Objective To study the effects of different concentrations of Quercetin on nitric oxide (NO) production and membrane fluidity of the injured human umbilical vein vascular endothelial cell line(ECV 304) with hypoxia and the lack of glucose. Methods The experiments were performed in the culture of ECV 304 injured with hypoxia and the lack of glucose in vitro. The releases of intracellular lactate dehydrogenase(LDH) of ECV 304 was measured with automatic biochemistry analysis. NO level of ECV 304 was monitored with colorimetry. The membrane fluidity of ECV 304 was measured with the fluorescence polarization method. Results After ECV 304 was cultured in hypoxia and the the lack of glucose for 24 hours, the release of LDH and the membrane fluidity were increased significantly; NO level was decreased. Preincubation of ECV 304 with 20, 80,160 μ mol·L -1  of Quercetin for 24 hours reduced LDH activity, membrane fluidity and increased the level of NO in hypoxia and the lack of glucose induced ECV 304. Conclusion These results demonstrate that Quercetin can produce the protective effect on hypoxia and the lack of glucose induced injury of ECV 304 by increasing release of NO and changing membrane fluidity.

The 3ω approach was used to measure the effective thermal conductivity of phase-change material microcapsules (PCMMs) based on urea formaldehyde and sliced paraffin. The effective thermal conductivities of PCMMs with different densities were measured within the phase-change temperature range. The relationships between effective thermal conductivity, density and temperature were analysed. The effective thermal conductivity reached peak values within the phase-change temperature range and the temperature peak value was consistent with the peak value of the phase-change temperature. The effective thermal conductivity increased with increasing density due to the decreased porosity of samples and their increased solid-phase conduction.

＜正＞ Pentoxifylline （PTX;50mg/kg daily i.p. for 4 - 8 weeks）, an aspecific phos-phodiesterase inhibitor with anti-inflammatory, anti-ischemic and anti-fibrotic activities, was administered to exercised mdx mice, a model for Duchenne muscular dystrophy. In vivo, the treatment contrasted the exercise-induced decrease in fore-limb muscle force. The ex vivo analysis included multidisciplinary biophysical, biochemical and histological approaches. PTX-treated limb muscle fibers had a restored calcium homeostasis. In fact the voltage threshold for contraction was returned to the control values;in parallel both the resting cytosolic calcium and the activity of calcium permeable TRP-like channels were markedly reduced. The treatment also fully counteracted the impairment of chloride channel conductance in diaphragm fibers. PTX-treated diaphragm and hind limb muscles showed a decrease in both non-muscle area and pro-fibrotic cytokine TGF-beta. In parallel the regenerating area was increased and the plasma level of creatine kinase reduced. The wide mechanism of action and a direct effect on the structures handling calcium may account for the beneficial effects of PTX in muscular dystrophy （Telethon GGP05130）.

给大鼠三七中人参二醇甙(PTS)150 mg/kgiv能明显抑制交感神经传出放电,减慢心率,暂时降低血压。PTS 75 mg/kg iv仅使血压呈现暂时升高。PTS 200μg/rat icv(约为iv有效量的1/150)也能明显抑制大鼠内脏交感神经传出放电,同时伴有血压降低和心率减慢,而用该剂量iv则对上述指标无任何影响。给大鼠icv PTS 400μg/rat可明显对抗iv乌头碱诱发的心律失常。上述结果表明PTS对中枢交感神经系统有抑制作用,此作用可能是其抗心律失常的作用机理之一。

In many tissues the parasympathetic and sympathetic nervous system regulate smooth musc tone via their transmitters aeetylcholine and noradrenaline, respectively. Direct smooth musc e e effects of acetylcholine via muscarinic receptors always promote contraction, but non-neuronal sources can importantly contribute to such stimulation. Direct smooth muscle effects of noradren- aline can promote contraction via al- and sometimes also α2-adrenoceptors but can promote re- laxation and inhibit contraction via β-adrenoceptors. I will focus on the interaction between sub- types of muscarinic and β-adrenergic receptors, largely using the urinary bladder as an exam- ple.

