Morphological and Genetical Variability among Rhizoctonia solani Isolates Causing Sheath Blight Disease of Rice

Eighteen isolates of Rhizoctonia solani collected from infected rice plants in four different locations of Bangladesh were studied by using morphological characters and molecular markers. Anastomosis study with a reference isolate confirmed that all the isolates belonged to R. solani. Significant variation was observed in sclerotial size, shape and distribution. Un-weighted pair group method with arithmetic mean dendrogram constructed based on the Gower＇s general similarity coefficient showed that these isolates were grouped into four clusters at the 0.68 similarity coefficent according to morphological characters. Cluster I was a major cluster consisting of 13 isolates, while clusters Ⅱ to Ⅳ consisted of 1 or 2 isolates. Analyses by variable number of tandem repeat and amplified fragment length polymorphism markers showed that the isolates were grouped into five and three clusters at a similarity coefficient of 0.64 and 0.69, respectively. Although most of the variability was found between isolates from different regions as expected, significant variation was observed within the isolates collected from similar agro-ecological regions. Our results suggest the presence of different races of R. solani within the same local geographic regions.

Postulation of Leaf Rust Resistance Genes in Seven Chinese Spring Wheat Cultivars

To detect the leaf rust resistance genes in the 7 Chinese spring wheat clultivars Shenmian 99025, Shenmia 99042, Shenmian 85, Shenmian 91, Shenmian 96, Shenmian 1167 and Shenmian 962, Thatcher, Thatcher backgrounded near-isogenic lines and 15 pathotypes of P. triticina were used for gene postulate at the seedling stage, and 9 of the 15 pathotypes were used in the field tests. Molecular markers closely linked to, or co-segregated with resistance genes Lrl, Lr9, LrlO, Lrl9, Lr20, Lr21, Lr24, Lr26, Lr28, Lr29, Lr32, Lr34, Lr35, Lr37, Lr38, and Lr47 were screened to assist detection of the resistance genes. As results, 4 known resistance genes, including Lrl, Lr9, Lr26, and Lr34, and other unknown resistance genes were postulated singly or in combination in the tested cultivars. Shenmian 85, Shenmian 91, Shenmian 96, Shenmian 962, Shenmian 1167, and Shenmian 99042 are potentially useful for wheat production and breeding programs. The result suggested that combining gene postulation, molecular markers and pedigrees is effective and more accuracy method to know the resistance genes in cultivars.

Introduction:The brown planthopper(BPH),Nilaparvata lugens(Stål)(Hemiptera:Delphacidae),is a notorious insect pest of *** supplementation can alter plant biochemical compositions which may indirectly affect on ecological fitness parameters of its insect ***,few findings are available on nutrient-induced ecological fitness of BPH and relationship of BPH fitness parameters with its host rice plant biochemical ***:We studied the main and interaction effects of nitrogen(N),phosphorus(P),and potassium(K)inputs on BPH fitness,as measured by the following variables:nymph survival,nymphal duration,adult body weight,and its *** planthopper fitness parameters were regressed as function of rice plant(Oryza sativa)biochemical composition.A completely randomized design with four replications in a factorial scheme was used considering N,P,and K levels as ***:Nitrogen application to the host rice plants provided greater survival and enhanced body weight of *** nymphal duration was found to decrease with N subsidy that caused shorter generation time of *** compounds N and soluble protein(SP),total free sugar(TFS),and silicon(Si)content in rice plant tissue correlated strongly with all fitness traits of ***,SP,and TFS associated positively with BPH survival and body weight,while negatively with nymphal *** contrast,Si had negative correlation with BPH survival and body weight,but positive with nymphal *** supplementation to host plant contributed to increase BPH body *** of P with all parameters of BPH were weak except body weight which was highly significant and *** between N and P inputs demonstrated significant effect on BPH body *** of K had no significant effect on BPH survival and *** analysis did not detect significant relationship of BPH fitness parameters with plant tissue content of ***:The res

Field evaluation of Streptomyces rubrogriseus HDZ-9-47 for biocontrol of Meloidogyne incognita on tomato

Streptomyces rubrogriseus HDZ-9-47,isolated from eggs of Meloidogyne spp.,was evaluated as a potential biocontrol agent of Meloidogyne incognita under in vitro and protective *** observations showed that HDZ-9-47 parasitized eggs of *** within 7 *** vitro,the culture filtrate of HDZ-9-47 caused 97.0%mortality of second-stage juveniles（J2s）of *** and inhibited more than 50%egg *** the field,compared with the control,the rootknot index and J2s density in the treatment of drench the broth contained 1012 HDZ-9-47 spores were respectively reduced by 51.1 and 80.7%at 90 days post transplantation,which were better than that in other application doses and *** addition,reduction rates of root-knot index and J2s density of the treatment of combined application of HDZ-9-47 with biofumigation was 87.1 and 91.0%,respectively,better than either of HDZ-9-47 or biofumigation used alone or fosthiazate *** tomato yield also increased by 16.1%.Together,our results suggest that HDZ-9-47 could be an effective biocontrol agent of ***,and that application of HDZ-9-47 combined with cabbage residue biofumigation was a promising and sustainable option for *** control.

