Rapid and sensitive detection of various analytes is in high *** from its application in genome editing,CRISPR-Cas also shows promises in nucleic acid detection *** further exploit the potential of CRISPR-Cas for detection of diverse analytes,we present a versatile biosensing platform that couples the excellent affinity of aptamers for broad-range analytes with the collateral single-strand DNA cleavage activity of CRISPR-Cas12 *** demonstrated that the biosensors developed by this platform can be used to detect protein and small molecule in human serum with a complicated background,i.e.,the tumor marker alpha fetoprotein and cocaine with the detection limits of 0.07 fmol/L and 0.34 lmol/L,respectively,highlighting the advantages of simplicity,sensitivity,short detection time,and low cost compared with the state-of-the-art biosensing ***,this biosensing platform with plug-and-play design show great potential in the detection of diverse analytes.

Risk calculators have offered a viable tool for clinicians to stratify patients at risk of prostate cancer(PCa)and to mitigate the low sensitivity and specificity of screening prostate specific antigen(PSA).While initially based on clinical and demographic data,incorporation of multiparametric magnetic resonance imaging(MRI)and the validated prostate imaging reporting and data system suspicion scoring system has standardized and improved risk stratification beyond the use of PSA and patient parameters ***-naïve patients with lower risk profiles for harboring clinically significant PCa are often subjected to uncomfortable,invasive,and potentially unnecessary prostate biopsy *** risk calculator data into prostate MRI reports can broaden the role of radiologists,improve communication with clinicians primarily managing these patients,and help guide clinical care in directing the screening,detection,and risk stratification of PCa.

LL1, a novel specific STAT3 inhibitor, displays anti-colorectal cancer activities in vitro and in vivo

OBJECTIVE The signal transducer and activator of transcription 3(STAT3) is associated with the development and progression of numerous types of human cancer, and STAT3 activation confers metastasis to ***, no STAT3 inhibitor was used clinically. Hence,we discovered a novel potent small-molecule inhibitor of STAT3, LL1, and studied its antitumor effects and the underlying mechanisms in two tumor models. METHODS Using structure-based drug design method, based on the crystal structure of STAT3 protein, we identified a potent STAT3 inhibitor(LL1) targeting STAT3 SH2 domain and characterized its therapeutic properties and potential toxicity in vitro and in vivo using the MTT assay, colony formation assays, histological, immunohistochemical,flow cytometric analysis, tumor xenograft model. RESULTS LL1 is highly selective among STATs family members and specifical y inhibits phosphorylation of STAT3 Tyr-705 site, blocking the whole transmission process of STAT3 signaling. Mechanistical y, LL1 inhibited proliferation, colony formation, migration and invasion of colonic cell ***3 is orally available to animals and suppresses tumor growth and metastasis in a dosage form compatible to clinical applications. Importantly, it doesn′t cause significant toxicity at several times of an effective dose. CONCLUSION LL1 inhibits tumor growth and metastasis by blocking STAT3 signaling pathway. LL1 could be a promising therapeutic drug candidate for colorectal cancer by inhibiting the STAT3 activation.

Introduction: Buschke Lowenstein Tumor (BLT) is a tumor, belonging to the group of verrucous carcinomas. It is endowed with an unpredictable potential of recurrence and degeneration. The aim of the work is to present the anatomical and therapeutic characteristics of BLT at the University Hospital of National Reference of N’Djamena in Chad. Patients and Method: Prospective descriptive study of 13 patients with BLT, from May 2009 to April 2019 in the department of urology was included. The patients more than 15, carriers of TBL located at: external genitalia, perineum and anal margin whose clinical, paraclinical assessment allowed the diagnosis of BLT, management and follow-up at the said service. Respect for the patient’s identity and consent, the patient’s consent is obtained and the anonymity of the photographs is observed. Result: A total of 13 patients had BLT, 11 males and 2 females. The average age of the patients was 32.5 years. The lesions were cauliflower-like and located on the vulva (n = 2), the penis, the perineum, the anorectal region in men (n = 11). Serology was positive for: HIV (n = 3), chlamydia (n = 4). The management was surgical by cauterization and excision. Cystoscopy and rectoscope were normal. The cure was obtained in all patients. Two patients had a recurrence at 5 months requiring a second successful cure. Conclusion: BLT is a frequent pathology in the urology department of the University Hospital of National Reference of N’Djamena in Chad. The surgical treatment by “cauterization exeresis” gives a better treatment.

Despite the low incidence, squamous cell carcinoma(SCC) remains the most common scrotal malignancy featuring a propensity for recurrence and metastasis. In recent years there has been a significant change in the epidemiology of scrotal SCC. Surgery is the mainstay of treatment for resectable disease. Sentinel lymph node dissection similar to algorithm for patients with penile SCC can reduce the morbidity of routine lymph node dissection. Emerging treatments for advanced and metastatic SCC are at the cusp of significantly changing management of this disease. We have performed a comprehensive review of scrotal SCC with a focus on these topics.

