AIM:To evaluate the safety and efficacy of stapled transanal rectal resection(STARR),and to analyze the outcome of the patients 12-mo after the ***:From May 2007 to October 2008,50 female patients with rectocele and/or rectal intussusception underwent *** preoperative status,perioperative and postoperative complications at baseline,3,6 and 12-mo were *** were collected prospectively from standardized questionnaires for the assessment of constipation[constipation scoring system,Longo’s obstructed defecation syndrome(ODS)score system,symptom severity score],patient satisfaction (visual analogue scale),and quality of life(Patient Assessment of Constipation-Quality of Life Questionnaire).RESULTS:At a 12-mo follow-up,significant improvement in the constipation scoring system,ODS score system,symptom severity score,visual analog scale and quality of life(Pmed safely without major ***,the procedure seems to be effective for patients with obstructed defecation associated with symptomatic rectocele and rectal intussusception.

Calmodulin (CaM), widely distributed in almost all eukaryotic cells, is a major intracellular calcium receptor responsible for mediating the Ca2 + signal to a multitude of different enzyme systems and is thought to play a vital role in the regulation of cell proliferative cycle[1，2]. Recently, many studies showed that CaM is also present in extracellular fluid such as cell culture media and normal body fluid and has been reported to stimulate proliferation in a range of normal and neoplastic cells, apparently acting as an autocrine growth factor[3-11]. In 1988, Crocker et al reported for the first time that addition of extracellular pure pig brain CaM could promote DNA synthesis and cell [7]proliferation in K562 human leukaemic lymphocytes[7].After that, more and more research was done on extracellular CaM and evidences demonstrated that extracellular CaM could also stimulate cell proliferation in normal human umbilical vein endothelial cells[5], keratinocytes[4], suspension-cultured cells of Angelica Dahurica, etc[6]. CaM is a monomeric protein of 148 amino acids that contains four homologous Ca2 + -binding domains. CaM has been highly conserved throughout the evolution. Only 1 out of 148 amino acids of human CaM is different from that of fish CaM. Complementary DNAs encoding rat, eel, chicken, human, and trypanosome CaM have been cloned.

Nerve cell metabolism in post brain ischemia depends on increased microcirculation perfusion and transport function of microvascular endothelial cells. In the present study, a rat model of middle cerebral artery occlusion was established to investigate the influence of electroacupuncture （EA） on hippocampal CA1 cerebral blood flow and glucose transporter 1 （GLUT1） expression in the microvascular endothelial cells. Following EA at Neiguan （PC 6）, the cerebral blood flow in the ischemic hippocampal CA1 region was significantly elevated, the number and microvascular integrated absorbance of the GLUTl-positive cells were significantly increased, nerve cell damage was ameliorated, and GLUT1 protein expression in the ischemic hippocampus was significantly increased. Results demonstrate that EA increased the cerebral blood flow of the hippocampal CA1 region and improved the glucose transport function, thereby attenuating neuronal injuries.

By homologous expressed sequence tag (EST) searching, one EST (GenBank: W29095) was obtained, which shows 75% identity in 435 bp overlap with the coding sequence of mouse Cacng2 gene. A 1 545 bp cDNA fragment was obtained from the nested polymerase chain reaction (PCR) and rapid applification of cDNA end (RACE) reaction in the human brain prefrontal cortex cDNA library and the human brain Ready cDNA with the primers designed on W29095. The fragment contained a 948-bp open reading frame (ORF) encoding 315 amino acids, and was named CACNG3. As it was identical to a BAC clone (GenBank: AC004125) from chromosome 16p12-p13.1, the CACNG3 gene was mapped to human chromosome 16p12-p13.1, and the coding region was composed of 4 exons. Reverse transcription PCR (RT-PCR) analysis showed that the CACNG3 gene expressed in human adult brain and fetal brain. Senegenin, an effective component of Polygala tenuifolia root extract, promotes proliferation and differentiation of neural progenitor cells in the hippocampus. However, the effects of senegenin on mesencephalon-derived neural progenitor cells remain poorly understood. Cells from a ventral mesencephalon neural progenitor cell line （ReNcell VM） were utilized as models for pharmaceutical screening. The effects of various senegenin concentrations on cell proliferation were analyzed, demonstrating that high senegenin concentrations （5, 10, 50, and 100 μmol/L）, particularly 50 μmol/L, significantly promoted proliferation of ReNcell VM cells. In the mitogen-activated protein kinase signal transduction pathway, senegenin significantly increased phosphorylation levels of extracellular signal-regulated kinases. Moreover, cell proliferation was suppressed by extracellular signal-regulated kinase inhibitors. Results suggested that senegenin contributed to in vitro proliferation of human neural progenitor cells by upregulating phosphorylation of extracellular signal-regulated kinase.

