Thymosin β4 promotes angiogenesis and increases muscle progenitor cell density in ischemic skeletal muscle in a mouse model of hind limb ischemia

Aim: To determine the therapeutic effect of thy- mosin β4 (Tβ4) for treatment of ischemic limb disease in a mouse model. Methods: A mouse model of hindlimb ischemia was created by permanent ligation of femoral arteries and internal iliac artery. Tβ4 was dissolved in sterile saline and intramuscularly injected into the centre and periphery of ligation area in the treatment group (n = 10) starting from the surgery day until 4 weeks after surgery, while control animals received saline injection only (n = 9). All animals were sacrificed at 6 weeks after surgery and used for immunohistochemistry studies. Results: Tβ4 stimulated angiogenesis was evidenced by increased vascular density based on CD31 immunostaining, which was sig- nifycantly increased in Tβ4 group (562.5 ± 78.4/mm2) as compared with control group (371.1 ± 125.7/mm2;p 0.05) groups. Tβ4 increased Pax3/7+ skeletal muscle progenitor cell density. Pax3/7+ cell density of Tβ4 group (13.7% ± 2%) was significantly higher than that of the control group (4.3% ± 1.6%, p < 0.05). However, the numbers of central nuclei fiber and central nuclei per fiber were insignificantly increased in Tβ4 group as compared to control group. The numbers of central nuclei fiber were 8.9 ± 2.1 and 9.5 ± 1.6, and the central nuclei per fiber were 0.25 ± 0.07 and 0.48 ± 0.09 for control and Tβ4 groups, respectively. Conclusions: This preliminary study suggests that localized delivery of Tβ4 increased angiogenesis and skeletal muscle progenitor cell density in ischemic skeletal muscle, but failed to promote skeletal muscle regeneration.

Due to rapid Industrialization and economic growth, engineering and technical education has been fast developing in the world. Engineering and Technical Education plays a vital role in human resource development of the country by creating skilled manpower, enhancing Industrial productivity and improving the quality of life. In the current scenario, technical education determines the development and socio- economic condition of the nation. There is a greater need for quality technical education to produce technically skilled manpower. Although engineering education has evolved in many ways that improve the readiness of graduates to meet the challenges of the twenty-first century, National and International organizations continue to seek for change. In response for the change from National and International organizations, engineering education has evolved in a number of ways that enhance the learning of students. One of the major responses has been the incorporation of more team-based, project-driven activities. One of the methods to prepare the students to meet these challenges is help them to build knowledge and skills which they can readily adapt to address the novel, complex problems that they come across. Downregulating activated epidermal growth factor receptor has no effect on RBM5 expression

Background We were interested in determining how the tumor suppressor gene RBM5 is regulated in lung cancers. Previous studies suggested that the gene expression is related to histological subtype and smoking exposure, since in small cell lung cancers the RBM5 gene is deleted whereas in non-small cell lung carcinomas （NSCLC） RBM5 expression is reduced. Of particular interest was the recent finding that in lung adenocarcinomas, a histological subtype of NSCLC, smoking exposure correlated with mutational activity in the transforming growth factor alpha （TGF-c~） signaling pathway. Lung adenocarcinomas from smokers were associated with activating KRAS mutations, whereas lung adenocarcinomas from never-smokers were associated with activating epidermal growth factor receptor （EGFR） mutations. We hypothesized that inhibition of RBM5 in lung adenocarcinomas is achieved indirectly via these activating mutations. The objective of the research described herein was to determine if EGFR activation and RBM5 expression are negatively correlated. Methods EGFR expression in the lung adenocarcinoma cell line NCI-H1975 was inhibited using small interfering RNA. RBM5 expression was examined by real-time quantitative polymerase chain reaction and Western blotting. Results Reduced EGFR expression did not correlate with any change in RBM5 expression at either the RNA or protein level. Conclusion These results suggest that RBM5 expression is not directly regulated by EGFR in non-smoker related lung adenocarinomas, and that some other mechanism operates to inhibit either the expression or function of this potential tumour suppressor in lung cancers that retain the RBM5 gene.

