The chitin is extracted from the cuttlebone of S. inermis and the mineral contents are predictable. The structure and degree of deacetylation of extracted cuttlebone chitin is dogged through Fourier Transform Infrared (FT-IR) spectroscopy. The extracted cuttlebone chitin is used as the substrate for the production of chitinase from Vibrio sp. The extra cellular proteins are concentrated by ammonium sulphate precipitation, dialysed and then purified by using gel (sephadex G-100) chromatography. Among the 40 fractions, only two fractions (active fractions) showed maximum absorbance at 280 nm, which are pooled, dialysed and free-dried. The enzyme activity (0.104 μmoles/ml) and molecular weight (50 - 60 kDa) of purified chitinase is also determined through SDS- PAGE. The optimal condition for chitinase activity is pH between 6.0 - 6.5 and 45℃.

＜正＞ A preliminary study on the pattern of the use of statins from May-July 2004 in three hospitals in Jakarta was conducted to see the responder rate of the patients. The inclusion criteria are 243 outpatients （127 male. 56±12 yrs and 116 female. 55±12 yrs）. diagnosed with dyslipidemia. treated with statins either as first choice or addon therapy and other lipid-lowering drugs. Atorvastatin is the most used statins as first choice （38.1% ） and as add-on （1.6% ）. followed by rosu-vastatin （20.5%）,fluvastatin （11.5%）, pravastatin （10.7%）;whereas lovas tatin （0.4% ） and simvastatin （3.7% ） were least prescribed. Patients’ TC levels were reduced significantly （△±14%. p ＜ 0.0001）. while Tg levels were not significantly reduced （△±8 % ）. Around 46 % cases （72 cases） met the NCEP ATP Ⅲ goals on TC level. The combinations most used, with respect to atorvastatin, are fenofibrate （11.3%）. pravastatin （9.3%）. rosuvastatin （3 % ）. and gemfibrozil （2 % ）. Regarding rosuvastatin are gemfibrozil （9 % ）. ciprofibrate （5%）. and atorvastatin （3%） . A large number, however, do not comply. Conclusion, the inappropriate usage of statins is due to various factors, will be discussed in the paper.

Objective: The increased perinatal morbidity in diabetes may be partly related to vascular dysfunction. Because potassium channels play an important role in the regulation of vascular tone, this study explores the impact of diabetes on potassium channel function in the fetoplacental vascular bed. Study design: Vascular potassium channel function was investigated by ex vivo dual perfusion of isolated placental cotyledons (n = 47). Appropriate control experiments were carried out to exclude nonspecific effects. Results: Glibenclamide (KATP channel blocker) increased perfusion pressure to a maximum fetoplacental arterial pressure of 37 ± .6 mm Hg in controls versus 15 ± 6 mm Hg in diabetes (P <. 05). 4- Aminopyridine (KV channel blocker) equally increased fetoplacental arterial pressure in controls, and in diabetes (21 ± 4 mm Hg vs 22 ± 2 mm Hg). Apamin and charybdotoxin (KCa channel blockers) caused a negligible rise in fetoplacental arterial pressure. Conclusion: In the fetoplacental circulation, KATP channels and KV channels significantly contribute to baseline vascular tone. In diabetes, vascular KATP channel function is impaired.

＜正＞ Previous study showed that 1.10 - phenantroline skeleton was active in vitro on chloroquine - resistant and sensitive strain of Plasmodium falciparum. Based on the skeleton. 8 derivatives of N - alkyl and N - benzyl 1.10 - phenantrolines have been synthesized. This study was conducted to evaluate in vitro antiplasmodialactivity and cytotoxicity of these compounds. The in vitro antiplasmodial on chloroquine-resistant P. falciparum strain （FCR- 3）. chloroquine - sensitive P. falciparum strain （D10） and cytotoxicity test on Vero cells were determined by radioactive method after 24 and 72 h incubation periods, and were expressed by the 50 % concentration inhibiting of the parasite or cell growth （IC50）. Cytotoxic/antiplasmodial ratio was calculated to evaluate its safety. The highest antiplasmodial activity was observed for （1） - N - benzyl - 1.10 - phenantroliniumiodide with IC50 0.08 - 0.59μM, IC50 on Vero cells was 2207.77 - 126631. 51 μM. and cytotoxic/antiplasmodial ratio showed that this compound was safe （9199.04-214629.67）.

