Material basis and sleep-improving mechanisms of Lily-Ziziphi spinosae semen Decoction：Systematic evidence from LC-MS， network pharmacology and animal experiments

Lily-Ziziphi spinosae semen Decoction (LZ) is known for its blood nourishing， mind calming， body sedation， and sleep promoting effects in traditional Chinese medicine. However， its material basis and underlying mechanisms have not yet been clearly defined. This study applies liquid chromatography-mass spectrometry， network pharmacology， and animal studies to reveal the material basis and sleep-improving mechanisms of LZ. The mixed decoction (LZ-ME) and single decoction (LZ-SE) were prepared to study their chemical components. Network pharmacology was used to predict the sleep-improving targets and signaling pathways of LZ. ICR mice were intragastrically administered saline (NC)， melatonin (positive control group， 0.50 mg/kg)， low (12.90 g/kg)， medium (25.70 g/kg)， and high (38.60 g/kg) dose of LZ-ME and LZ-SH for 30 days.. The results showed that LZ-ME could prolong the sleep duration and shorten the sleep latency in sodium barbiturate induced mice model. The results of chemical composition showed that total polysaccharides， total flavonoids， total saponins， and total alkaloids in LZ-ME were significantly higher than those in LZ-SE (177.20%， 82.34%， 30.58%， and 11.66%， respectively). A total of 58 chemical components were identified by UPLC-QTOF-MS， and eight representative difference components of LZ-ME and LZ-SE were found. LZ-ME significantly increased tumor necrosis factor (TNF-α) in mice serum and neurotransmitter γ-aminobutyric acid (GABA) levels in mice hippocampus， decreased dopamine (DA) and glutamate (Glu) levels in mice hippocampus (P < 0.05). Furthermore， LZ-ME up-regulated the abundance of the beneficial bacteria Lactobacillus and Eubacterium＿R， down-regulated the abundance of the harmful bacteria Lachnospiraceae et al. The polysaccharides， flavonoids (spinosin and 6?-feruloylspinosin) and saponins (jujuboside A and jujuboside B) were the main material bases for the sleep-promoting effects of LZ. Farnesoid X receptor(FXR)is a ligand-activated nuclear receptor and plays important roles in the regulation of metabolism and homeostasis of several important physiological substances,such as bile acids,glucose,and *** such,FXR has become a promising therapeutic target for the treatment of several metabolic diseases and liver ***,fargesone A(FA),a natural product from Magnolia fargesii was identified to be a novel,potent FXR agonist that demonstrated good in vitro and in vivo ***,the detailed interaction mechanism of FA with FXR remains *** this study,we employed multiple computational approaches including molecular docking,molecular dynamics simulation,and binding free energy calculation to address the *** comparisons of the structural dynamics and binding free energies,an optimal binding mode was identified,in which FA interacts with FXR via a direct hydrogen bond with His447 and hydrophobic interactions with multiple residues,such as Leu287,Met290,Met328,Ile352,and *** mutants,namely,H447F and L287N,were further constructed to validate the importance of the identified *** anticipate that these findings could be helpful for future rational design of new FA analogues targeting FXR.

In recent years, biopharmaceuticals have witnessed remarkable advancements, transforming the landscape of therapeutic interventions. Biopharmaceuticals encompassing therapeutics generated through cutting-edge biotechnological methods have shown promising therapeutic outcomes. However, their clinical success hinges significantly on overcoming drug delivery challenges related to stability, intracellular delivery, immunogenicity, and pharmacokinetic properties. Herein, we provide an overview of various marketed macromolecules, including nucleic acids, and immunotherapeutic agents such as cytokines and monoclonal antibodies, as well as other therapeutic peptides/proteins like enzymes, hormones, and coagulation factors. Our primary focus is on elucidating the delivery challenges associated with these macromolecules and highlighting the pivotal role played by drug delivery platforms in the development of currently marketed products, offering valuable insights for both scientific research and the pharmaceutical industry.