Anatomic Evaluation of Uterosacral and Cardinal Ligament during Robotic and Laparoscopic Surgery for Pelvic Organ Prolapse

Introduction: It is widely accepted that the uterosacral ligaments (UTSL), together with the cardinal ligament (CL), hold the upper vagina and cervix over the levator plate. The aim of this study is to evaluate the anatomical relationship between the right vs. left CL and UTSL during robotic and laparoscopic utero-sacral ligament suspension (UTSLS) and its implication with the surgical technique during UTSL suspension. Material and Methods: We evaluated 25 women with apical pelvic organ prolapses of stages 2 - 4 and we analyzed 100 uterosacral and cardinal ligaments. They were assigned (non-randomly) to: a) robotic-assisted laparoscopic uterosacral ligament suspension (RAL-UTSLS), b) robotic-assisted single-site utero-sacral ligament suspension (RASS-UTSLS) or laparo-endoscopic single site utero-sacral ligament suspension (LESS-UTSLS). We evaluated the length (distance between origins and insertions) of the aforementioned ligaments using the Da Vinci Si and other laparoscopic instruments like calipers. Results: The mean length of the UTSL in their caudal-cranial extent was 3.5 ± 0.5 cm (right side) and 2.58 ± 0.3 cm (left side). Measurements were performed on the same way for the CL, resulting in 5.1 ± 0.3 cm (both side). The only significant difference was observed when comparing the right vs. left UTSL. This anatomic difference translates to 5 ± 1 suture stitches on the right UTSL vs. 2 ± 1 on the left UTSL. Conclusion: In our evaluation on cardinal and uterosacral ligament, the right UTSL was significantly longer as compared to the left and this allowed us to take 3 additional stitches on the right UTSL vs. left during RAL-UTSLS. Future studies are necessary to compare females with/without POP.

The Historical Review of the 1755 Lisbon Tsunami

The original accounts reporting the 1755 Lisbon Tsunami were compiled, focusing on the descriptions related to the tsunami parameters, damage and fatalities in the affected regions （Atlantic northeast and Caribbean）. The accounts show the tsunami reached the southwest municipalities of Portugal in less than 30 minutes after the earthquake, and about one hour later the entire coasts of Portugal, Gulf of Cadiz （Spain） and Morocco were hit by the first tsunami waves. The tsunami took about five hours to reach Ireland and UK, and hit the Caribbean region about 9-10 hours after the earthquake. In addition, significant damage and fatalities were reported in Portugal （Peniche, the Lisbon Metropolitan Area and seven coastal areas in the south）, Spain （Cadiz） and in all Morocco coastline, including Marrakech which is located about 200 km inland. In Ireland, UK and the Caribbean minor damage was observed, and no fatalities were reported.

Effect of Different Reproductive Stages and Culture Times on Domestic Cat <i>in Vitro </i>Oocyte Maturation

The domestic cat has been used as a model to carry out comparative research in assisted reproduction, to be applied in wild cats. The efficiency in domestic cat IVM concerning the reproductive status and/or cultivation times has previously been investigated;however, the studies were carried out separately. The objective of this research was to evaluate the maturation of oocytes of domestic cats of different reproductive stages using two different in vitro culture times. The ovaries were obtained by Ooforo-Salpingo-Hysterectomy of cats that were of the following groups: 1) prepubertal, 2) follicular, 3) pregnant or 4) in anestrus. Maturation was carried out with TCM199 medium supplemented with BSA for 24 h and 48 h. On average, 29 ± 25, 20 ± 15, 17 ± 9 and 17 ± 13 oocytes/cat were recovered from the prepubertal follicular, pregnant, and anestrus stages, respectively, but did not show a significant difference (P > 0.05). Also, meiotic maturation did not show a significant difference between the different reproductive stages at 24 h and 48 h, respectively (P > 0.05). However, in the prepubertal and follicular stages, greater oocyte maturation numbers were observed at 48 h compared to 24 h (P result was not observed in the pregnant and anestrus stages (P > 0.05), indicating that the in vitro culture duration is an important factor during in vitro maturation of domestic cat oocytes.