Expression of Ki-67,p53,and K-ras in chronic pancreatitis and pancreatic ductal adenocarcinoma

AIM： To examine surgical specimens of pancreas with either chronic pancreatitis or pancreatic cancer in order to study whether ductal hyperplasia and dysplasia in pancreas represent precursor lesions for pancreatic cancer. METHODS： We examined expression of Ki-67, CEA, p53, and K-ras, in the surgical specimens of pancreas with adenocarcinomas （n = 11） and chronic pancreatitis （n = 12）. Cellular proliferation was assessed by Ki-67 proliferation index using the proliferation marker Ki-67. In specimens with pancreas cancer, we divided pancreas epithelium into normal （n = 7）, ductal hyperplasia （n = 3）, dysplasia （n = 4）, and cancerous lesion （n = 11） after hematoxylin and eosin staining, Ki-67, and CEA immunohistochemical staining. In cases with chronic pancreatitis, the specimen was pathologically examined as in cases with pancreas cancer, and they were also determined as normal （n = 10）, ductal hyperplasia （n = 4）, or dysplasia （n = 5）. p53 and K-ras expression were also studied by immunohistochemical staining. RESULTS： In pancreatic cancer, the Ki-67 index was 3.73±3.58 in normal site, 6.62±4.39 in ductal hyperplasia, 13.47：1：4.02 in dysplasia and 37.03±10.05 in cancer tissue, respectively. Overall, p53 was positive in normal ducts, ductal hyperplasia, dysplasia, and carcinoma cells in 0 of 14 （0%）, 0 of 7 （0%）, 7 of 9 （78%）, and 10 of 11 （91%）, respectively, and K-ras was positive in 0 of 8 （0%）, 1 of 3 （33%）, 4 of 6 （67%）, 4 of 5 （80%）, respectively. CONCLUSION： Our results favorably support the hypothesis that ductal hyperplasia and dysplasia of the pancreas might be precursor lesions for pancreas cancer. Further evaluation of oncogenes by the molecular study is needed.

Parasitism and pathogenicity of Radopholus similis to Ipomoea aquatica, Basella rubra and Cucurbita moschata and genetic diversity of different populations

Ten populations of Radopholus similis from different ornamental hosts were tested for their parasitism and pathogenicity to water spinach （Ipomoea aquatic）, malabar spinach （Basella rubra）, and squash （Cucurbita moschata） in pots. The re- sults showed all three plants were new hosts of R. similis. Growth parameters of plants inoculated with nematodes were significantly lower than those of healthy control plants. All R. similis populations were pathogenic to the three plants, but pathogenicity differed among populations from different hosts. The same R. similis populations also showed different pathogenic effects in the three different plants. RadN5 population from Anthurium andraeanum had the highest pathoge- nicity to the three studied plants. RadN1 from A. andraeanum had the lowest pathogenicity to squash and RadN7 from Chrysalidocarpus lutesens had the lowest pathogenicity to water spinach and malabar spinach. R. similis is usually associated with root tissues, but here we report that it could be found to move and feed in the stem bases of all three studied plants. Sequence and phylogenetic analyses of DNA markers of the 18S rRNA, 28S rRNA, ITS rRNA, and mitochondrial DNA gene sequences of ten R. similis populations revealed significant genetic diversity. RadN5 and RadN6 populations from anthurium showed a close genetic relationship and could be distinguished from other populations by PCR-RFLP. At the same time, RadN5 and RadN6 populations were the most pathogenic to three studied plants. These results confirm the existence of large biological variability and molecular diversity among R. similis populations from the same or different hosts, and these characteristics are related to pathogenic variability.

Strong Suppression of Systemic Acquired Resistance in Arabidopsis by NRR is Dependent on its Ability to Interact with NPRland its Putative Repression Domain

