Gene cloning in repeat-rich polyploid genomes remains ***,we describe a strategy for overcoming major bottlenecks in cloning of the powdery mildew resistance gene(r-gene)Pm69 derived from tetraploid wild emmer wheat.A conventional positional cloning approach was not effective owing to suppressed *** sorting was compromised by insufficient purity.A Pm69 physical map,constructed by assembling Oxford Nanopore Technology(ONT)long-read genome sequences,revealed a rapidly evolving nucleotide-binding leucine-rich repeat(NLr)r-gene cluster with structural variations.A single candidate NLr was identified by anchoring rNA sequencing reads from susceptible mutants to ONT contigs and was validated by virus-induced gene ***69 is likely a newly evolved NLr and was discovered in only one location across the wild emmer wheat distribution range in ***69 was successfully introgressed into cultivated wheat,and a diagnostic molecular marker was used to accelerate its deployment and pyramiding with other r-genes.

Various types of ground baits are loaded to lakes by anglers,but the ecological role of these materials is often ***,the aims of this study were to conduct a quantitative and qualitative assessment of the baits used by anglers in the largest lake of Central-Europe(Lake Balaton,Hungary),estimate the annual nutrient balance of recreational fishing,and relate the nutrient loading from this source to the total external *** study demonstrated that popular angler baits varied substantially in their nutrient contents and in their molar nitrogen to phosphorus(N:P)*** net nutrient balance of recreational fishing was+19.3 t total N year^(-1)and+5.2 t total P year^(-1),equivalent to 0.7%and 3.2%of the estimated total annual N and P loads,*** rate of nutrient loading may be doubled if the present trends continue:anglers will use 20%more bait with an even higher nutrient content and will also show higher propensity to release the fish after *** turn,sustainable nutrient balance of fisheries management could be achieved by limiting the anglers’bait use to 1.5 kg day^(-1)and restricting baits to those which have relatively low P(d N(<1.5%)contents.

Wheat sharp eyespot, a stem disease caused by the soilborne fungus rhizoctonia cerealis van der Hoeven,has become a threat to wheat production worldwide. Exploiting resistance resources from wild relatives of wheat is a promising strategy for controlling this disease. In this study, a new wheat–dasypyrum villosum T2dS·2V#4L translocation line in the background of Chinese Spring(CS) showed stable resistance to r. cerealis. Introgression of the T2dS·2V#4L chromosome into wheat cultivar Aikang 58 by backcrossing produced a marked increase in sharp eyespot resistance in NIL-T2dS·2V#4L in comparison with NILT2dS·2dL, and no detrimental effects of 2V#4L on agronomic traits were observed in the BC2F2, BC2F2:3,and BC2F2:4generations. Flow-sorted sequencing of 2V#4L yielded 384.3 Mb of assembled sequence, and8836 genes were predicted of which 6154 had orthologs in at least one of the 2AL, 2BL, and 2dL arms of CS, whereas 1549 genes were unique to 2V#4L. About 100,000 SNPs were detected in genes of 2V#4L and2dL in 10 sequenced bread wheat cultivars. A Kompetitive Allele Specific Polymerase chain reaction and30 conserved ortholog sequence markers were developed to trace the 2V#4L chromatin in wheat backgrounds. T2dS·2V#4L compensating translocation lines represent novel germplasm with sharp eyespot resistance and the markers will allow rapid detection in breeding programs.