Berberine, a major constituent of Coptidis rhizoma, exhibits neural protective effects. The present study analyzed the potential protective effect of berberine against amyloid β-induced cytotoxicity in rat cerebral cortical neurons. Alzheimer＇s disease cell models were treated with 0.5 and 2 μmol/L berberine for 36 hours to inhibit amyloid β-induced toxicity. Methyl thiazolyl tetrazolium assay and terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling staining results showed that berberine significantly increased cell viability and reduced cell apoptosis in primary cultured rat cortical neurons. In addition, western blot analysis revealed a protective effect of berberine against amyloid β-induced toxicity in cultured cortical neurons, which coincided with significantly decreased abnormal up-regulation of activated caspase-3. These results showed that berberine exhibited a protective effect against amyloid β-induced cytotoxicity in cultured rat cortical neurons.

Objective To investigate the efficacy and safety of acute stent implantation during endovascular treatment for patients with emergent large vessel occlusion due to intracranial atherosclerotic *** A retrospective analysis was carried out on 46 patients with emergent large vessel occlusion due to intracranial atherosclerotic stenosis whoreceivedendovascular treatment at the Strategic Support Force medical Center from January 2015 to August ***-seven patients underwent balloon angioplasty alone and 19 patients underwent acute stent *** baseline characteristics,modified thrombolysisincerebral infarction(mTICI)score of the responsible vessels,modified Rankin scale(mRS)score 90 days after operation,incidence of symptomatic intracranial hemorrhage and mortality of the two groups were *** The proportion of effective recanalization of the offending vessels(mTICI≥2b)in the acute stenting group was slightly higher than that in the balloon angioplasty group(16/19 vs.81.5%),but the difference was not statistically significant(P>0.05).Besides,there was no significant difference in the median of mRS between the acute stenting group[3.0(0,4.0)]and the balloon angioplasty group[4.0(1.0,5.0)]90 days after operation(P>0.05).

Objective To investigate the safety and efficacy of donor-derived CD19+or sequential CD19+CD22+chimeric antigen receptor T-cell(CAR-T)therapy in patients with B-cell acute lymphoblastic leukemia(BALL)afterallogeneic hematopoieticstem cell transplantation(allo-HSCT).Methods The data of 22 patients with B-ALL who relapsed after allo-HSCT and underwent donor-derived CAR-T therapy at the Zhujiang Hospital of Southern medical University and the 920th Hospital of Joint Logistics Support Force of the People's Liberation Army of China from September 2015 to December 2022 were retrospectively *** primary endpointwasoverall survival(OS),and the secondary endpoints wereeevent-free survival(EFS),complete remission(CR)rate,and Grade 3-4 adverse *** A total of 81.82%(n=18)of the 22 patients achieved minimal residual disease-negative CR after CAR-T *** median follow-up time was 1037(95%CI 546-1509)days,and the median OS and EFS were 287(95%CI 132-441)days and 212(95%CI 120-303)days,*** 6-month 0S and EFS rates were 67.90%(95%CI 48.30%-84.50%)and 58.70%(95%CI 37.92%-79.48%),respectively。