＜正＞ The present study aimed to elucidate the therapeutic effects of melatonin against E. coli - induced renal injury. Wistar albino rats, injected intrarenally with E. Coli. were administered with either saline or melatonin （10 mg/kg/day;intraperitoneally）.Twenty - four hours or one week after pyelonephritis induction, rats were decapitated. In kidney samples, malondialdehyde （MDA）,glutathione （GSH） levels, myeloperoxidase （MPO） activity and collagen content were measured and histological analyses were made. In the saline - treated pyelonephritis group, a decrease in renal GSH along with increases in MDA level. MPO activity, and collagen content were observed （p＜0.05-0.00l）. while serum TNF -α. lactate dehydrogenase. BUN and creatinine levels were elevated as compared to control. However, melatonin treatment reversed all these biochemical indices （p ＜ 0.05）. as well as renal injury observed histologically. The protective effects of melatonin may be due to its ability to inhibit neutrophil infiltration and to balance oxidant- antioxidant status, suggesting a future role for melatonin in acute pyelonephritis treatment.

Objective:To evaluate the wound healing activity of the methanolic root extract of Buchanania lanzan Spreng.(***),with a focus on antimicrobial and anti-biofilm ***:The extract was evaluated for its wound healing properties(excision and incision models)as evident from the analysis of tensile strength and wound *** extract was also screened for antibacterial properties against different Cram positive and Gram negative bacterial *** was also studied for its effect on biofilm formation and disruption of preformed *** synergistic effect of *** was determined in combination with ***:Topical application of ***(10%w/w ointment)significantly increased(40.84%)the tensile strength in the incision wound *** also showed significant wound healing activity in excision model and such significant activity was observed from the 9th *** Soframycin displayed significant wound healing activity from the 6th *** was found that root extracts of *** revealed significant inhibition against all tested *** displayed antimicrobial activity against Cram positive(MIC 0.625 mg/mL)and Cram negative(MIC0.625-1.25 mg/mL).*** was able to reduce biofilm formation and also caused disruption of preformed biofilms in a manner similar to ***,gentamicin was found to be ineffective against biofilms formed by Gram negative *** to the fractional inhibitory concentration index,*** displayed synergistic activity when it was combined with ***:From this study it may be concluded that the root extract of *** revealed significant wound healing potential,which was supported and well correlated with pronounced antibacterial activity of the tested plant parts.

A Portable Device for Intermittent Oxygen Supplementation during High-Intensity Exercise

Introduction: Oxygen supplementation increases physical performance during strenuous work, as known for more than 100 years. There is no widespread use among athletes, which could be due to logistical challenges with gas supply when breathing over 200 L/min. We hypothesized that a positive effect could be reached with a moderate increase in alveolar PO2. If so, a technical solution here called “Oxelerator” could be constructed combining intermittent gas supply with oxygen concentrators running of electricity and a small bottle. Methods. This study describes the theoretical details regarding gas flow and consumption as well as a practical test of the Oxelerator when used during intense exercise by two athletes with high VO2 and work capacity. Results: The Oxelerator generates a peak gas flow of 32 L/min dosed intermittently in 400 - 700 ms boluses adjusted to the inspiratory phase of breathing, mixing oxygen with surrounding air. Theoretical calculations show that 1 liter tank suffice for 20 - 40 minutes of high intensity exercise and hyperoxic FEO2 while breathing above 200 L/min. The first subject ran on a treadmill at a VO2 of 4.64 L/min, RER 0.94. Then intermittent oxygen supplementation was initiated, and FEO2 reached 25.96% after 1 minute. The second subject rowed an ergometer at 330 W. VO2 stabilized at 5.67 L/min, with RER 0.91. After 1 minute of oxygen supplementation mixed expired FEO2 was 22.58% suggesting that inspired FO2 averaged 28 - 31%. Conclusions: A portable technical solution is proposed that enable intermittent O2 supplementation enough to increase end-expiratory FO2 during intense workloads. This will enable hyperoxic exercise for athletes.

＜正＞ The aim of our study was to compare hypoglycemic effects of cholic henodeoxy-cholic and chemicaly synthetized monoketocholic （MKH） acid and in combination with insulin （INS） after intranasal （IN） and intravenous （IV） administration. Their toxicity was tested by glutathione and protein content in liver. Anaesthetized rats received bile acids as 2% sodium salt solutions （2 mg/kg） as in combination with INS （10 IU/kg）. Glucose levels were measured in 0, 15, 30, 60, 90, 120 and 180th min. Liver samples were taken in 0 min. and in every hour after IV injection （4 mg/kg）. According to the initial time blood glucose levels were changing significantly after IN application of all bile salts. Areas under the curve had shown great hypoglycaemic effect （p ＜ 0, 001） in relation to controls and INS. There were no significant changes in glutathione and protein content compared to control groups. Our results confirmed that bile salts enhance the INS effects, but also indicate that they per se can affect blood glucose levels. Based on our results we might presume that IV or IN applied bile salts could be safely used as insulin promoters.
