Mechanisms underlying lineage commitment and plasticity of human yo T cells

Phenotypic and functional heterogeneity are the hallmarks of effector and memory T cells. Upon antigen stimulation, y T cells differentiate into two major types of memory T cells： central memory cells, which patrol the blood and secondary lymphoid organs, and effector memory cells, which migrate to peripheral tissues, y T cells display in vitroa certain degree of plasticity in their function that is reminiscent of that which is observed in conventional CD4 T cells. Similar to CD4 T cells, in which a plethora of specialized subsets affect the host response, y8 T cells may readily and rapidly assume distinct Thl-, Th2-, Th17-, TFH and T regulatory-like effector functions, suggesting that they profoundly influence cell-mediated and humoral immune responses. In addition to differences in cytokine repertoire, y~ T cells exhibit diversity in homing, such as migration to lymph node follicles, to help B cells versus migration to inflamed tissues. Here, we review our current understanding of y T-cell lineage heterogeneity and flexibility, with an emphasis on the human system, and propose a classification of effector y T cells based on distinct functional phenotypes.

Sensor Deployment and Relocation:A Unified Scheme

Sensor networks are envisioned to revolutionize our daily life by ubiquitously monitoring our environment and/or adjusting it to suit our needs. Recent progress in robotics and low-power embedded systems has made it possible to add mobility to small, light, low-cost sensors to be used in teams or swarms. Augmenting static sensor networks with mobile nodes addresses many design challenges that exist in traditional static sensor networks. This paper addresses the problem of topology control in mobile wireless networks. Limitations in communication, computation and energy capabilities push towards the adoption of distributed, energy-efficient solutions to perform self-deployment and relocation of the nodes. We develop a unified, distributed algorithm that has the following features. During deployment, our algorithm yields a regular tessellation of the geographical area with a given node density, called monitoring configuration. Upon the occurrence of a physical phenomenon, network nodes relocate themselves so as to properly sample and control the event, while maintaining the network connectivity. Then, as soon as the event ends, all nodes return to the monitoring configuration. To achieve these goals, we use a virtual force-based strategy which proves to be very effective even when compared to an optimal centralized solution. We assess the performance of our approach in the presence of events with different shapes, and we investigate the transient behavior of our algorithm. This allows us to evaluate the effectiveness and the response time of the proposed solution under various environmental conditions.

Finite Sample Properties of Virtual Reference Feedback Tuning Control

In this paper, finite sample properties of virtual reference feedback tuning control are considered, by using the theory of finite sample properties from system identification. To design a controller in closed loop system structure, the idea of virtual reference feedback tuning is proposed to avoid the identification process corresponding to the plant model. After constructing one identification cost without any knowledge of plant model, the author derives one bound on the difference between the expected identification cost and its sample identification cost under the condition that the number of data points is finite. Also the correlation between the plant input and external noise is considered in the derivation of this bound. Furthermore, the author continues to derive one probability bound to quantify this difference by using some probability inequalities and control theory.

Role of blood AFP mRNA and tumor grade in the preoperative prognostic evaluation of patients with hepatoceiluiar carcinoma

AIM： To explore the potential prognostic role of preoperative tumor grade and blood AFP mRNA in a cohort of patients with hepatocellular carcinoma （HCC） eligible for radical therapies according to a well-defined treatment algorithm not including nodule size and number as absolute selection criteria. METHODS： Fifty patients with a diagnosis of HCC were prospectively enrolled in the study. Inclusion criteria were：（1） histological assessment of tumor grade by means of percutaneous biopsies; （2） determination of AFP mRNA status in the blood; （3） patient＇s eligibility for radical therapies. RESULTS： At preoperative evaluation, 54% of the study group had a well-differentiated HCC, 42% had AFP mRNA in the blood, 40% had a tumor larger than 5 cm and 56% had more than one nodule. Surgery （resection or liver transplantation） was performed in 29 patients, while 21 had percutaneous ablation procedures. After a median follow-up of 28 too, 12-, 24-, and 36-mo survival rates were 78%, 58%, and 51%, respectively. Surgical therapy, performance status and three tumor-related variables （AFP mRNA, HCC grade and gross vascular invasion） resulted as significant survival predictors at univariate analysis. Nodule size and number did not perform as significant prognosticators. Multivariate study selected only surgical therapy and a biologically early HCC profile （AFP mRNA negative and well-differentiated tumor without gross vascular invasion） as independent survival variables. CONCLUSION： The preoperative determination of tumor grade and blood AFP mRNA status may potentially refine the prognostic evaluation of HCC patients and improve the selection process for radical therapies.

