Chronic hepatitis B virus(HBV)infection is one of the major health care burdens throughout the *** chronicity of HBV infection is supported by the intracellular persistence of a multicopy viral episome called covalently closed circular DNA(cccDNA),which is the bona fide viral transcription template and resistant to currently available antivirals.1 Therefore,noncytolytic elimination of cccDNA is considered as the“holy grail”for a cure of hepatitis B,and the metabolism and transcription regulation of cccDNA have become a hot topic in basic,translational,and clinical HBV research.

The ever-growing research on lymphatic biology has clearly identified lymphatic vessels as key players that maintain human health through their functional roles in tissue fluid homeostasis,immunosurveillance,lipid metabolism and *** is therefore not surprising that the list of human diseases associated with lymphatic malfunctions has grown larger,including issues beyond lymphedema,a pathology traditionally associated with lymphatic drainage ***,the discovery of factors and pathways that can promote optimal lymphatic functions may offer new therapeutic *** evidence indicates that aside from biochemical factors,biomechanical signals also regulate lymphatic vessel expansion and functions ***,we review how mechanical forces induced by fluid shear stress affect the behavior and functions of lymphatic vessels and the mechanisms lymphatic vessels employ to sense and transduce these mechanical cues into biological signals.

The mammalian target of rapamycin(mTOR) is an evolutionarily conserved Ser/Thr protein kinase with essential cellular function via processing various extracellular and intracellular inputs. Two distinct multi-protein m TOR complexes(mTORC), m TORC1 and m TORC2, have been identified and well characterized in eukaryotic cells from yeast to human. Sin1, which stands for ■,also known as mitogen-activated protein kinase■ (MAPKAP)1, is an evolutionarily conserved adaptor protein. Mammalian Sin1 interacts with many cellular proteins, but it has been widely studied as an essential component of m TORC2, and it is crucial not only for the assembly of m TORC2 but also for the regulation of its substrate specificity. In this review, we summarize our current knowledge of the structure and functions of Sin1, focusing specifically on its protein interaction network and its roles in the m TOR pathway that could account for various cellular functions of m TOR in growth,metabolism, immunity and cancer.

AIM:To elucidate the molecular mechanisms underlying hepatitis B virus(HBV)occult infection of genotype ***:A total of 10 types of hepatitis B surface antigen(HBs Ag)variants from a Korean occult cohort were *** a complete HBV genome plasmid mutated such that it does not express HBs Ag and plasmid encoding,each HBs Ag variant was transiently co-transfected into Hu H-7 *** secretion capacity and intracellular expression of the HBV virions and HBs Ags in their respective variants were analyzed using real-time quantitative polymerase chain reaction assays and commercial HBs Ag enzyme-linked immunosorbent assays,***:All variants exhibited lower levels of HBs Ag secretion into the medium compared with the wild *** particular,in eight of the ten variants,very low levels of HBs Ag secretion that were similar to the negative control were *** contrast,most variants(9/10)exhibited normal virion secretion capacities comparable with,or even higher than,the wild *** provided new insight into the intrinsic nature of occult HBV infection,which leads to HBs Ag sero-negativeness but has horizontal ***,most variants generated higher reactive oxidative species production than the wild *** finding provides potential links between occult HBV infection and liver disease ***:The presently obtained data indicate that deficiency in the secretion capacity of HBs Ag variants may have a pivotal function in the occult infections of HBV genotype C.

AIM： To investigate the correlation between expression of CD58 and severity of hepatitis B. METHODS： The level of soluble CD58 （sCD58） in serum of patients with hepatitis B was detected by enzymelinked immunosorbent assay. The level of expression of membrane CD58 molecule in PBMC was detected by direct immunofluorescence. The levels of serumal TBIL, DBIL, IBIL, ALT and AST were detected by the automated biochemistry analyzer as well. RESULTS： The levels of sCD58 in serum and membrane CD58 molecule in PBMC of patients with hepatitis B were significantly higher than that in normal controls （P 〈 0.05）. Level of CD58 was related to the levels of serumal TBIL, DBIL, IBIL, ALT and AST. CONCLUSION： The level of CD58 molecule （in both serum and PBMC form） of patients with hepatitis B is related to the degree of liver damage.

Epstein-Barr virus （EBV） is prevalent throughout the world and is associated with several malignant diseases in humans. Latent membrane protein 2 （LMP2） of EBV plays a crucial role in the pathogenesis of EBV-associated tumors; therefore, LMP2 has been considered to be a potential immunodiagnostic and immunotherapeutic target. A multi-epitope-based antigen is a promising option for therapeutic vaccines and diagnoses of such malignancies. In this study, we systematically screened cytotoxic T lymphocyte （CTL）, helper T cell （Th） and B-cell epitopes within EBV-LMP2 using bioinformatics. Based on the screen, two peptides rich in overlapping epitopes of both T cells and B cells were selected to construct a plasmid containing the sequence for a chimeric multi-epitope protein referred to as EBV-LMP2m, which is composed of LMP2aa195-232 and LMP2aa419-436. The EBV-LMP2m protein was expressed in E. coil BL21 （DE3） after prokaryotic codon optimization. Inoculation of the purified chimeric antigen in BALB/c mice induced not only high levels of specific IgG in the serum and secretory IgA in the vaginal mucus but also a specific CTL response. By using purified EBV-LMP2m as an antigen, the presence of specific IgG in the serum specimens of 202 nasopharyngeal carcinoma （NPC） patients was effectively detected with 52.84% sensitivity and 95.40% specificity, which represents an improvement over the traditional detection method based on VCA-IgA （60.53% sensitivity and 76.86% specificity）. The above results indicate that EBV-LMP2m may be used not only as a potential target antigen for EBV-associated tumors but also a diagnostic agent for NPC patients.