Dementia is characterized by synaptic and neuronal dysfunction in disease-specific brain *** failure of dementia clinical trials with therapeutic drugs targeting abnormal protein aggregates has caused researchers to shift their focus to synaptic functions and increased the importance of clinically available imaging for synaptic density and the development of synapse-targeted *** density imaging with positron emission tomography(PET)tracer enables non-invasive detection of synaptic loss and hence investigates the association with other neuropathological events exemplified by disease-specific abnormal protein *** studies have reviewed the progress of synaptic density imaging;however,to our knowledge,there is no article yet that summarizes the research progress of multimodal imaging of synaptic density tracers combined with other dementia ***,synaptic function intervention for dementia therapy has not yet been *** this review,first we detail the progress of synaptic density imaging including tracer development and preclinical/clinical application,followed by a discussion of multimodal imaging of synaptic density tracers combined with classic dementia biomarkers in the clinical research ***,we briefly summarize the synapse-targeted drugs for dementia therapy.

The present study aims to establish a reproducible large animal experimental unit using a minipig model to monitor cardiac function changes.A 90-min closed-chest bal-loon occlusion of the left anterior descending branch of the coronary artery was used to induce myocardial infarction in Pannon *** monitor the cardiac function,measurements were made by cardiac magnetic resonance imaging(cMRI),invasive pressure monitoring,and a Pulse index Continuous Cardiac Output(PiCCO)hemo-dynamic system at 0,72,and 720 h during the follow-up ***-diastolic and end-systolic volumes(EDV,ESV),left ventricular ejection fraction(LVEF)obtained by cMRI evaluation,global ejection fraction and aortic dP/dt_(max)obtained by the invasive method,were recorded and *** 72-and 720-h EDV data showed a signifi-cant increase(p=0.012,<0.001)compared to baseline,and the Day 30 data showed a significant increase compared to Day 3(p=0.022).The ESV 72 h after the infarction showed a significant increase(p=0.001)compared to baseline,which did not change significantly by Day 30(p=0.781)compared to Day *** and ESV were signifi-cantly negatively correlated with aortic dp_(max),and ESV was significantly correlated with *** LVEF and dP_(max),a significant(p<0.001 and p=0.002)worsening was demonstrated at Day 3 compared to baseline,which was no longer statistically de-tectable for LVEF at Day 30(p=0.141),while the difference for dP_(max)was maintained(p=0.002).The complementary use of PiCCO hemodynamic measurements in large animal models makes the previously used methodologies more robust and reliable.

The first total synthesis of(+)-taberdicatine B and(+)-tabernabovine B has been accomplished in 10steps with 26.9% overall yield and 15 steps with 7.3% overall yield, respectively. The prominent features of this efficient synthetic strategy include the following:(1)(+)-Taberdicatine B and(+)-tabernabovine B were accessed from common advanced intermediates by varying the substituents;(2) A one-pot asymmetric bromocyclization/hydrolysis was explored to assemble HPI skeleton;(3) Dieckmann condensation to form β-keto ester for the assembly of seven-membered ring;(4) An ester reduction/amide semireduction/cyclization sequence was applied to form the cage-like framework.

Objective:Myocardial inflammation during myocardial infarction(MI) could be inhibited by regulating arachidonic acid(AA) *** studies demonstrated that Sini Decoction(SND) was identified to be an effective prescription for treating heart failure(HF) caused by *** the anti-inflammatory mechanism of SND remained *** work was designed to investigate the anti-inflammato ry mechanism of SND through the AA metabolism pathway in vitro and in vivo ***:An inflammatory injury model of H9c2 cells was established by lipopolysaccharide(LPS)-stimulated macrophage-conditioned medium(CM).The MI model was built by the ligation of left anterior descending(LAD) branch of coronary artery in ***,the rats were divided into five groups:sham group,MI group,MI+Celecoxib group,MI+low-dose SND group(SND-L) and MI+high-dose SND group(SND-H).Cardiac function,histopathological changes and serum cytokines were examined four weeks *** blot analysis was conducted to verify the key enzymes levels in the AA metabolic pathway,including phospholipase A2(PLA2),cyclooxygenases(COXs) and lipoxygenases(LOXs).Results:These in vivo results demonstrated that SND could improve the cardiac function and pathological changes of rats with MI,and regulate the key inflammatory molecules in the AA metabolism pathway,including sPLA2,COX-1,COX-2,5-LOX and *** vitro,SND could decrease the release of proinflammatory cytokines including TNF-α and IL-6 and inhibit cell apoptosis in CM-induced H9c2 ***,SND could protect H9c2 cells from the damage of CM by regulating nuclear factor kappa-B(NF-κB) signal pathway and the expression of ***: SND may be a drug candidate for anti-inflammatory treatment during MI by regulating the multiple targets in the AA metabolism pathway.