Serum chromogranin-A in hepatocellular carcinoma: diagnostic utility and limits

AIM: The utility of serum alpha-fetoprotein (α-FP) for the detection of hepatocellular carcinoma (HCC) is questionable. High serum levels of chromogranin-A (CgA) have recently been reported in HCC. Impaired hepatic, renal, and heart functions influence circulating CgA. The aim of this study was to assess sensitivity and specificity of serum CgA as a marker of HCC in patients with liver cirrhosis (LC). METHODS: Serum CgA levels were measured by RIA in 339 patients of which 54 HCC, 132 LC, 45 chronic hepatitis (CH), 27 chronic heart failure (CHF), 36 chronic renal failure (CRF), 45 chronic inflammatory bowel disease (IBD) as disease controls and in 75 healthy controls. Patients with liver disease or IBD and concomitant renal and/or heart failure were excluded. Pearson correlation, non-parametric combination test and confidence interval analysis were used for statistical analysis. RESULTS: Serum CgA above normal values (100 ng/mL) were found in 83% of HCC patients, in 48% of LC patients, in 20% of CH patients, in 33% of IBD patients, in 92% of CRF patients, in 100% of CHF patients, and in none of the healthy controls. The mean CgA values in HCC (769±1046), in LC (249±369), in CH (87±94), in CRF (1390±1401), in CHF (577±539), in IBD (146±287) were significantly higher than those in healthy controls (48±18). HCC patients had higher CgA values (Pdid not differ from those with CRF or CHF. The 95% CI for the mean (250-1289 ng/mL) in HCC patients was selected as a CgA range and the lower value of such range was assumed as cut-off. Sensitivity and specificity of CgA, calculated in relation to the cut-off in patients with cirrhosis and HCC, were respectively 61% (CI 48-73%) and 82% (CI 75-88%). Serum a-FP values were >200 ng/mL in 21% of the HCC patients and in none of the LC patients. No significant correlation was found between a-FP and CgA in patients with HCC and in patients with cirrhosis. CONCLUSION: When HCC is suspected and a-FP

Impact of chronic exposure to bevacizumab on EpCAM-based detection of circulating tumor cells

Background： Circulating tumor cells （CTCs） are often undetected through the immunomagnetic epithelial cell adhesion molecule （EpCAM）-based CellSearch~ System in breast and colorectal cancer （CRC） patients treated with bevacizumab （BEV）, where low CTC numbers have been reported even in patients with evidence of progression of disease. To date, the reasons for this discrepancy have not been clarified. This study was carried out to investigate the molecular and phenotypic changes in CRC cells after chronic exposure to BEV in vitro. Methods： The human CRC cell line WiDr was exposed to a clinically relevant dose of BEV for 3 months in vitro. The expression of epithelial and mesenchymal markers and EpCAM isoforms was determined by western blotting and immunofluorescence. To evaluate the impact of EpCAM variant isoforms expression on CTC enumeration by CellSearch, untreated and treated colon cancer cells were spiked into 7.5 mL of blood from a healthy donor and enumerated by CellSearch. Results： Chronic exposure of CRC cell line to BEV induced decreased expression of EpCAM 40 kDa isoform and increased expression EpCAM 42 kDa isoform, together with a decreased expression of cytokeratins （CK）, while no evidence of epithelial to mesenchymal transition （EMT） in treated cells was observed. The recovery rate of cells through CellSearch was gradually reduced in course of treatment with BEV, being 84% , 70% and 40% at l, 2 and 3 months, respectively. Conclusions： We hypothesize that BEV may prevent CellSearch from capturing CTCs through altering EpCAM isoforms.

An Integrated Approach for Optimal Design of Micro Gas Turbine Combustors

The present work presents an approach for the optimized design of small gas turbine combustors, that integrates a0-D code, CFD analyses and an advanced game theory multi-objective optimization algorithm. The output of the0-D code is a baseline design of the combustor, given the required fuel characteristics, the basic geometry (tubularor annular) and the combustion concept (i.e. lean premixed primary zone or diffusive processes). For the optimizationof the baseline design a simplified parametric CAD/mesher model is then defined and submitted to a CFDcode. Free parameters of the optimization process are position and size of the liner hole arrays, their total area andthe shape of the exit duct, while different objectives are the minimization of NOx emissions, pressure losses andcombustor exit Pattern Factor. A 3D simulation of the optimized geometry completes the design procedure. As afirst demonstrative example, the integrated design process was applied to a tubular combustion chamber with alean premixed primary zone for a recuperative methane-fuelled small gas turbine of the 100 kW class.
