Dear Editor,Inflammatory bowel diseases(IBD)such as ulcerative colitis(UC)and Crohn's disease are affecting increasing numbers of patients worldwide.1 These chronic diseases significantly impact patients'quality of life as they are incurable and disabling.1 Patients with IBD have had an increasing rate of hospitalizations,half of all UC patients are hospitalized at least once during their lifetime,and readmissions after a UC-related hospitalization are common.

Individuals with inflammatory bowel disease(IBD)have been reported to be at an increased risk of infertility and sexual dysfunction.Although the relationship between them remains unclear,IBD severity is suspected to affect hormone levels and fertility.To analyze the impact of IBD severity on semen parameters and sex hormone levels in ulcerative colitis-type IBD(UC-IBD),we conducted a cross-sectional study involving 120 patients with UC-IBD in Adam Malik General Hospital,Medan,Indonesia.The patients were classified into three groups based on the Mayo score for Uc,followed by a comparison of various semen and hormone parameters among these groups.In addition to the cross-sectional analysis,a simple correlation test was conducted irrespective of the patient grouping.Sperm concentration,motility,and morphology were found to decline significantly with an increase in IBD severity.Without classifying patients with IBD into subgroups,the Mayo score showed negative correlations with sperm concentration(r=-0.375,P<0.0001),rapid progressive motility(r=-0.660,P<0.0001),free testosterone(r=-0.732,P<0.0001),and total testosterone(r=-0.721,P<0.0001),and positivecorrelations with immotile sperm(r=0.660,P<0.0001),abnormal morphology(r=0.657,P<0.0001),and sex hormone-binding globulin(SHBG;r=0.278,P=0.002).Sperm concentration,motility,and morphology declined significantly with the severity of IBD.This study suggests a significant negative impact of IBD severityonsemenqualityand sexhormones.

Chuanxiong Rhizoma extracts prevent liver fibrosis via targeting CTCF-c-MYC-H19 pathway

Objective:Hepatic fibrosis has been widely considered as a conjoint consequence of almost all chronic liver diseases.Chuanxiong Rhizoma(Chuanxiong in Chinese,CX)is a traditional Chinese herbal product to prevent cerebrovascular,gynecologic and hepatic diseases.Our previous study found that CX extracts significantly reduced collagen contraction force of hepatic stellate cells(HSCs).Here,this study aimed to compare the protection of different CX extracts on bile duct ligation(BDL)-induced liver fibrosis and investigate plausible underlying mechanisms.Methods:The active compounds of CX extracts were identified by high performance liquid chromatography(HPLC).Network pharmacology was used to determine potential targets of CX against hepatic fibrosis.Bile duct hyperplasia and liver fibrosis were evaluated by serologic testing and histopathological evaluation.The expression of targets of interest was determined by quantitative real-time PCR(qPCR)and Western blot.Results:Different CX extracts were identified by tetramethylpyrazine,ferulic acid and senkyunolide A.Based on the network pharmacological analysis,42 overlap targets were obtained via merging the candidates targets of CX and liver fibrosis.Different aqueous,alkaloid and phthalide extracts of CX(CX_(AE),CX_(AL) and CXP_(HL))significantly inhibited diffuse severe bile duct hyperplasia and thus suppressed hepatic fibrosis by decreasing CCCTC binding factor(CTCF)-c-MYC-long non-coding RNA H19(H19)pathway in the BDL-induced mouse model.Meanwhile,CX extracts,especially CX_(AL) and CX_(PHL) also suppressed CTCF-c-MYC-H19 pathway and inhibited ductular reaction in cholangiocytes stimulated with taurocholate acid(TCA),lithocholic acid(LCA)and transforming growth factor beta(TGF-β),as illustrated by decreased bile duct proliferation markers.Conclusion:Our data supported that different CX extracts,especially CX_(AL) and CX_(PHL) significantly alleviated hepatic fibrosis and bile duct hyperplasia via inhibiting CTCF-c-MYC-H19 pathway,providing novel insights into the anti-f

Integrins and their potential roles in mammalian pregnancy

Integrins are a highly complex family of receptors that, when expressed on the surface of cells, can mediate reciprocal cell-to-cell and cell-to-extracellular matrix(ECM) interactions leading to assembly of integrin adhesion complexes(IACs) that initiate many signaling functions both at the membrane and deeper within the cytoplasm to coordinate processes including cell adhesion, migration, proliferation, survival, differentiation, and metabolism. All metazoan organisms possess integrins, and it is generally agreed that integrins were associated with the evolution of multicellularity, being essential for the association of cells with their neighbors and surroundings, during embryonic development and many aspects of cellular and molecular biology. Integrins have important roles in many aspects of embryonic development, normal physiology, and disease processes with a multitude of functions discovered and elucidated for integrins that directly influence many areas of biology and medicine, including mammalian pregnancy, in particular implantation of the blastocyst to the uterine wall, subsequent placentation and conceptus(embryo/fetus and associated placental membranes) development. This review provides a succinct overview of integrin structure, ligand binding, and signaling followed with a concise overview of embryonic development, implantation, and early placentation in pigs, sheep, humans, and mice as an example for rodents. A brief timeline of the initial localization of integrin subunits to the uterine luminal epithelium(LE) and conceptus trophoblast is then presented, followed by sequential summaries of integrin expression and function during gestation in pigs, sheep, humans, and rodents. As appropriate for this journal, summaries of integrin expression and function during gestation in pigs and sheep are in depth, whereas summaries for humans and rodents are brief. Background Calcifying pseudoneoplasm of the neuraxis(CAPNON)is indeed a rare central nervous system lesion that can occur in central nervous system(CNS).Due to its infrequency and limited literature reports,it is challenging to diagnose and manage CAPNON.Case presentation In this intriguing study,we embarked on a quest to uncover the story of a 16-year-old girl who experienced bothersome headaches.Through advanced imaging techniques like computed tomography(CT)and magnetic resonance imaging(MRI),we glimpsed a delicate calcified growth within the lateral ventricles’posterior horn.Motivated by our unwavering commitment to solving mysteries,we embarked on a surgical journey that not only freed the young patient from her ailment but also shed light on the true nature of her puzzling adversary—a remarkable CAPNON.Conclusions For patients with CAPNON who have multiple or non-respectable lesions,the primary goal is to alleviate symptoms.After alleviating the symptoms with partial resection,close monitoring of any residual lesions is essential.If there is no evidence for disease progression,a strategy of continued close observation is appropriate.

