Objective:To establish the prevalence and associated risk factors of Schistosoma mansoni(***) infection among schoolchildren at a village in Wolaita *** Ethiopia,Methods:A cross-sectional study was carried out among primary schoolchildren.A total of 384 randomly selected study subjects provided stool samples for parasitological examination by Kato-Katz and Formalin-Ether concentration *** parasitological data were obtained from Health Center Laboratory to see the previous history *** infection in the *** analysis was performed using SPSS software version ***:From the total children examined.85.4% were found positive for at least one helminth *** infection(81.3% ) was the most prevalent and the prevalence of STH was 32% ..Moderate and heavy infection intensities were only observed in S,mansoni *** overall heavy intensity of infection was 56.4% .Contact to Bisarc stream was the most important factor for *** infection(OR 3.9) followed by herding cattle near the stream(OR2.527).Males were twice more likely to get the infection than females(OR 1.923).Analysis of secondary parasitological data showed that *** infection was a leading helminthic infection over the past ***:The present study found a higher intensity and prevalence of *** infection in a rural village of Wolaita ***,appropriate integrated control and prevention measures need to be implemented in the study area.

Role of brain Cain electro-acupuncture analgesia and the development of analgesic tolerance to electro-acupuncture and morphine were studied. At the same time, the inhibition by protein synthesis inhibiturs of the development of analgesic tolerance to electro-acupuncture was observed. The results showed that like morphine tolerance, the brain Caand cAMP levels in mice were enhanced with the development of analgesic tolerance to electro-acupuncture. After treatment with protein synthesis inhibitors anisomycin, actinomycin or cycloheximide the development of analgesic tolerance to electro-acupuncture was inhibited, and concurrently, the brain Caand cAMP levels in the animals greatly reduced. From the changes of brain Caand cAMP levels, the analgesic effects by electro-acupuncture, morphine and lanthanides seem to be very similar and share a mutual ion basis and the mechanism of action. Objective:To determine repellent activity of hexane,ethyl acetate,benzene,chloroform and methanol extract of Cardiospermum halicacabum,(***) against Culex quinque/ascialus(***),Aedes aegypti(***) and Anopheles slephensi(***). Methods:Evaluation was carried out in a net cage(45 cm×30 cm×25 cm) containing 100 blood starved female mosquitoes of three mosquito species and were assayed in the laboratory condition by using the protocol of WHO 2005;The plant leaf crude extracts of *** was applied at 1.0,2.5,and 5.0 me/cm^2 separately in the exposed area of the fore *** ethanol served as ***:In this observation,the plant crude extracts gave protection against mosquito bites without any allergic reaction to the test person,and also,the repellent activity was dependent on the strength of the plant *** tested plant crude extracts had exerted promising repellent against all the three ***:From the results it can be concluded the crude extract of *** was potential for controlling ***, *** and *** mosquitoes.

One goal of precise oncology is to re-classify cancer based on molecular features rather than its tissue origin. Integrative clustering of large-scale multi-omics data is an important way for molecule-based cancer classification. The data heterogeneity and the complexity of inter-omics variations are two major challenges for the integrative clustering analysis. According to the different strategies to deal with these difficulties, we summarized the clustering methods as three major categories： direct integrative clustering, clustering of clusters and regulatory integrative clustering. A few practical considerations on data pre-processing, post-clustering analysis and pathway-based analysis arc also discussed.

MicroRNA （miRNA） plays key roles in post-transcriptional regulations. Recently, a competing endogenous RNA （ceRNA） hypothesis has been proposed that miRNA targets could communicate and regulate each other through titrating shared miRNAs, which provides a new layer of gene regulation. Though a number of ceRNAs playing biological functions have been identified, the ceRNA hypothesis remains controversial. Recent experimental and theoretical studies argued that the modulation of a single RNA species could hardly change the expression level of competing miRNA targets through ceRNA effect under normal physiological conditions. Here, we reviewed a common framework to model miRNA regulations, and summarized the current theoretical and experimental studies for quantitative understanding ceRNA effect. By revisiting a coarse-grained ceRNA model, we proposed that network topology could significantly influence the competing effect and ceRNA regulation at protein level could be much stronger than that at RNA level. We also provided a conditional independent binding equation to describe miRNA relative repression on different target, which could be applied to quantify siRNA off-target effect.

