Ethanol(EtOH) is a common trigger for gastric mucosal diseases, and mitigating oxidative stress is essential for attenuating gastric mucosal damage. Capsaicin(CAP) has been identified as a potential agent to counteract oxidative damage in the gastric mucosa; however, its precise mechanism remains unclear. This study demonstrates that CAP alleviates EtOH-induced gastric mucosal injuries through two primary pathways: by suppressing the chemokine receptor 4(CCR4)/Src/p47phox axis, thereby reducing oxidative stress, and by inhibiting the phosphorylation and nuclear translocation of nuclear factor-κB p65(NF-κB) p65, resulting in diminished inflammatory responses. These findings elucidate the mechanistic pathways of CAP and provide a theoretical foundation for its potential therapeutic application in the treatment of gastric mucosal injuries.

General expert consensus on the application of network pharmacology in the research and development of new traditional Chinese medicine drugs

The research and development of new traditional Chinese medicine(TCM) drugs have progressively established a novel system founded on the integration of TCM theory, human experience, and clinical trials(termed the “Three Combinations”). However, considering TCM's distinctive features of “syndrome differentiation and treatment” and “multicomponent formulations and complex mechanisms”, current TCM drug development faces challenges such as insufficient understanding of the material basis and the overall mechanism of action and an incomplete evidence chain system. Moreover, significant obstacles persist in gathering human experience data, evaluating clinical efficacy, and controlling the quality of active ingredients, which impede the innovation process in TCM drug development. Network pharmacology, centered on the “network targets” theory, transcends the limitations of the conventional“single target” reductionist research model. It emphasizes the comprehensive effects of disease or syndrome biological networks as targets to elucidate the overall regulatory mechanism of TCM prescriptions. This approach aligns with the holistic perspective of TCM, offering a novel method consistent with TCM's holistic view for investigating the complex mechanisms of TCM and developing new TCM drugs. It is internationally recognized as a “next-generation drug research model”. To advance the research of new tools, methods, and standards for TCM evaluation and to overcome fundamental, critical, and cutting-edge technical challenges in TCM regulation, this consensus aims to explore the characteristics, progress, challenges, applicable pathways, and specific applications of network pharmacology as a new theory, method, and tool in TCM drug development. The goal is to enhance the quality of TCM drug research and development and accelerate the efficiency of developing new TCM products.

Recent advances in spatially resolved transcriptomics(SRT)have provided new opportunities for characterizing spatial structures of various ***-based geometric deep learning has gained widespread adoption for spatial domain identification ***,most methods define adjacency relation between cells or spots by their spatial distance in SRT data,which overlooks key biological interactions like gene expression similarities,and leads to inaccuracies in spatial domain *** tackle this challenge,we propose a novel method,SpaGRA(https://***/sunxue-yy/SpaGRA),for automatic multi-relationship construction based on graph *** uses spatial distance as prior knowledge and dynamically adjusts edge weights with multi-head graph attention networks(GATs).This helps SpaGRA to uncover diverse node relationships and enhance message passing in geometric contrastive ***,SpaGRA uses these multi-view relationships to construct negative samples,addressing sampling bias posed by random *** results show that SpaGRA presents superior domain identification performance on multiple datasets generated from different *** SpaGRA,we analyze the functional regions in the mouse hypothalamus,identify key genes related to heart development in mouse embryos,and observe cancer-associated fibroblasts enveloping cancer cells in the latest Visium HD ***,SpaGRA can effectively characterize spatial structures across diverse SRT datasets.

Many proteins are regulated by post-translational modifications,such as the reversible covalent attachment of ubiquitin and ubiquitin-like proteins in eukaryotes(Kerscher et al, 2006). Post-translational modification of proteins by the SUMO protein family is involved in diverse cellular processes, including development, hormonal responses, and biotic and abiotic stress signaling(Park et al, 2011). SUMO modification can modulate protein-protein interactions, intracellular localization or the activities of the protein(Gareau and Lima, 2010).