Cancers in the Elderly Seen in Anatomical Pathology Laboratory in Lomé, Togo

Background: The elderly cancers are worldwide a major public health issue. Their exact incidence is unknown in Togo. The purpose of this study was to describe epidemiological and histological data of cancers in the elderly in Togo. Materials and Methods: This was a cross-sectional and descriptive study of cases of elderly cancers diagnosed at the laboratory of pathology of the university teaching hospital of Lomé from 1995 to 2014 (20 years). Cases of examination on samples of the elderly (biopsy, excision, surgical specimens) were collected from the data records of that laboratory. Results: Overall, 792/5200 cases of elderly cancer were collected representing 15.2% of all cancers diagnosed in the laboratory. The annual incidence was 39.6 cases. Patient age ranged from 65 to 103 years old, with a mean of 68.5 ± 4 years old. The sex ratio (M/F) was 1.3. At pathological level, the study material included pieces (n=290 cases;36.6%) and biopsies (n=502 cases;63.4%). Cancers located preferably in prostate in men (38%), and in women the most frequent locations were cervix (16.2%) and breast (15.8%). We noticed four histological groups: carcinomas (n = 706 cases;89.1%), sarcomas (n = 43 cases;5.5%), lymphoma (n = 34 cases;4.3%) and melanoma (n = 9 cases;1.1%). Carcinomas were dominated by adenocarcinomas (50.1%) and squamous cell carcinomas (33.8%);Kaposi’s sarcoma (32.6%) was the most common histological type in sarcomas. Conclusion: Our results showed that the elderly cancers were frequent in Togo, locating mostly in uterus cervix and breast in female, and prostate in men. This study could help to advocate the establishment of a cancer registry in Togo.

Prevalence and Etiologies of Obstructive Renal Failure in the Nephrology Department of the University Hospital Center of Point G, Bamako, Mali

Introduction: Obstructive renal failure (ORF) or obstructive uropathy is defined by the simultaneous presence of impaired renal function and urinary excretory tract dilatation on medical imaging. It accounts for 2% to 10% of the causes of acute renal failure (ARF). Objective: To determine the prevalence and etiologies of ORF in the nephrology department of the University Hospital Center of Point G. Methodology: This was a descriptive study with retrospective data collection conducted from January 1, 2017 to June 30, 2018. All patients hospitalized with renal failure due to urinary tract obstruction were included. The parameters studied were age, gender, etiology, type of renal impairment, procedures performed, surgical outcomes of the procedure, pre-and post-management creatinine levels. Information on these clinical and paraclinical variables was collected from individual hospitalization and patient follow-up records. Results: Among 1133 patients, 83 had obstructive renal failure, a prevalence of 7.32%. The sex ratio was 1.86. The mean age was 48.99 ± 6.81 years with extremes of 7 and 102 years. Signs suggestive of lower urinary tract obstruction were, in order of frequency: dysuria (50.6%), urinary burns (44.6%), urinary frequency (44.6%), pollakiuria (44.6%), macroscopic hematuria (25.3%), low back pain (21.6%), total anuria (18.1%). The etiologies were dominated by lithiasis 36 cases (43.4%), tumours 32 cases (38.55%) followed by sequelae of bilharziasis 12 cases (14.50%). Conclusion: Obstructive renal failure is becoming more and more common. The etiology is essentially of lithiasic and tumor origin. It must be treated early to allow total or partial recovery of renal function.

Regenerative medicine based applications to combat stress urinary incontinence

Stress urinary incontinence(SUI), as an isolated symptom, is not a life threatening condition. However, the fear of unexpected urine leakage contributes to a significant decline in quality of life parameters for afflicted patients. Compared to other forms of incontinence, SUI cannot be easily treated with pharmacotherapy since it is inherently an anatomic problem. Treatment options include the use of bio-injectable materials to enhance closing pressures, and the placement of slings to bolster fascial support to the urethra. However, histologic findings of degeneration in the incontinent urethral sphincter invite the use of tissues engineering strategies to regenerate structures that aid in promoting continence. In this review, we will assess the role of stem cells in restoring multiple anatomic and physiological aspects of the sphincter. In particular, mesenchymal stem cells and CD34+cells have shown great promise to differentiate into muscular and vascular components,respectively. Evidence supporting the use of cytokines and growth factors such as hypoxia-inducible factor1-alpha, vascular endothelial growth factor, basic fi-broblast growth factor, hepatocyte growth factor and insulin-like growth factor further enhance the viability and direction of differentiation. Bridging the benefits of stem cells and growth factors involves the use of synthetic scaffolds like poly(1,8-octanediol-co-citrate)(POC) thin films. POC scaffolds are synthetic, elastomeric polymers that serve as substrates for cell growth,and upon degradation, release growth factors to the microenvironment in a controlled, predictable *** combination of cellular, cytokine and scaffold elements aims to address the pathologic deficits to urinary incontinence, with a goal to improve patient symptoms and overall quality of life.
