A hidden fault prediction model based on the belief rule base with power set and considering attribute reliability

Hidden faults are important characteristics of complex systems that cannot be observed *** hidden behavior of a system, such as the health state and safety level, has a direct correlation with these hidden faults. After the predicted hidden behavior reaches a fault boundary, certain measures must be taken to avoid fault occurrences. Thus, hidden faults can be predicted by the hidden behavior of a *** belief rule base(BRB) has been used to predict hidden behaviors. However, two problems remain to be solved in engineering practice. First, when the observed information is absent, ignorance may exist in the output. If only global ignorance is considered, it may be unreasonable in certain cases, which can influence the prediction model. Second, the effects of disturbance factors such as noise and sensor quality may cause the reliability of the gathered information to decline, which indirectly leads to unreliability of the hidden behavior. Thus, to address the global ignorance and unreliable hidden behavior, a new hidden BRB model with a power set and considering attribute reliability(PHBRB-r) is proposed for hidden fault prediction. In the PHBRB-r model, the effects of disturbance factors on hidden behavior are considered using attribute reliability, and the discernment frame is a power set. A case study of hidden fault prediction is conducted to demonstrate the effectiveness of the PHBRB-r model.

Objective: The molecular mechanism of prostate cancer is poorly understood. The aim of the study was to investigate the prevalence and prognostic value of promoter hypermethylation of retinoic acid receptor beta (RARB) and p16 among benign prostatic hyperplasia (BPH) and prostate cancer patients. Methods: In this case-control study, 63 patients were included in three groups; 21 with BPH as the control group, 21 with prostate cancer and good prognostic factors (based on prostate-specific antigen, Gleason score and stage) as good prognosis group, and 21 with prostate cancer and poor prognostic features as poor prognosis group. The prostate biopsy specimen of each individual was examined for hypermethylation of RARB and p16 promoters by methylation specific PCR (MSPCR). Results: Seven (33.3%) patients with good prognosis and 15 (71.4%) patients with poor prognosis were positive for RARB methylation, which were significantly higher than controls (P <0.0001). p16 promoter methylation was shown in 19.0% and 47.6% patients with good and poor prognosis, respectively. The RARB and p16 promoter methylation in the poor prognosis group was significantly higher than that in the good prognosis group (P =0.02 for RARB and P<0.0001 for p16). Conclusion: Hypermethylation of RARB and p16 promoters may predict prognosis in prostate cancer.

Selectively enhancing radiosensitivity of cancer cells via in situ enzyme-instructed peptide self-assembly

The radiotherapy modulators used in clinic have disadvantages of high toxicity and low *** the first time,we used the in situ enzyme-instructed self-assembly(EISA)of a peptide derivative(Nap-GDFDFpYSV)to selectively enhance the sensitivity of cancer cells with high alkaline phosphatase(ALP)expression to ionizing radiation(IR).Compared with the in vitro pre-assembled control formed by the same molecule,assemblies formed by in situ EISA in cells greatly sensitized the ALPhigh-expressing cancer cells to y-rays,with a remarkable sensitizer enhancement *** results indicated that the enhancement was a result of fixing DNA damage,arresting cell cycles and inducing cell ***,in vitro pre-formed assemblies mainly localized in the lysosomes after incubating with cells,while the assemblies formed via in situ EISA scattered in the cell *** accumulation of these molecules in cells could not be inhibited by endocytosis *** believed that this molecule entered cancer cells by diffusion and then in situ self-assembled to form nanofibers under the catalysis of endogenous *** study provides a successful example to utilize intracellular in situ EISA of small molecules to develop selective tumor radiosensitizers.

