Allergic Phenotypes and Sarcopenia:Evidence from Observational Studies and Mendelian Randomization Analysis

Commonly afected in early-life population,the impact of allergic phenotypes on mid-or late-life health is less *** study is to explore the association of allergic phenotypes including atopic dermatitis(AD),asthma,eosinophils count(EC),and *** conducted observational studies and mendelian randomization(MR)analysis based on UK Biobank(UKB),the China Health and Retirement Longitudinal Study(CHARLS)and data from genome-wide association study(GWAS).Based on the UKB data,AD,asthma and EC were positively correlated with pre-sarcopenia and decreased skeletal muscle mass index and hand grip in fully adjusted *** and EC were signifcantly associated with sarcopenia while AD was marginally associated(p=0.095).Based on the CHARLS cohort,asthma signifcantly added 109.4%risk for pre-sarcopenia in adjusted model(relative risk=2.094;p=0.002),*** asthma(β=0.100,p=0.006)and EC(β=0.023,p=0.017)exerted signifcantly casual efects on ***,as for sarcopenia,merely EC exhibited a signifcantly casual efect(β=0.005,p=0.048).Signifcant casual efects of AD(β=–0.027,p=0.003),asthma(β=–0.029,p=0.027)and EC(β=–0.041,p<0.001)on decreased appendicular lean mass(ALM)were observed using the inverse-variance weighted method and the Mendelian randomization pleiotropy residual sum and outlier(MR-PRESSO)*** results revealed a contributory role of AD,asthma and EC on sarcopenia,especially in terms of decreased ALM,an indicator for sarcopenia *** fndings of our study will raise the awareness of preventing aging-related disorders or geriatric syndromes among allergic populations.

The Association between Sugar-Sweetened Beverages and Hair Loss in Young Women

The association between sugar-sweetened beverages （SSBs） intake and hair loss condition might give scientific support for better dietary practices to prevent hair loss. We aimed to explore the association of SSBs intake with the hair loss condition in women. This online cross-sectional study， conducted from January to April 2022， recruited 1169 women aged 18 - 45 years from 31 provinces on mainland of China. We used the updated version of the 15-item Beverage Intake Questionnaire （BEVQ-15） to evaluate the usual beverage consumption over the past month and the Hair-Shedding Visual Scale for Asian Women （HSVS-A） to evaluate the extent of hair shedding. We utilized binary logistic regression to assess the association between SSBs intake and hair loss. Among 1169 female participants with an average age of 27.2， there were 211 （18.0%） participants with hair loss. Compared with participants who never drank SSBs， those who drank more than 7 times/week or 1000 mL/weeks were more likely to have hair loss， with an adjusted odds ratio （OR） of 2.25 （95% confidence interval [CI] = 1.32，3.83） or 2.22 （95% CI = 1.28，3.85）. Furthermore， the mediation effect of anxiety status on the association between SSBs and female hair loss was observed， with a significant indirect effect (βindirect =0.0199， 95% CI = 0.0032，0.0418). In conclusion， our study showed that women who consumed higher amounts of SSBs had an increased risk of hair loss， which might attract more attention from young women and benefit from reducing SSBs consumption. Further longitudinal and interventional studies are still needed to confirm the current association and provide evidenced-based health education.

Approximately 2.5% of the global population experiences allergic reactions to seafood， making it one of the most prevalent and life-threatening allergies. Seafood allergy can lead to the disruption of the intestinal barrier， possibly due to abnormalities in intestinal glycosylation. In this study， the mechanisms underlying seafood allergy were explored through the lens of intestinal glycobiology. Mice were first sensitized with tropomyosin， resulting in significant increases in allergy symptom scores and specific antibody and Th2 cytokine levels. Intestinal damage was confirmed by histopathology assessments and levels of diamine oxidase and claudin-1. Moreover， alterations in glycosylated proteins within the jejunum were analyzed using high-throughput mass spectrometry and the pGlyco3.0 search engine， yielding 2283 glycosylation peptides corresponding to 655 unique glycosylation sites on 399 proteins in precision N-glycoproteomics. Differential expression and enrichment analyses revealed that differentially expressed glycoproteins play significant roles in the extracellular matrix (ECM)-receptor interaction pathway and focal adhesion pathway. In conclusion， tropomyosin sensitization leads to intestinal glycome changes， accompanied by remodeling of the intestinal ECM. Our research establishes an essential theoretical basis for targeting the intestinal glycome and ECM remodeling in a precise and fine-tuned manner for the treatment of food allergies.

Background: Chronic spontaneous urticaria (CSU) is an autoimmune skin disorder that lasts for >6 weeks and may last for years. It is a disabling skin disease that impairs quality of life. Set-up treatment with antihistamines, immunosuppressives, immune modulators and lately Omalizumab are expensive or have significant side effects. In this retrospective study, we describe our experience with the use of hydroxychloroquine (HCQ) as a maintenance therapy for those with severe forms of CSU after Corticosteroids (C) induction phase. Patients and Methods: 16 adult patients (aged 44 ± 7) with severe CSU for 5 ± 1 months, were included in the study. Eight patients had attacks of angioneurotic oedema. Their previous treatments were antihistaminic and short-courses of C. Results: After 2 weeks of remission with C and HCQ 200 mg twice daily, the dose of C was tapered down and discontinued by the end of the first month. The seven days Urticaria Activity Score decreased from 30 ± 3 to 6 ± 1 by the first month and remained low at 3 ± 1 by the end of 2 years of follow-up. Moreover, IgE levels and CRP had similar trends. Remission persisted after 37 ± 9 months of follow-up. Conclusion: HCQ is a safe, efficacious and inexpensive drug for the treatment of CSU.