Rice E3 Ligase-Like Protein OsPIAL1 Positively Regulated the Drought Stress Response but Negatively Regulated the Salt Stress Response

Small ubiquitin-like modifier(SUMO)E3 ligases that facilitate the conjugation of SUMO proteins to target substrates contain an SP-RING domain which is like the RING domain found in ubiquitin E3 *** this study,we isolated and characterized the Oryza sativa protein inhibitor of activated STAT like1(OsPIAL1)containing SP-RING domains,as the rice homolog of Arabidopsis ***1 interacts with OsSUMO proteins but does not interact with rice SUMO-conjugating enzymes(OsSCEs).An analysis of transgenic rice plant shows that OsPIAL1 is involved in SUMO conjugation to SCEs but not in SUMO conjugation to *** addition,this OsPIAL1 activity requires drought stress *** profiles show that the OsPIAL1 gene is induced by only drought stress in the leaves,whereas it is repressed by ABA and abiotic stresses in the *** stress leads to the fastest decrease in OsPIAL1 transcripts in the ***,the stress experiments indicate that the transgenic rice plants overexpressing OsPIAL1 exhibit a drought stress-tolerant phenotype but a salt stress hypersensitive *** results and those from Arabidopsis pial mutants suggest that PIALs act as a positive regulator in the drought stress response but as a negative regulator in the salt stress response.

Monascus pilosus-fermented black soybean inhibits lipid accumulation in adipocytes and in high-fat diet-induced obese mice

Objective:To explore the anti-obesity effects and the mechanism of action of Monascus pilosus(***)-fermented black soybean(MFBS)extracts(MFBSE)and MFBS powders(MFBSP)in adipocytes and high-fat diet(HFD)-induced obese mice,***:Black soybean was fermented with ***,and the main constituents in MFBS were analyzed by HPLC *** vitro,MFBSE were examined for anti-adipogenic effects using Oil-Red O *** vivo,mice were fed a normal-fat diet(NFD)control,HFD control or HFD containing 1 g/kg MFBSP for 12 weeks,and then body weight gain and tissues weight ***-time PCR and western blot assay were used to determine the mechanism of anti-adipogenic ***:MFBSE inhibited lipid accumulation in 3T3-L1 adipocytes without exerting cell *** treatment in HFD-fed mice significantly decreased the body weight gain compared with the HFD control *** and MFBSP treatment resulted in significantly lower mRNA levels of adipogenesis-related genes,such as peroxisome proliferator-activated receptorγ(PPARγ),fatty acid-binding protein 4(FABP4),and fatty acid synthase(FAS),in adipocytes and in white adipose tissue(WAT)of HFD-induced obese ***:These results suggest that the anti-obesity effects of MFBS are elicited by regulating the expression of adipogenesis-related genes in adipocytes and WAT of HFDinduced obese mice.

Feature Rescaling of Support Vector Machines

Support vector machines （SVMs） have widespread use in various classification problems. Although SVMs are often used as an off-the-shelf tool, there are still some important issues which require improvement such as feature rescaling. Standardization is the most commonly used feature rescaling method. However, standardization does not always improve classification accuracy. This paper describes two feature rescaling methods： multiple kernel learning-based rescaling （MKL-SVM） and kernel-target alignment-based rescaling （KTA-SVM）. MKL-SVM makes use of the framework of multiple kernel learning （MKL） and KTA-SVM is built upon the concept of kernel alignment, which measures the similarity between kernels. The proposed meth- ods were compared with three other methods： an SVM method without rescaling, an SVM method with standardization, and SCADSVM. Test results demonstrate that different rescaling methods apply to different situations and that the proposed methods outperform the others in general.

Gene selection is an indispensable step for analyzing noisy and high-dimensional single-cell RNA-seq(scRNA-seq)*** with the commonly used variance-based methods,by mimicking the human maker selection in the 2D visualization of cells,a new feature selection method called HRG(Highly Regional Genes)is proposed to find the informative genes,which show regional expression patterns in the cell-cell similarity *** mathematically find the optimal expression patterns that can maximize the proposed scoring *** comparison with several unsupervised methods,HRG shows high accuracy and robustness,and can increase the performance of downstream cell clustering and gene correlation ***,it is applicable for selecting informative genes of sequencing-based spatial transcriptomic data.
