Krill oil attenuates obesity-induced skeletal muscle atrophy in mice

Obesity is associated with skeletal muscle mass loss and physical *** oil(KO)has been shown to be beneficial in human ***,the effect of KO on obesity-induced skeletal muscle atrophy is still *** this study,the male C57BL/6J mice were fed a high-fat diet(HFD)for 12 weeks to induce obesity,and then were intragastric administration with 400 mg/kg bw KO for an additional 6 *** results showed that KO treatment reduced body weight,fat accumulation and serum pro-inflammatory cytokines in HFD-induced obese ***,KO treatment attenuated skeletal muscle atrophy in HFD-fed mice,as evidenced by preserving skeletal muscle mass,average myofiber cross-sectional area and grip *** administration also mitigated obesity-induced ectopic lipid deposition and inflammatory response in skeletal ***,KO treatment inhibited the transcriptional activities of nuclear factor-κB(NF-κB)p65 and forkhead box O 3a(FoxO3a),and then down-regulated muscle atrophy F-box(MAFbx)and muscle-specific RING finger protein 1(MuRF1)protein levels in skeletal muscle from HFD-fed *** administration also improved obesity-induced impaired muscle protein synthesis via activating PI3K/Akt ***,KO treatment enhanced muscle mitochondrial biogenesis in HFD-induced obese mice via activating PGC-1α***,KO might be developed as a potential nutritional supplement for the prevention and treatment of obesity-induced skeletal muscle atrophy.

Salvianolic acids improve liver lipid metabolism in ovariectomized rats via blocking STAT-3/SREBP1 signaling

Postmenopausal women, who have reduced circulating estrogen levels, are more prone to develop obesity and related metabolic diseases than premenopausal women. The absence of safe and effective treatments for postmenopausal obesity has changed the focus to natural products as alternative remedies. Total salvianolic acids(TSA) are the major water-soluble ingredients of Danshen. Salvianolic acid(SA) is the major constituent of the TSA. Salvianolic acids, including TSA and SA, are widely used in traditional Chinese medicine. In the present study, ovariectomized rats and LO2 cells were used to study the effects of salvianolic acids on body weight gain and hepatic steatosis. Salvianolic acids reduced ovariectomy(OVX)-induced body weight gain, attenuated the expressions of hepatic lipogenic genes, such as sterol regulatory element binding protein(SREBP)1, fatty acid synthase(FAS), and stearoyl-CoA desaturase(SCD)1, and decreased the liver triglyceride(TG) and total cholesterol(TC). For the molecular mechanisms, OVX and high glucose-induced phosphorylation of signal transducer and activator of transcription(STAT)-3 was inhibited by salvianolic acids treatment. In LO2 cells, inhibition of STAT-3 by siRNA attenuated the increased expression of SREBP1 and TG induced by high glucose. Salvianolic acids reduced the upregulation of SREBP1 and TG induced by high glucose in LO2 cells. In conclusion, these findings illustrated that salvianolic acids markedly alleviated the lipid metabolism disorders and protected against the postmenopausal obesity. The underlying mechanism was probably associated with the regulation of STAT-3 signaling.

Plasma Homocysteine and Gene Polymorphisms Associated with the Risk of Hyperlipidemia in Northern Chinese Subjects

Objective To examine the relationship between occurrence of hyperlipidemia, plasma homocysteine and polymorphisms of methylenetetra hydrofolate reductase （MTHFR） gene and methionine synthase （MS） gene. Methods A total of 192 hyperlipidemia patients were selected and divided into hypercholesterolemia group, hypertriglyceridemia group, and combined hyperlipidemia group. Another 208 normal individuals were selected as control. Total plasma homocysteine （tHcy） concentration was measured by high-performance liquid chromatography （HPLC）. Lipid profiles were measured for all subjects The polymorphisms of MTHFR gene C677T and MS gene A2756G were analyzed by PCR-RFLP. Results The tHcy concentration in the combined hyperlipidemia patients was significantly higher than that in the control （15.95μmol/L vs 13.43 μmol/L, P〈0.05）. The prevalence of hyperhomocysteinemia （HHcy） in the combined hyperlipidemia group was significantly higher than that in the control （42.2% vs 23.0%, P=0.015）, with the odds ratio （OR） of 3.339 （95%CI： 1.260-8.849）. The hyperlipidemia patients with HHcy had a higher concentration of total cholesterol （TC） than that in the normal tHcy patients （5.67±0.95 mmol/L vs 5.47±0.92 retool/L, P=0.034）. There was no significant difference in genotype or allele frequencies of MTHFR C677T between the hyperlipidemic and control groups. The hyperlipidemia patients with MTHFR CT/TT genotype had a higher concentration of triglyceride （TG） than those with CC genotype （2.24±1.75 mmol/L vs 1.87±0.95 mmol/L, P〈0.05）. Individuals with CT/TT genotype had a higher concentration of tHcy than those with 677CC genotype both in the hyperlipidemia group （12.61±1.24μmol/L vs 11.20±1.37 μmol/L, P〈0.05） and in the control group （14.04±1.48 μmol/L vs 12.61±1.24 μmol/L, P〈0.05）. The percentage of MS 2756 GG/AG genotype in the combined hyperlipidemia group was significantly higher than that in the control （26.7% vs 13.0%, P=0.012）, with the OR of 3.121 （95%C1： 1.288-7.65/）. It is a multifactorial metabolic disorder that results from a combination of resistance to insulin action and an inadequate compensatory insulin secretory response. 27-Hydroxycholesterol-induced EndMT acts via STAT3 signaling to promote breast cancer cell migration by altering the tumor microenvironment

