzed statistically. In the study, all the neutralizing antibody ti-ters in test groups were higher than the control group, and the recombinant proteins appeared a dose dependent adjuvant effect. The tested vaccines with 2 μg ml-1 pIL-2 and with 10 μg ml-1 pIL-2/pIL-6 got significant and extremely significant differences, compared with the vaccines without pILs. The difference of the daily weight gain indicated the potential positive influence of pIL-2 and pIL-6 on immune protection.

Cholesterol-25-hydroxylase(CH25 H)is a membrane protein associated with endoplasmic reticulum,and it is an interferon-stimulated factor regulated by ***25 H catalyzes cholesterol to produce 25-hydroxycholesterol(25 HC)by adding a second hydroxyl to the 25 th carbon atom of *** studies have shown that both CH25 H and 25 HC could inhibit the replication of many *** this study,we found that ectopic expression of CH25 H in HEK-293 T and BHK-21 cell lines could inhibit the replication of Seneca Valley virus(SVV)and that there was no species *** the other hand,the knockdown of CH25 H could enhance the replication of SVV in HEK-293 T and BHK-21 cells,indicating the importance of CH25 *** some extent,the CH25 H mutant without hydroxylase activity also lost its ability to inhibit SVV *** studies demonstrated that 25 HC was involved in the entire life cycle of SVV,especially in repressing its adsorption *** study reveals that CH25 H exerts the advantage of innate immunity mainly by producing 25 HC to block virion adsorption.

Hepatic steatosis is associated with dysregulated cholesterol metabolism and altered protein acetylation dynamics in chickens

Background Hepatic steatosis is a prevalent manifestation of fatty liver, that has detrimental effect on the health and productivity of laying hens, resulting in economic losses to the poultry industry. Here, we aimed to systematically investigate the genetic regulatory mechanisms of hepatic steatosis in laying *** Ninety individuals with the most prominent characteristics were selected from 686 laying hens according to the accumulation of lipid droplets in the liver, and were graded into three groups, including the control, mild hepatic steatosis and severe hepatic steatosis groups. A combination of transcriptome, proteome, acetylome and lipidome analyses, along with bioinformatics analysis were used to screen the key biological processes, modifications and lipids associated with hepatic *** The rationality of the hepatic steatosis grouping was verified through liver biochemical assays and RNA-seq. Hepatic steatosis was characterized by increased lipid deposition and multiple metabolic abnormalities. Integration of proteome and acetylome revealed that differentially expressed proteins(DEPs) interacted with differentially acetylated proteins(DAPs) and were involved in maintaining the metabolic balance in the liver. Acetylation alterations mainly occurred in the progression from mild to severe hepatic steatosis, i.e., the enzymes in the fatty acid oxidation and bile acid synthesis pathways were significantly less acetylated in severe hepatic steatosis group than that in mild group(P < 0.05). Lipidomics detected a variety of sphingolipids(SPs) and glycerophospholipids(GPs) were negatively correlated with hepatic steatosis(r ≤-0.5, P < 0.05). Antimicrobial peptides(AMPs)in insects are endogenous peptides that are effector components of the innate defense system of the *** may serve as antimicrobial agents because of their small molecular weight and broad-spectrum antimicrobial *** this study,we performed transcriptome analysis of cigarette beetle(Lasioderma serricorne)larvae,parasitized by the ectoparasitic wasp,Anisopteromalus *** AMP genes were significantly upregulated following *** parasitism,postulating the hypothesis that the parasitization enhanced the host's resistance against pathogenic microorganisms through the regulation of host AMP ***,3 AMP genes(LsDef1,LsDef2,and LsCole)were significantly upregulated and we studied their immune function in *** challenge and functional analysis showed that LsCole was responsible for the immune response against Gram-negative and Gram-positive bacteria,while LsDef1 and LsDef2 were involved in insect defense against Gram-positive *** recombinant LsCole exhibited antimicrobial activities against the Gram-negative bacterium Escherichia coli and the Gram-positive bacterium Staphylococcus ***2 showed an antibacterial effect against *** and LsDef2 exhibited antibiofilm activity against *** 2 AMPs disrupted cell membranes and caused leakage of *** cell *** results indicated that the 3 AMPs in *** are involved in the innate immunity of this pest *** AMPs may have potential as antimicrobial agents for bacterial infection ***,data are discussed in relation to new control strategies with greater biosafety against pest insects with use of microbial biocontrol agents in combination with RNA interference against the insect's defensive AMP genes.

Effects of biochanin A on lactational performance,nitrogen metabolism,and blood metabolites in dairy cows

Optimizing nitrogen utilization efficiency and mitigating nitrogen losses in cows plays a pivotal role in fostering economic sustainability within contemporary agricultural *** A(BCA),a natural component in red clover,has the potential to improve nitrogen metabolism in dairy *** primary objective of this study was to probe the impact of biochanin A supplementation on lactational performance,nitrogen metabolism,and blood metabolites in dairy cows.A complete randomized block design experiment was conducted over 28 d,involving 36 multiparous Holstein cows(comparable milk yield=37.1±2.90 kg,BW=642±70.0 kg,days in milk=92±8.0 d,and parity=2.4±0.50),which were allocated to three treatment groups:the Control group(with 0 g/d BCA),the Low group(with 10 g/d per cow BCA),and the High group(with 40 g/d per cow BCA).Biochanin A supplementation improved the lactational performance of cows by increasing milk yield by 6.3%(P=0.007)and feed efficiency by 12.7%(P=0.009).Total intestinal apparent digestibility was unaffected by BCA supplementation(P>0.05),but microbial nitrogen was increased by 30.0%(P=0.002)for promoting nitrogen utilization efficiency by 20.7%(P=0.004).Milk competent yields(protein,lactose,and non-fat milk solid)were increased with increasing BCA supplementation(P 0.05).BCA did not affect body health of dairy ***,none of the plasma endocrine hormones were affected(P>0.05).A total of 95 significantly different metabolites were screened from the plasma metabolites of cows in the BCA-added and non-added *** performing an enrichment analysis of the metabolic pathways associated with the different metabolites,six specific pathways were identified:bile acid biosynthesis,aspartate metabolism,pyrimidine metabolism,arginine and proline metabolism,the urea cycle,and ammonia *** inclusion of BCA is suggested to enhance milk yield and modulate nitrogen metabolism by influencing relevant metabolites within the metabolic pathwa