Effects of host identity on the gut microbiota:A comparative study on three microtinae species

Background:Gut microbiota exert an immense effect on host health and host environmental adaptation.Furthermore,the composition and structure of gut microbiota are determined by the environment and host genetic factors.However,the relative contribution of the environment and host genetic factors toward shaping the structure of gut microbiota has been poorly understood.Methods:In this study,we characterized the fecal microbial communities of the closely related voles Neodon fuscus,Lasiopodomys brandtii,and L.mandarinus after caged feeding in the laboratory for 6 months,through high-throughput sequencing and bioinformatics analysis.Results:The results of pairwise comparisons of N.fuscus vs.L.brandtii and L.mandarinus vs.L.brandtii revealed significant differences in bacterial diversity and composition after domestication.While 991 same operational taxonomic units(OTUs)were shared in three voles,there were 362,291,and 303 species-specific OTUs in N.fuscus,L.brandtii,and L.mandarinus,respectively.The relative abundances of Proteobacteria and Prevotella,which are reported to be enriched in high-altitude populations,were significantly higher in high-altitude N.fuscus than in low-altitude L.brandtii after domestication.Firmicutes,which produce various digestive enzymes for energy metabolism,and Spirochaetes,which can degrade cellulose,were found in higher abundance in subterranean L.mandarinus than that in L.brandtii which dwells on the earth surface.Conclusion:Our findings showed that some components of gut microbiota still maintained dominance even when different host species are reared under the same environmental conditions,suggesting that these bacteria are substantially influenced by host factors.

The Anxiolytic and Antidepressant Effect of Stachys Lavandulifolia in the Experimental Pentylenetetrazole-Induced Rat Model of Acute Seizure

Objective:In traditional medicine,Stachys lavandulifolia or mountain tea has been used in the treatment of arthritis and rheumatoid arthritis.A leaf extract from this plant is also thought to be effective in treating epilepsy as well as other neurological ailments.To assess the effect of S.lavandulifolia extract(SLE),rats were exposed to a stress and anxiety model caused by seizures.Materials and Methods:A challenge dose of pentylenetetrazole(PTZ)(60 mg/kg)was used to induce seizures in male Wistar rats(200-250 g).Animal treated with an intraperitoneal injection of either SLE(50 mg/kg)or sodium valproate(100 mg/kg)alone or in combination 30 min before PTZ challenge.The effect of stress and anxiety was assessed using an open field test(OFT),forced swim test(FST),and elevated plus maze(EPM).At the end of the experiment,anesthesia was used to euthanize rats,and their blood serum was collected for measurements of glutathione(GSH),malondialdehyde(MDA),and GSH peroxidase(GPx).Results:In contrast to the control group,those treated with PTZ exhibited depressive behavior.At 50 mg/kg,SLE provided protection against seizures induced by PTZ.SLE also reduced seizure-induced stress and anxiety,significantly improving FST,EPM,and OFT compared with the PTZ-treated group.By decreasing MDA levels and restoring the antioxidant enzymes GSH and GPx,SLE treatment protects against seizure-induced anxiety and stress.Conclusions:SLE inhibits oxidative stress damage and seizure-induced stress by inhibiting seizures'inhibitory effects.

Retinitis pigmentosa(RP)is a group of genetic disorders characterized by progressive degeneration of photoreceptors and retinal pigment epithelium(RPE)cells.Its main clinical manifestations include night blindness and progressive loss of peripheral vision,making it a prevalent debilitating eye disease that significantly impacts patients’quality of life.RP exhibits significant phenotypic and genetic heterogeneity.For instance,numerous abnormal genes are implicated in RP,resulting in varying clinical presentations,disease progression rates,and pathological characteristics among different patients.Consequently,gene therapy for RP poses challenges due to these complexities.However,stem cells have garnered considerable attention in the field of RPE therapy since both RPE cells and photoreceptors can be derived from stem cells.In recent years,a large number of animal experiments and clinical trials based on stem cell transplantation attempts,especially cord blood mesenchymal stem cell(MSC)transplantation and bone marrow-derived MSC transplantation,have confirmed that stem cell therapy can effectively and safely improve the outer retinal function of the RP-affected eye.However,stem cell therapy also has certain limitations,such as the fact that RP patients may involve multiple types of retinal cytopathia,which brings great challenges to stem cell transplantation therapy,and further research is needed to solve various problems faced by this approach in the clinic.Through comprehensive analysis of the etiology and histopathological changes associated with RP,this study substantiates the efficacy and safety of stem cell therapy based on rigorous animal experimentation and clinical trials,while also highlighting the existing limitations that warrant further investigation.
