The cytokine granulocyte-macrophage-colony stimulating factor (GM-CSF) possesses the capacity to differentiate monocytes into macrophages (MØs) with opposing functions, namely, proinflammatory M1-like MØs and immunosuppressive M2-like MØs. Despite the importance of these opposing biological outcomes, the intrinsic mechanism that regulates the functional polarization of MØs under GM-CSF signaling remains elusive. Here, we showed that GM-CSF-induced MØ polarization resulted in the expression of cytokine-inducible SH2-containing protein (CIS) and that CIS deficiency skewed the differentiation of monocytes toward immunosuppressive M2-like MØs. CIS deficiency resulted in hyperactivation of the JAK-STAT5 signaling pathway, consequently promoting downregulation of the transcription factor Interferon Regulatory Factor 8 (IRF8). Loss- and gain-of-function approaches highlighted IRF8 as a critical regulator of the M1-like polarization program. In vivo, CIS deficiency induced the differentiation of M2-like macrophages, which promoted strong Th2 immune responses characterized by the development of severe experimental asthma. Collectively, our results reveal a CIS-modulated mechanism that clarifies the opposing actions of GM-CSF in MØ differentiation and uncovers the role of GM-CSF in controlling allergic inflammation.

Manufacturing plants are increasingly complex and integrated, requiring control systems able to identify the interactions between the various operating units. Production planning and control design of a process are tools that, if combined, bring many economic benefits to the processes since they aim to identify and maintain optimal decision operations to a system. This work uses such integration between production planning and plantwide control to propose a control system for the Williams-Otto plant from the definition of the operating optimal point for coordinated decentralized optimization, in which the original optimization problem decomposition into smaller coordinated problems ensure that the found local optimum also meets the requirements of the global system. The results for decentralized optimization are satisfactory and very similar to the global optimum problem and to the control system response proposed based on the optimal obtained. It is effective taking smooth actions, working with （economic） optimal set points （economically） of operation. The unification of production planning techniques and plantwide control techniques is an effective tool for the control system design for entire plants.

Interleukin 27(IL-27),a heterodimeric cytokine composed of Epstein-Barr virus-induced 3 and p28,is a pleiotropic cytokine with both pro-and anti-inflammatory ***,the precise role of IL-27 in acute graft-versus-host disease is not yet fully *** this study,utilizing mice with IL-27 p28 deficiency in dendritic cells(DCs),we demonstrated that IL-27 p28 deficiency resulted in impaired Treg cell function and enhanced effector T cell responses,corresponding to aggravated aGVHD in *** addition,using single-cell RNA sequencing,we found that loss of IL-27 p28 impaired Treg cell generation and promoted IL1R2^(+)TIGIT^(+)pathogenic CD4+T cells in the thymus at a steady ***,IL-27 p28 deficiency promoted STAT1 phosphorylation and Th1 cell responses,leading to the inhibition of Treg cell differentiation and ***,patients with high levels of IL-27 p28 in serum showed a substantially decreased occurrence of grade II-IV aGVHD and more favorable overall survival than those with low levels of IL-27 ***,our results suggest a protective role of DC-derived IL-27 p28 in the pathogenesis of aGVHD through modulation of the Treg/Teff cell balance during thymic ***-27 p28 may be a valuable marker for predicting aGVHD development after transplantation in humans.

This work aimed to study the derivatization unprecedented of glyphosate and AMPA solutions using N,O-bis (trimethylsilyl)trifluoroacetamide (BSTFA) combined with trimethylchlorosilane (TMCS), evaluating the composition of the reaction medium, use of ultrasound, volume of BSTFA:pyridine and pH of the reaction medium. From this study it was inferred that the reaction medium was composed of BSTFA:pyridine in ratio 60:100, respectively, without ultrasonic vibration and pH adjustment that provide optimal conditions for analysis by GC-MS. Furthermore, the methodology used was simple and fast, and that was the most practical method commonly used.

Background: Tropical forests play a fundamental role in the provision of diverse ecosystem services, such as biodiversity,climate and air quality regulation, freshwater provision, carbon cycling, agricultural support and culture. To understand the role of forests in the carbon balance, aboveground biomass(AGB) estimates are needed. Given the importance of Brazilian tropical forests, there is an urgent need to improve AGB estimates to support the Brazilian commitments under the United Nations Framework Convention on Climate Change(UNFCCC). Many AGB maps and datasets exist, varying in availability, scale and coverage. Thus, stakeholders, policy makers and scientists must decide which AGB product, dataset or combination of data to use for their particular goals. In this study, we assessed the gaps in the spatial AGB data across the Brazilian Amazon forests not only to orient the decision makers about the data that are currently available but also to provide a guide for future ***: We obtained a map of the gaps in the forest AGB spatial data for the Brazilian Amazon using statistics and differences between AGB maps and a spatial multicriteria evaluation that considered the current AGB datasets. The AGB spatial data gap map represents areas with good coverage of AGB data and, consequently, the main gaps or priority areas where further biomass assessments should focus, including the northeast of Amazon State, Amapá and northeast of Pará. The objective of this study was to characterize and verify the in vitro antitumor activity of essential oils (EOs) extracted from the leaves and flowers of Callistemon viminalis. The EOs were extracted by hydrodistillation using a modified Clevenger apparatus. The identification and quantification of constituents were performed on a gas chromatograph coupled to a mass spectrometer and a gas chromatograph with a flame ionization detector. The antitumoral activity was evaluated by a colorimetric assay (MTS) using different cell lines derived from human tumors (breast, lung, glioblastoma, and melanoma). The major constituents of the EOs of leaves and flowers were similar, only quantitative differences being observed. The compounds 1,8-cineole, α-pinene and α-terpineol were found in concentrations of 50.4%, 25.8% and 8.7% in the EOs obtained from the leaves and 48.8%, 24.5% and 3.9% in the EOs obtained from the flowers, respectively. The cytotoxic activity of the EOs was observed only in melanoma cultures (HT144). Cultures treated for 48 h with EOs from leaves and flowers (200 μg·mL-1) reduced the viability by 40% and 25%, respectively. Thus, the antiproliferative activity of the EO from leaves was more pronounced than the EO from flowers in cells derived from melanoma.