Chemotherapy has limited efficacy in the treatment of advanced and metastatic pancreatic cancer(PC), and has serious side effects. The development of novel effective agents, especially targeted therapy, is essential for patients with PC. We present a 58-year-old Chinese woman initially diagnosed with locally advanced PC. As the disease progressed to Stage Ⅳ, the patient was unable to tolerate chemotherapy after the fourth-line treatment. She was then treated with apatinib, a novel and highly selective tyrosine kinase inhibitor of vascular endothelial growth factor receptor-2 and achieved a progression-free-survival of 7 mo. All drug-related side effects were well controlled with medication. To the best of our knowledge, this is the first case of PC which responded to apatinib. Considering this remarkable response, apatinib may be a promising agent in the treatment of PC. We also reviewed the literature on chemotherapy and targeted therapy, especially the anti-angiogenesis therapy for patients with PC, and investigated the effect of apatinib in other solid tumors as well.

The aim of this study was to design a cage-like radiotherapy system(CRTS)to further promote the clinical application of noncoplanar *** CRTS comprises two stands,two O-rings,several arc girders,an X-ray head,an imaging subsystem,and a treatment *** X-ray head rotates with O-rings around the patient’s body and slides along the arc *** with the C-arm linear accelerator(C-Linac),the clinically available spatial irradiation angle ranges(SIARs)of the CRTS for the head,chest,and abdomen were 33%,63.6%,and 62.6%larger,***,according to a preliminary planning comparison based on the dose distribution simulation method,the CRTS achieved much better protection of normal tissue than the ***,the CRTS enabled accurate noncoplanar irradiation without movement of the body being irradiated,allowed automatic control of the movements of different parts without risk of collisions,and provided continuous radiation over an angle that considerably exceeded a full *** advantages make CRTS very promising for noncoplanar radiotherapy.

AIM:To analyze the outcome of patients who received concurrent capecitabine(Xeloda) and radiation(XRT) compared to the established concurrent 5-fluorouracil(5-FU) with radiation(5FU-RT) and fluoropyrimidine-based chemotherapy alone as adjuvant treatment in gastric ***:All patients with gastric cancers who received adjuvant treatment at the National Cancer Centre Singapore between 1996 and 2006 were *** outcomes of patients who received XRT were compared with those who had 5FU-RT or chemotherapy alone as adjuvant therapy for gastric ***:A total of 108 patients were *** age at diagnosis was *** majority of the patients(64.8%) had advanced stage Ⅲ and Ⅳ disease(with no distant metastasis).All except 4 patients had D2 *** one patients(19.4%) had positive surgical resection *** three patients received XRT compared with 52 who had 5FU-RT and 23 who received chemotherapy *** the patients in the chemotherapy-only group,all had fluoropyrimidine-based therapy,with added cisplatin in 7 patients and epirubicin in 2 *** recurrence-free survival was longer for the XRT group(52 mo) compared to the 5FU-RT(35 mo) and chemotherapy-only groups(25 mo)(P=0.48).The patients in the XRT group achieved similar median overall survival(53 mo) as the 5FU-RT(54 mo) and the chemotherapy-only groups(44 mo)(P=0.5).CONCLUSION:Capecitabine with concurrent radiation was as effective as concurrent 5FU with radiation or fluoropyrimidine-based chemotherapy alone when used as adjuvant treatment in patients with gastric cancers.

Esophageal squamous cell carcinoma(ESCC)is one of the most lethal cancers *** this study,we aimed to investigate the underlying mechanisms of metastasis inhibition by miR-205 in *** microRNA(miRNA)array and quantitative RT-PCR analyses,we found that the expression level of miR-205 was significantly lower in patients with lymph node metastasis compared with that in patients without lymph node *** transfection of miR-205 mimics or inhibitors into ESCC cell lines,a significant negative correlation was observed between the expression level of miR-205 and *** luciferase reporter assays,we revealed that miR-205 inhibited the expression of SMAD1 by targeting the 30untranslated region(30-UTR)of SMAD1 mRNA in ESCC ***,our results showed that miR-205 suppressed the invasion and migration of ESCC cells,whereas Smad1 increased their invasion and *** together,our study demonstrates that miR-205 functions as a suppressor of tumor metastasis by regulating SMAD1expression through targeting the 30-UTR of SMAD1 mRNAin ***,miR-205 may be a potential therapeutic target for miRNA-based therapy of ESCC.