Epimedin B(EB)is one of the main flavonoid ingredients present in Epimedium brevicornum Maxim.,a traditional herb widely used in *** previous study showed that EB was a stronger inducer of melanogenesis and an activator of tyrosinase(TYR).However,the role of EB in melanogenesis and the mechanism underlying the regulation remain ***,as an extension to our previous investigation,we provide comprehensive evidence of EB-induced pigmentation in vivo and in vitro and elucidate the melanogenesis mechanism by assessing its effects on the TYR family of proteins(TYRs)in terms of expression,activity,and *** results showed that EB increased TYRs expression through microphthalmia-associated transcription factor-mediated p-Akt(referred to as protein kinase B(PKB))/glycogen synthase kinase 3β(GSK3β)/β-catenin,p-p70 S6 kinase cascades,and protein 38(p38)/mitogen-activated protein(MAP)kinase(MAPK)and extracellular regulated protein kinases(ERK)/MAPK pathways,after which EB increased the number of melanosomes and promoted their maturation for melanogenesis in melanoma cells and human primary melanocytes/skin ***,EB exerted repigmentation by stimulating TYR activity in hydroquinone-and N-phenylthiourea-induced TYR inhibitive models,including melanoma cells,zebrafish,and ***,EB ameliorated monobenzone-induced depigmentation in vitro and in vivo through the enhancement of TYRs stability by inhibiting TYR misfolding,TYR-related protein 1 formation,and retention in the endoplasmic reticulum and then by downregulating the ubiquitination and proteolysis *** data conclude that EB can target TYRs and alter their expression,activity,and stability,thus stimulating their pigmentation function,which might provide a novel rational strategy for hypopigmentation treatment in the pharmaceutical and cosmetic industries.

Objective:Vulvar lichen planus(VLP),especially erosive VLP(EVLP),seriously affects women’s physiological function and quality of life.A retrospective analysis of erosive vulvar lichen planus(EVLP)was performed to raise awareness on this chronic,deforming,and painful vulvar ***:This observational retrospective study involved 40 patients with EVLP among 1,368 patients with vulvar disease from 2017 to *** patients’vulvar clinical features and extravulvar mucosal involvement were *** patients’pain and quality of life were evaluated with the numeric rating scale and the dermatology Life Quality Index(DLQI),*** characteristics were reported using frequencies and proportions for categorical *** variables are expressed as mean±standard *** association between NRS score for pain and DLQI score was analyzed by Pearson’s correlation ***:EVLP accounted for 2.9%(40/1,368)of all cases of vulvar disease,and the median age at onset was 42 *** mostly occurred on the labia minora and ***(20/40)of the patients had extravulvar mucosal ***(42.5%)patients had oral lichen planus,and seven(17.5%)patients were diagnosed with vulvovaginal-gingival *** numeric rating scale score showed that 47.5%(19/40)of patients experienced mild pain,whereas 12.5%(5/40)described the pain as ***-one patients(77.5%,31/40)had dyspareunia,and nine patients had aggravated pain after sexual *** mean DLQI score was 9.1±6.0,with 37.5%(15/40)of patients indicating that EVLP had severely affected their *** numeric rating scale score for pain was significantly correlated with the DLQI score(P=0.007).Conclusion:EVLP can affect the quality of women’s lives and cause severe deformations and painful architectural *** description of the characteristics of EVLP in this report will benefit the diagnosis and early treatment of this disease.

Objective:Adalimumab(ADA)is expensive and rarely used among ***,its cost for moderate to severe chronic plaque psoriasis can now be reimbursed since its addition to the National Reimbursement Drug List(NRDL)in January ***,we conducted this study to estimate the medical cost of ADA and its influencing factors in patients with psoriasis using ADA and thus gain a better understanding of the effect of the NRDL adjustment in ***:This retrospective study evaluated the medical cost of ADA before and after the addition of ADA to the NRDL(2018—2022)among 320 patients in a large professional dermatology specialty *** analysis,Welch’s analysis of variance,and multiple regression analysis were used to evaluate the medical cost and influencing ***:Before ADA was added to the NRDL,patients’mean total cost and ADA cost were¥24,243.56±16,346.53 and¥20,391.11±14,440.69,*** ADA became covered by the NRDL,the above 2 expenses decreased to¥10,461.19±7,554.66 and¥8,775.27±7,092.16,*** significant decrease in the cost of ADA resulted in a significant 110.5-times increase in ADA *** significant differences were found in the total cost,patient type(outpatient/inpatient),insurance status(yes/no),reimbursement status(before/after),and *** insurance reimbursement(P<0.001,β=-0.468,before reimbursement=1,after reimbursement=2)was the most significant influencing factor,followed by patient type(P=0.018,β=-0.215,outpatient=1,inpatient=2)and age(P=0.0046,β=-0.174).Conclusion:Medical insurance reimbursement is the most important factor affecting patients’total cost of ADA by reducing the economic burden,enhancing the availability of ADA,and stimulating the need for treatment.
