Effect of environmental stresses on the survival and cytotoxicity of Shiga toxin-producing Escherichia coli

The objective of this study was to determine the effect of environmental stresses on the cytotoxicity of Shiga toxin-producing Escherichia coli (STEC). STEC O157:H7 and six non-O157 STEC strains (O26:H11, O103:H1, O104:H4, O111:NM, O121:NM, and O145:NM) were subjected to osmotic (aw 0.95-0.98), acid (pH 4-7), and chlorine (1-5 ppm) stresses. After stress treatments, bacterial populations, expression of virulence-associated genes, and Vero-cytotoxicity were determined. Among the strains, O145:NM survived at aw 0.97 longer than other serotypes, while O111:NM was significantly more sensitive to osmotic stress. At pH 4, O103:H1 was more resistant to the stress, while O26:H11 and O111:NM had significantly less growth. For 2 ppm chlorine stress, O26:H11, O103:H1, and O145:NM had higher populations (>3 log) than other strains. Stressed strains showed a significant increase in relative gene expression levels of stx1, stx2, and eae in O103:H1, O104:H4, and O145:NM than non-stressed control. Additionally, significantly higher Vero-cytotoxicity, as indicated by lactate dehydrogenase assay, of stressed O26:H11, O103:H1, O104:H4, and O145:NM was observed. The results suggest that the growth and cytotoxicity of selected pathogenic E. coli may be enhanced after being exposed to environmental stresses.

Objectives: The occurrence of Cryptosporidium oocysts in drinking source water can present a serious public health risk. To rapidly and effectively assess the source and human-infective potential of Cryptosporidium oocysts in water, sensitive detection and correct identification of oocysts to the species level(genotyping) are essential. Thus we developed three real-time PCR genotyping assays for rapid tracking of Cryptosporidium contamination. Methods: Light Cycler PCR and melt curve analysis using fluorescence resonance energy transfer(FRET) probes were used in the development of three real-time PCR assays, two targeting the small subunit(SSU) rRNA gene(18S-LC1 and 18S-LC2 assays) and one targeting the 90 k Da heat shock protein(hsp90) gene(hsp90 assay). We evaluated the sensitivity and Cryptosporidium species detection range of these assays. Results: The 18S-LC1 and hsp90 assay could differentiate common human-pathogenic species(C. parvum, C. hominis, and C. meleagridis), while the 18S-LC2 assay was able to differentiate non-pathogenic species(such as C. andersoni) from human-pathogenic ones commonly found in source water. In sensitivity evaluations, the 18S-LC2 and hsp90 genotyping assays could detect as few as one Cryptosporidium oocyst per sample, indicating it is possible for these two assays to detect low levels of oocysts in water samples. Conclusion: The 18S-LC2 and hsp90 genotyping assays might be used in environmental monitoring, whereas the 18S-LC1 genotyping assay could be useful for genotyping Cryptosporidium spp. in clinical specimens or wastewater samples.

In vitro meat production: Challenges and benefits over conventional meat production

Although initially described as Cryptosporidium parvum,the Cryptosporidium in the small intestine of mice(Mus musculus)differs genetically from what is now known as *** in humans and *** this study,the morphological,biological,and genetic characteristics of the murine parasite are described,and the parasite is renamed as Cryptosporidium *** of the new species(4.64±0.05×4.19±0.06μm,with a mean shape index of 1.11±0.02;n=69)are slightly smaller than those of Cryptosporidium *** prepatent period was six and seven days,and the patent period was 24-28 and 28-29 days in neonatal and adult mice,*** are not infectious to lambs and ***,transmission electron and scanning electron microscopy studies of the new species showed the presence of developmental stages in the microvillar brush border of the jejunum and ileum of experimentally infected mice,with the infection most intensive in the *** the parasite was probably initially described as ***,it had nucleotide sequences significantly different from what is now known as *** at the small subunit r RNA,70 k Da heat shock protein,oocyst wall protein,actin,and the 60 k Da glycoprotein genes,and was widely known as the Cryptosporidium mouse genotype or mouse genotype *** on the morphological,genetic and biological data and in compliance of established Cryptosporidium species naming criteria,this geographically widespread parasite is named as a new species in honor of Ernest Edward Tyzzer,who pioneered Cryptosporidium research and probably first detected the parasite near 100 years ago.

In vitro meat production system is the production of meat outside the food animals by culturing the stem cells derived from farm animals inside the bioreactor by using advanced tissue engineering techniques. Besides winning the favour of animal rights activists for its humane production of meat, in vitro meat production system also circumvents many of the issues associated with conventional meat production systems, like excessively brutal slaughter of food animals, nutrition-related diseases, foodborne illnesses, resource use, antibiotic-resistant pathogen strains, and massive emissions of methane that contribute to global warming. As the conditions in an in vitro meat production system are controlled and manipulatable, it will be feasible to produce designer, chemically safe and disease-free meat on sustainable basis. However, many challenges are to be faced before cultured meat becomes commercially feasible. Although, the production cost and the public acceptance are of paramount importance, huge funds are desperately required for further research in the field.

Background Despite the increasing focus on strengthening One Health capacity building on global level, challenges remain in devising and implementing real-world interventions particularly in the Asia-Pacific region. Recognizing these gaps, the One Health Action Commission (OHAC) was established as an academic community for One Health action with an emphasis on research agenda setting to identify actions for highest *** text This viewpoint describes the agenda of, and motivation for, the recently formed OHAC.
