Inflammation is a driving force of hematopoietic stem cells(HSCs) aging, causing irreversible exhaustion of functional HSCs. However, the underlying mechanism of HSCs erosion by inflammatory insult remains poorly understood. Here, we find that transient LPS exposure primes aged HSCs to undergo accelerated differentiation at the expense of self-renewal, leading to depletion of HSCs. Meanwhile, the central regulator nuclear factor kappa B(NF-κB) mediating functional impairment by inflammation insult induces differential transcriptional response in aged HSCs compared with young HSCs, with precocious activation of myeloid lineage genes. Altered compartmentalization and chromatin loop formation are associated with aging-related differential transcriptional response in HSCs upon lipopolysaccharide(LPS)stimulation. Mechanistically, enhancer and promoter regions of myeloid lineage genes in aged HSCs are more accessible and display more rapid and prominent CTCF occupancy upon LPS stimulation. Our study provides comprehensive resources for the three-dimensional(3D)genome structure of HSCs and sheds light into the ordered genome organization and the associated transcriptome signature underlying HSCs aging.

The human microbiota-associated(HMA) mice model, especially the germ-free(GF)-humanized mice, has been widely used to probe the causal relationships between gut microbiota and human diseases such as type 1 diabetes(T1D). However, most studies have not clarified the extent to which the reconstruction of the human donor microbiota in recipient mice correlates with corresponding phenotypic reproducibility. In this study, we transplanted fecal microbiota from five patients with T1D and four healthy people into GF mice, and microbiota from each donor were transplanted into 10 mice. Mice with similar microbiota structure to the donor exhibited better phenotypic reproducibility. The characteristics of the microbial community assembly of donors also influenced the phenotypic reproducibility in mice, and individuals with a higher proportion of stochastic processes showed more severe disorders. Microbes enriched in patients with T1D had a stronger colonization potential in mice with impaired glucose metabolism, and microbiota functional features related to T1D were better reproduced in these mice. This indicates that assembly traits and colonization efficacy of microbiota influence phenotypic reproducibility in GF-humanized mice. Our findings provide important insights for using HMA mice models to explore links between gut microbiota and human diseases.

Aim: Yiqi-Liangxue Recipe(YL) is a compound preparation of Chinese medicine used for preventing cardiovascular events after percutaneous coronary intervention(PCI)(Patent No. 200810240175.4). Maintaining the integrity of endothelia is one of the most effective approaches to prevent restenosis. Given its clinical protective effects on long-term prognosis, we investigated the mechanisms of YL in protecting vascular endothelial ***: We prepared drugs serum and human umbilical vein endothelial cell(HUVEC) lines. The injured model was employed with Angiotensin II(Ang II). The YL group was employed with YL treatment. The ARB medicine group was employed with Losartan Potassium treatment. The combination of Chinese medicine and western medicine(YL+ARB) group was employed with both YL and ARB. The control group was unemployed with injury and medical treatment. For each group the cell migration rate(CM) and cell proliferation rate(CP) were measured. The concentration of Nitric Oxide(NO), Reactive Oxygen Species(ROS), Endothelin-1(ET-1) were assessed. The gene expression level of ET-1 was ***: YL+ARB group significantly promoted the speed of the CM/CP of the injured HUVECs. Compared with model group, the concentration of NO increased after the drug intervention, and the concentration of ET-1 decreased. Compared with YL+ARB group, YL and ARB group each had the similar weaker effects, but did not have significant difference. The fluorescence of ROS in YL, ARB and YL+ARB group had no difference because the fluorescence was too strong. Compared with model group, the-2ΔΔCt values were decreased in the YL, ARB and YL+ARB group.
