BCNU/PLGA microspheres:a promising strategy for the treatment of gliomas in mice

Objective：To investigate the effects of BCNU/PLGA microspheres on tumor growth,apoptosis and chemotherapy resistance in a C57BL/6 mice orthotopic brain glioma model using GL261 cell ***：BCNU/PLGA sustained-release microspheres were prepared by the water-in-oil-in-water emulsion ***261 cells were intracranially injected into C57BL/6 mouse by using the stereotactic technology.A total of 60 tumor-bearing mice were randomly and equally divided into three groups：untreated control,PLGA treated,BCNU/PLGA *** resonance imaging （MRI） was taken to evaluate tumor ***/PLGA sustained-release wafers were implanted in the treatment group two weeks after *** time and quality were *** were harvested,and immunohistochemical staining was used to check the expression of Bax,Bcl-2,and O6-methylguanine-DNA methyltransferase （MGMT）.Statistical methods was used for analysis of relevant ***：BCNU/PLGA sustained-release wafers were fabricated and implanted *** is statistical difference of survival time between the BCNU/PLGA treated group and control groups （P＜0.05）.MRIscan showed inhibitory effect of BCNU/PLGA on tumor *** to the group A and B,BCNU/PLGA decreased the expression of apoptosis related gene Bcl-2 （P＜0.05）,but did not elevate the expression level of Bax （P＞0.05）,with the ratio of Bax/Bcl-2 *** MGMT protein expression,no statistically significant change was found in treated group （P＞0.05）.Conclusions：Local implantation of BCNU/PLGA microspheres improved the survival quality and time of GL261 glioma-bearing mice significandy,inhibited the tumor proliferation,induced more cell apoptosis,and did not increase the chemotherapy resistance.

目的 :进一步研究脑血管痉挛的发生机理 ,为临床治疗服务。方法 :采用 1 4只成年家犬 ,实验组 9只 ,对照组 5只 ,通过 2次枕大池注血法建立蛛网膜下腔出血 (SAH)模型。观察痉挛血管的自由基变化、血管活性、血管造影像、超微结构变化。另外 2 0只犬用来观察血管扩张剂的作用。结果 :实验组较对照组自由基含量高 ,血管活性降低 ,血管狭窄 ,管壁损害重。结论 :尼莫地平对急性痉挛有效 ,对慢性痉挛无效 ;慢性血管痉挛是以管壁结构性狭窄为特点。

Nanocatalytic medicine triggering in situ catalytic reactions has been considered as a promising strategy for tumor-selective ***,the targeted distribution of nanocatalysts was still low,considering the absence of targeting propulsion ***,encouraged by the fast-developing controllable microrobotics for targeting delivery,a sunflower-like nanocatalytic active swarm(SNCAS)controlled by a three-dimensional(3D)magnetic field was proposed for synergistic tumorselective and magnetic-actively tumor-targeting ***,a patient-derived renal cancer cell 3D organoid was utilized for the verification of the effective tumor therapeutic *** the targeted control of 3D magnetic field,the multiple cascade catalytic efficiency of SNCAS based on Fenton reaction was evaluated,resulting in efficient tumor cell apoptosis and *** the patient-derived organoid treatment,the SNCAS presented significant lethality toward 3D organoid structure to induce cell apoptosis with the collapse of organoid *** targeting efficiency was further enhanced under the magnetic-controllable of ***,empowered by the magnetic control technology,the synergistic therapeutic strategy based on controllable swarm combined active targeting and tumor-specific catalytic nanomedicine has provided a novel way for advanced cancer ***,3D patient-derived organoids were proved as a powerful tool for the effectiveness verification of nanocatalytic medicine.

Spinal cord——Interlaminar micro-neurosurgical discectomy in lumbar disk hermiation

Objective To investigate the surgical results of interlaminar microsurgical discectomy in lumbar disk herniation. Methods A study of 36 consecutive patients undergoing microsurgical lumbar discectomy was conducted. The extend of anular deficiency and the presence of disc fragment were determined. The clinical raiological parameters, type of surgery performed, follow-up, were assessed. Results We found that interlaminar disectomy without laminotomy was adequate in 28 cases （78 % ）. Seletive foraminotomy in addition to diseectomy was performed in 17 cases （47 % ）. Patients in the Fragment-Fissure and Fragment-Contained group had the best clinical outcomes, and lower rates of reherniation. Conclusion Interlaminar discectomy without laminotomy is a safe,

Fluorescence-guided microsurgical resection of human glioma with using 5-aminolevulinic acid

Objectitive The study was to evaluate the effectiveness of fluorescence-guided microsurgical resection of human glioma,and to investigate this technical tip and *** From Apil,2006 to October,2007,25 patients (13 women and 12 men;meanage 48.5 years old;agerange 15～66 years old) with human glioma underwent fluorescence-guided microsurgical resection by 5-aminolevulinic acid (5-ALA). The patients with glioma received 20 mg/kg 5-ALA before *** fluorescent regions of glioma were resected through a fluorescein surgical microscope after administration of *** evaluate the effectiveness of the resection,operative findings were compared with histological findings from different fluore scent areas of these *** patients were studied with MRI for detection of residual *** The rate of fluorescence positive was 96%, only one fluorescence negative case in all 25 *** viable tumor tissue was distinguished by its deep red fluorescence,whereas normal brain tissue was not *** histological findings revealed a large number of tumor cells and a conspicuous vascular reaction such as proliferation of endothelial cells in the fl uorescein-positive *** the other hand,the fluorescein-negative area showed only a small number of tumor cells and little vascular *** after operation result demonstrated the completeness of tumor removal in these patients. Conclusion These results suggest that fluorescence-guided microsur gical technique by administrating 5-ALA may be practical and useful for the resection of human *** this method,surgeon can easily and rapidly gained objecti vestandard to judge the extent of removal in the *** technique may also be useful in indentifying tumour margins during ***-up on patients who have undergone this surgery should be done to evaluate precisely effectiveness of this technique.