The androgen receptor （AR） remains the primary molecular target forprostate cancer （PCa） treatment and for development of novel therapies. Profiling and other analyses of human prostate adenocarcinoma have shown that the AR is still functional during latestage disease in the absence of circulating hormone fol lowing castration therapy. The molecular mechanisms that operate during this ＇cas tration resistant＇ phase are still not well understood. Qi et al. have now implicated the ubiquitin ligase Siah2 as an important mediator of AR action in castration resis tant prostate cancer （CRPC）. Siah2 was found to target repressed AR chromatin complexes for degradation, resulting in activation of ARregulated genes involved in tumor cell proliferation, cell motility and lipid metabolism. The authors show a requirement for Siah2 activity for PCa cell growth under conditions of low androgen, and also that targeting Siah2 results in tumor growth suppression under castrate conditions. These findings identify a new mechanism of AR regulation in progressing disease as well as a novel enzymatic target for therapeutic intervention. The AR is a wellstudied hormone receptor that belongs to the large nuclear receptor gene superfamily. AR is activated by androgen binding （principally 5：~dihydrotestosterone, 5DHT）, which elicits changes in AR resident cytosolic complexes, translocation of the receptor to cell nuclei, formation of multiprotein transcriptional complexes onchromatin, and activation or repression of gene expression. Experiments with in vivo model systems have provided evidence that virtually all physiologic processes affected by androgens require the AR as a mediator of molecular effects at the gene level.1 AR is widely expressed beyond the reproductive systems and is believed to play important roles in several nonreproductive tissues, including muscle and brain. A performance assisted enhancement Kalman filtering algorithm(PAE-KF) for GPS/INS integration navigation in urban areas was presented in this work. The aim of this PAE-KF algorithm was to prevent "deep contamination" caused by error GPS data. This filtering algorithm effectively combined fault estimation of raw GPS data and nonholonomic constraint of vehicle. In fault estimation, a change point detection algorithm based on abrupt change model was proposed. Statistical tool was then used to infer the future bound of GPS data, which can detect faults in GPS raw data. If any kinds of faults were detected, dead reckoning mechanism begins to compute current position. Nonholonomic constraint condition of vehicle was used to estimate velocity of vehicle and change point detection was added into classic Kalman filtering structure. Experiment on vehicle shows that even when the GPS signals are unavailable for a period of time, this method can also output high accuracy data.

The prostate secretory protein of 94 amino acids （PSP94） has been shown to interact with cysteine-rich secretory protein 3 （CRISP-3） in human seminal plasma. Interestingly, PSP94 expression is redaced or lost in the majority of the prostate tumours, whereas CRISP-3 expression is upregulated in prostate cancer compared with normal prostate tissue. To obtain a better understanding of the individual roles these proteins have in prostate tumourigenesis and the functional relevance of their interaction, we ectopically expressed either PSP94 or CRISP-3 alone or PSP94 along with CRISP-3 in three prostate cell lines （PC3, WPE1-NB26 and LNCaP） and performed growth inhibition assays. Reverse transcription-polymerase chain reaction and Western blot analysis were used to screen prostate cell lines for PSP94 and CRISP-3 expression. Mammalian expression constructs for human PSP94 and CRISP-3 were also generated and the expression, localization and secretion of recombinant protein were assayed by transfection followed by Western blot analysis and immunofluorescence assay. The effect that ectopic expression of PSP94 or CRISP-3 had on cell growth was studied by clonogenic survival assay follokving transfection. To evaluate the effects of coexpression of the two proteins, stable clones of PC3 that expressed PSP94 were generated. They were subsequently transfected with a CRISP-3 expression construct and subjected to clonogenic survival assay. Our results showed that PSP94 and CRISP-3 could each induce growth inhibition in a cell line specific manner. Although the growth of CRISP-3-positive cell lines was inhibited by PSP94, growth inhibition mediated by CRISP-3 was not affected by the presence or absence of PSP94. This suggests that CRISP-3 may participate in PSP94-independent activities during prostate tumourigenesis.

Insights into erlotinib action in pancreatic cancer cells using a combined experimental and mathematical approach

AIM:To gain insights into the molecular action of erlotinib in pancreatic cancer (PC) cells. METHODS:Two PC cell lines, BxPC-3 and Capan-1, were treated with various concentrations of erlotinib, the specific mitogen-activated protein kinase kinase (MEK) inhibitor U0126, and protein kinase B (AKT) inhibitor XIV. DNA synthesis was measured by 5-bromo-2'-deoxyuridine (BrdU) assays. Expression and phosphorylation of the epidermal growth factor receptor (EGFR) and downstream signaling molecules were quantified by Western blot analysis. The data were processed to calibrate a mathematical model, based on ordinary differential equations, describing the EGFRmediated signal transduction. RESULTS:Erlotinib significantly inhibited BrdU incorporation in BxPC-3 cells at a concentration of 1 mol/L, whereas Capan-1 cells were much more resistant. In both cell lines, MEK inhibitor U0126 and erlotinib attenuated DNA synthesis in a cumulative manner, whereas the AKT pathway-specific inhibitor did not enhance the effects of erlotinib. While basal phosphorylation of EGFR and extracellular signal-regulated kinase (ERK) did not differ much between the two cell lines, BxPC-3 cells displayed a more than five-times higher basal phospho-AKT level than Capan-1 cells. Epidermal growth factor (EGF) at 10 ng/mL induced the phosphorylation of EGFR, AKT and ERK in both cell lines with similar kinetics. In BxPC-3 cells, higher levels of phospho-AKT and phospho-ERK (normalized to the total protein levels) were observed. Independent of the cell line, erlotinib efficiently inhibited phosphorylation of EGFR, AKT and ERK. The mathematical model successfully simulated the experimental findings and provided predictions regarding phosphoprotein levels that could be verified experimentally. CONCLUSION:Our data suggest basal AKT phosphorylation and the degree of EGF-induced activation of AKT and ERK as molecular determinants of erlotinib efficiency in PC cells.