Systemic Acquired Resistance （SAR） in plants confers lasting broad-spectrum resistance to pathogens and requires the phytohormone salicylic acid （SA）. Arabidopsis NPR1/NIM1 is a key regulator of the SAR response. Studies attempting to reveal the function of NPR1 and how it mediates SA signaling have led to isolation of two classes of proteins that interact with NPRI＂ the first class includes rice NRR, Arabidopsis NIMIN1, NIMIN2, and NIMIN3, and tobacco NIMIN2-1ike proteins; the second class belongs to TGA transcription factors. We have previously shown that overexpression of N RR in rice suppresses both basal and Xa21-mediated resistance. In order to test whether NRR affects SA-induced, NPRl-mediated SAR, we have transformed Arabidopsis with the rice NRR gene and tested its effects on the defense response. Expression of NRR in Arabidopsis results in suppression of PR gene induction by SAR inducer and resistance to pathogens. These phenotypes are even more severe than those of the nprl-1 mutant. The ability of NRR to suppress PR gene induction and disease resistance is correlated with its ability to bind to NPR1 because two point mutations in NRR, which reduce NPR1 binding, fail to suppress NPR1. In contrast, wild-type and a mutant NRR, which still binds to NPR1 strongly, retain the ability to suppress the SAR response. Replacing the C-terminal 79 amino acids of NRR with the VP16 activation domain turns the fusion protein into a transcriptional co-activator. These results indicate that NRR binds to NPR1 in vivo in a protein complex to inhibit transcriptional activation of PR genes and that NRR contains a transcription repression domain for active repression.

This review focuses on the relationship between hepatitis C virus(HCV) infection and glucose metabolism ***-sectional and longitudinal studies have shown that the chronic HCV infection is associated with an increased risk of developing insulin resistance(IR) and type 2 diabetes(T2D).The direct effect of HCV on the insulin signaling has been analyzed in experimental *** currently available data should be considered as preliminary,HCV seems to affect glucose metabolism via mechanisms that involve cellular pathways that have been implicated in the host innate immune *** and T2D not only accelerate the histological and clinical progression of chronic hepatitis C,but also reduce the early and sustained virological response to interferon-alpha-based ***,a detailed knowledge of the mechanisms underlying the HCV-associated glucose metabolism derangements is warranted,in order to improve the clinical management of chronic hepatitis C patients.

Carcinoid tumor of the duodenum and accessory papilla associated with polycythemia vera

Carcinoid tumors have been reported in a wide range of organs but most frequently involve the gastrointestinal tract; however, duodenal carcinoid tumors are rare. We report a 50-year-old male patient complaining of multiple melenas for 3 wk. The panendoscopy and endoscopic retrograde cholangiopancreaticography revealed swelling accessory papilla with an ulcer. The biopsy taken showed a carcinoid tumor. The lesion was removed by wide resection. Patient was found to have an abnormal blood cell count during the follow-up period with elevated levels of hemoglobin and hematocrit of 21.2 g/dL and 63.5%,respectively, thrombocytosis of 501 000/μL, and leukocytosis of 20 410/μL. He was diagnosed as a polycythemia vera by a hematologist after further evaluation. He received periodic phlebotomy and hydroxyurea treatment. The response was good and his hematocrit was stabilized by periodic phlebotomy in the range of 44-49% during the last 2 years. The possible origin of UGI bleeding by a duodenal carcinoid tumor, although rare, should be considered. There has been one case report of a duodenal carcinoid tumor that involved accessory papilla of the pancreas divisum and one case report of metastatic carcinoid tumor associated with polycythemia vera. It is different in our patient as compared with the latter report, which mentioned a polycythemia vera patient who was found to have a metastatic carcinoid in the 17 years follow-up period. Chemotherapy had been given before the carcinoid tumor was revealed. Our patient had no previous chemotherapy for polycythemia vera before he was found to have duodenal carcinoid tumor; this excludes the possibility of chemotherapy induced carcinoid tumor, although it had been suspected in the previous report. In our patient, the existence of both diseases may be by predisposition of each other since both diseases have an increased incidence of other neoplasm, or they may be coexistent incidentally.

Isolation and identification of Serratia marcescens Ha1 and herbicidal activity of Ha1 ‘pesta’ granular formulation

A total of 479 bacterial strains were isolated from brine （Bohai, Qinhuangdao City, Hebei Province, China）. Bioassay results indicated that 4 strains named Hal, Hal7, Ha38, and Ha384 had herbicidal activity. And strain Hal had the highest effective herbicidal activity. As a result, this study aims to JdentJfy strain Hal, characterize its physiological and biological activities, evaluate the herbicidal activity of its metabolites, and develop a ＇pesta＇ formulation and assess its effectiveness on Digitaria sanguinalis. Hal was identified as Serratia marcescens based on 16S rDNA sequencing. This strain has a flagellum, a diameter of 0.5 to 0.8 IJm, and a length of 0.9 to 2.0 IJm. The indole test shows positive results, and the catalase enzyme exhibits strong positive reactions. Results further showed that the inhibitory concentration （IC50） of the crude extracts to D. sanguinalis radicula and coleoptile were 3.332 and 2.828 mg mL-1, respectively. Both the suppression of D. sanguinalis and the cell viability of the Hal formulation in ＇pesta＇ were higher when stored at 4℃ than at （25＋2）℃. These results indi- cated that S. marcescens Hal can potentially be used as a biocontrol agent against D. sanguinalis.
