Anti-hepatitis C virus potency of a new autophagy inhibitor using human liver slices model

AIM: To evaluate the antiviral potency of a new antihepatitis C virus(HCV) antiviral agent targeting the cellular autophagy machinery. METHODS: Non-infected liver slices, obtained from human liver resection and cut in 350 μm-thick slices(2.7 × 106 cells per slice) were infected with cell culture-grown HCV Con1b/C3 supernatant(multiplicity of infection = 0.1) cultivated for up to ten days. HCV infected slices were treated at day 4 post-infection with GNS-396 for 6 d at different concentrations. HCV replication was evaluated by strand-specific real-time quantitative reverse transcription- polymerase chain reaction. The infectivity titers of supernatants were evaluated by foci formation upon inoculation into naive Huh-7.5.1 cells. The cytotoxic effect of the drugs was evaluated by lactate dehydrogenase leakage assays. RESULTS: The antiviral efficacy of a new antiviral drug, GNS-396, an autophagy inhibitor, on HCV infection of adult human liver slices was evidenced in a dosedependent manner. At day 6 post-treatment, GNS-396 EC50 was 158 nmol/L without cytotoxic effect(compared to hydroxychloroquine EC50 = 1.17 μmol/L).CONCLUSION: Our results demonstrated that our ex vivo model is efficient for evaluation the potency of autophagy inhibitors, in particular a new quinoline derivative GNS-396 as antiviral could inhibit HCV infection in a dosedependent manner without cytotoxic effect.

A comparison and interpretation of the international competitiveness of China and India

This paper first presents a comparison between China and India with respect to their overall economic and social development conditions and finds that China is currently at a higher development stage than India in terms of key economic indicators or social *** paper also conducts an assessment of the international competitiveness of the Chinese and Indian manufacturing industries and finds that the Chinese manufacturing industry is more competitive than its Indian *** paper then conducts an evaluation of the international competitiveness of the Chinese and Indian service industries and finds that(1)the Indian service industry has remained more globally competitive than its Chinese counterpart in most of the years examined;(2)the Chinese service industry has derived its international competitiveness primarily from its competitive *** spite of its weaker comprehensive competitiveness,the Chinese services industry has consistently secured a stronger competitive edge over its Indian *** Chinese service industry’s comparative competitiveness is weaker than India’s simply because the Chinese manufacturing industry has grown at a faster *** but not least,this paper conducts an in-depth analysis of the causes of competitive difference between China and India in respect of labor quality,infrastructure,government role,social heterogeneity,religion and culture as well as overseas compatriots.

Nickel-rich layered oxide cathode(LiNi_(x)Co_(y)Mn_(1−x−y)O_(2),x>0.5,NCM)shows substantial potential for applications in longer-range electrical ***,the rapid capacity decay and serious safety concerns impede its practical *** work provides a hydrogen-bonded organic framework(HOF)modification strategy to simultaneously improve the electrochemical performance,thermal stability and incombustibility of *** cyanurate(MCA),as a low-cost and reliable flame-retardant HOF,was implemented in the separator modification layer,which can prevent the battery short circuit even at a high *** addition,the supermolecule properties of MCA provide unique physical and chemical microenvironment for regulating ion-transport behavior in *** MCA coating layer enabled the nickel-rich layered oxide cathode with a high-capacity retention of 90.3%after 300 cycles at 1.0 ***,the usage of MCA in lithium-ion batteries(LIBs)affords a simple,low-cost and efficient strategy to improve the security and service life of nickel-rich layered cathodes.

The global campaign “Vision 2020： The Right to Sight” was officially launched in Geneva byDr. Gro Harlem Brundtland, the director General of World Health Organization （ WHO ） in February 1999. This represents the global initiative for the elimination of avoidable blindness by the year 2020.1 Seven months later, the International Agency for the Prevention of Blindness （IAPB） General Assembly was held in Beijing, China. During the IAPB General Assembly, the Chinese Ministry of Health, China Disabled Person＇ s Federation together with WHO launched the national campaign “Vision 2020： The Right to Sight” in China.

Study on New Pretreatment Combining Dilute Acid Treatment and Organic Solvent Water / Ethanol for Biorefinery： Application to Switch Grass

Switch grass was developed as a pioneer energy crop in USA with great industrial prospect. It contains about 60% sugars and 18% lignin. The purpose of this research is to find pretreatment process to fractionate cellulose, hemicellulose and lignin from switch grass to obtain much more useful chemicals and enhance the residue solid to be hydrolyzed by enzymes. The six different pretreatment methods were studied, such as hot water pretreatment （HWP） alone, dilute acid pretreatment （DAP）, ammonia pretreatment （AP）, lime pretreatment （LP）, organosolv water / ehanol pretreatment （OWEP）, and organosolv water / acetone pretreatment （OWAP）. It was the best method combining DAP with OWEP because the hemicellulose sugars were recovered in the first residual liquid while a varied amount of cellulose was retained in the residual solid and the lignin fraction was obtained by simply adjusting the pH from the second liquid. The result shows the optimal two-stage process consisted of the first stage DAP at 428 K for 7 min with 0.8% sulfuric acid, resulting in 79.82% glucose recovery yield and 98.74% xylose removal and the second stage OWEP at 468K for 20 min in 45% （v / v） ethanol with 0.4% NaOH, resulting in 62% total glucose yield 99% xylose and 80% lignin removal. After enzymatic hydrolysis, the glucose yield was up to 92.6%, compared with 16% yield from untreated switch grass. Scanning electron microscopy （SEM） highlighted the differences in switch grass structure from the various pretreatment methods during biomass fractionation.