The discovery of the water channel aquaporin has greatly expanded our understanding of the regulation of the water permeability of biological membranes。The atomic structure of aquaporin-1 (AQP1) demonstrated how aquaporin is freely permeated by water but not protons and provided marked insight into several human disorders, 13 mammalian aquaporins have been identified, each with a distinct distribution, and these are selectively permeated by water or water plus glycerol。Aquaporins are suspected in numerous pathological conditions such as brain edema, diabetes mellitus。Knowledgeof structure, regulation and function of aquaporins may provide insight into the development of new therapeutics through appropriate drug design。

Recently, intense interest has focused on the antioxidant properties of natural products. In particular, Chinese herbal medicines (CHM) have become hot topics for life science researchers since many are reported to possess cardioprotective compounds, many of which remain to be indentified. Indeed, the exact mechanisms how many CHM work remain unknown. Although many of these herbal remedies are undoubtedly efficacious, few have been scientifically investigated for their active chemical constituents and biological activities. We have previously reported higher activities of antioxidant defence enzymes such as superoxide dismutase, catalase, glutatione perioxidase and glutathione S-transferases in the liver of rats treated with the herb Salviae Miltiorrhizae (SM) in a rat model of acute myocardial infarction. Using well established in vitro antioxidant assays employing 2'-azino-bis(3-ethylbenz-thiazoline-6-sulfonic acid (ABTS) and diphenyl-1-plcrylhydrazyl (DPPH), we have shown that in addition to elevating endogenous antioxidant enzyme activity, SM and other CHM traditionally used for cardiovascular disorders (Rhizoma ligustici, Herba leonuri and Radix achyranthis bidentatae) contained potent antioxidant moieties in addition to their phenolic constituents. Furthermore, these novel non-phenolic components were effective inhibitors of oxidative reactions mediated by the inflammatory oxidants peroxynitrite, hypochlorous acid and hydroxyl radical as well as iron-dependent lipid peroxidation. The various antioxidant properties of CHM will have great clinical application in the new era.

The Role of TNF Related Apoptosis-Inducing Ligand in Neurodegenerative Diseases

＜正＞ Recently,we have presented the antifibrillatory interactions between lidocaine and two antihypertensive drugs,propranolol （Almotrefi et al., 1999） and valsartan （Almotrefi ＆ Arif, 2004）. This abstract reports its interactions with captopril. Studies were carried out on hearts isolated from New Zealand white rabbits of either sex weighing 1 to 2 Kg. The method used has been described previously （Almotrefi ＆ Baker. 1981）. Perfusion with lidocaine produced significant,dose-dependent increase in VFT while perfusion with captopril did not cause any significant change. In addition,there was no significant difference in VFT with the combined infusion of 3.46 μmol of lidocaine and μmol of captopril. in contrast to a synergistic antifibrillatory effect of the combined use of lidocaine and propranolol （Almotrefi et al., 1999）. This suggests that captopril does not have antifibrillatory interactions with lidocaine. indicating its safety in combining with class 1 antiarrhythmic ***, lidocaine, captopril.

A hallmark of all forms of neurodegenerative diseases is impairment of neuronal functions,and in many cases neuronal cell *** the etiology of neurodegenerative diseases may be distinct,different diseases display a similar pathogenesis,for example abnormal immunity within the central nervous system(CNS),activation of macrophage/microglia and the involvement of proinflammatory *** studies show that neurons in a neurodegenerative state undergo a highly regulated programmed cell death,also called ***-related apoptosis-inducing ligand(TRAIL),a member of the TNF family,has been shown to be involved in apoptosis during many *** one member of a death ligand family,TRAIL was originally thought to target only tumor cells and was not present in ***,recent data showed that TRAIL was unregulated in HIV-1-infected and immune-activated macrophages,a major disease inducing cell during HIV-1-assoeiated dementia(HAD).TRAIL is also induced on neuron by β-amyloid protein,an important pathogen for Alzheimer's *** this review,we summarize the possible common aspects that TRAIL involved those neurodegenerative diseases,TRAIL induced apoptosis signaling in the CNS cells,and specific role of TRAIL in individual *** & Molecular Immunology.2005;2(2):113-122.