Objective:The endothelial to mesenchymal transition(EndMT)plays a major role in cancer metastasis by regulating the complexity of the tumor microenvironment(TME).Here,we investigated whether 27-hydroxycholesterol(27 HC)induces EndMT in endothelial cells(ECs).Methods:EndMT markers in the human microvascular endothelial cell-1(HMEC-1)cell line and human umbilical vein endothelial cells(HUVECs)stimulated with 27 HC were evaluated with Western *** to mesenchymal transition(EMT)markers in breast cancer(BC)cells cultured in conditioned medium were investigated with quantitative real time polymerase chain reaction(qRT-PCR).The MMP-2 and MMP-9 mRNA expression and activity were detected with qRT-PCR and gelatin zymography assays,*** effect of activated STAT3 on 27 HC-induced EndMT was validated by Western blot,immunofluorescence staining,and cell transfection *** migration ability of BC cells was evaluated with Transwell ***:We found that 27 HC induced EndMT in HMEC-1 and HUVECs,and 27 HC-induced EndMT facilitated EMT and BC cell *** 27 HC-induced EMT of BC cells also promoted EndMT and HUVEC *** of the underlying molecular mechanisms revealed that STAT3 knockdown repressed EndMT in HUVECs as well as migration in BC cells induced with 27 *** addition,C646 and resveratrol,inhibitors of STAT3 acetylation,repressed the expression of Ac-STAT3,p-STAT3,and EndMT markers in HUVECs exposed to 27 HC;these HUVECs in turn attenuated the migration ability of BC cells in 27 HC-induced ***:Cross-talk between 27 HC-induced EndMT and EMT was observed in the ***,activation of STAT3 signaling was found to be involved in 27 HC-induced EndMT.

Effect of Zinc on Bone Metabolism in Fetal Mouse Limb Culture

Objective To determine the effects of zine-deficiency and zine-excess on hone metabolism. Methods We developed the culture model of fetal mouse limbs (16th day) cultivated in self-made rotator with continuing flow of mixed gas for six days in vitro. The cultured limbs were examined by the techniques of 45Ca tracer and X-roentgenography. Results The right limbs cultivated had longer bone length, higher bone density than the left limbs uncultivated from the same embryo; and histologically, the right limbs had active bone cell differentiation, proliferation, increased bone trabecula. clearly calcified cartilage matrix, and osteogenic tissue. Compared with the control group, the zinc-deficient group and zine-excess (Zn2+ l20) μmol/L) group contained less osteocalcin (BGP) and 45Ca content, and lower AKP activity; whereas zine-normal (Zn2+ 45 μmol/L and Zn2+ 70 μmol/L) groups contained more BGP and 45Ca contents, and higher AKP (alkaline phosphatase) activity. Conclusion Both zine-deficiency and zine-excess can alter bone growth and normal metabolism. The results indicate that the culture model of fetal mouse limbs (16th day) in vitro can be used as a research model of bone growth and development.

The purpose of this research was to evaluate radiological safety in pediatric radiology in hospitals in the Kongo Central province of the DRC. To this end, we surveyed a convenience sample of 50 health professionals, including 10 radiologists working in the hospitals covered by the survey, to assess the practice of pediatric radiology and the degree of compliance with radiation protection principles for the safety of children and the environment. We collected radiophysical parameters to calculate entrance doses in pediatric radiology in radiology departments to determine the dosimetric level by comparison with the diagnostic reference levels of the International Commission on Radiological Protection (ICRP). All in all, we found that in Kongo Central in the DRC, many health personnel surveyed reported that more than 30% of requested radiological examinations are not justified. Also, after comparing the entrance doses produced in the surveyed departments with those of the International Commission on Radiological Protection (ICRP), a statistically significant difference was found in pediatric radiology between the average doses in five out of six surveyed departments and those of the ICRP. Therefore, almost all of the surveyed departments were found to be highly irradiating in children, while excessive X-ray irradiation in children can have significant effects due to their increased sensitivity to radiation. Among the risks are: increased cancer risks, damage to developing cells, potential genetic effects, and neurological effects. This is why support for implementing radiation protection principles is a necessity to promote the safety of patients and the environment against the harmful effects of X-rays in conventional radiology.
