Dietary supplementation with citrus peel extract in transition period improves rumen microbial composition and ameliorates energy metabolism and lactation performance of dairy cows

Background During the transition period,excessive negative energy balance(NEB)lead to metabolic disorders and reduced milk *** microbes are responsible for resolving plant material and producing volatile fatty acids(VFA),which are the primary energy source for *** this study,we aimed to investigate the effect of citrus peel extract(CPE)supplementation on rumen microbiota composition,energy metabolism and milk performance of peri-partum dairy *** Dairy cows were fed either a basal diet(CON group)or the same basal diet supplemented with CPE via intragastric administration(4 g/d,CPE group)for 6 weeks(3 weeks before and 3 weeks after calving;n=15 per group).Samples of serum,milk,rumen fluid,adipose tissue,and liver were collected to assess the effects of CPE on rumen microbiota composition,rumen fermentation parameters,milk performance,and energy metabolic status of dairy *** CPE supplementation led to an increase in milk yield,milk protein and lactose contents,and serum glucose levels,while reduced serum concentrations of non-esterified fatty acid,β-hydroxybutyric acid,insulin,aspartate aminotransferase,alanine aminotransferase,and haptoglobin during the first month of *** supplemen-tation also increased the content of ruminal *** to the CON group,the abundance of Prevotellaceae,Methanobacteriaceae,Bacteroidales_RF16_group,and Selenomonadaceae was found increased,while the abun-dance of Oscillospiraceae,F082,Ruminococcaceae,Christensenellaceae,Muribaculaceae UCG-011,Saccharimona-daceae,Hungateiclostridiaceae,and Spirochaetaceae in the CPE group was found *** adipose tissue,CPE supplementation decreased lipolysis,and inflammatory response,while increased insulin *** the liver,CPE supplementation decreased lipid accumulation,increased insulin sensitivity,and upregulated expression of genes involved in *** Our findings suggest that CPE supplementation during the peripartum period

Recombination and Mutation of a New Chinese Strain of HP-PRRSV SCMY2023

In this study, RT-PCR was performed on lung tissue samples from sick pigs in a suspected outbreak of porcine reproductive and respiratory syndrome (PRRS) at a pig farm in Mianyang City, Sichuan Province, China. Positive samples were inoculated into Marc-145 cells to observe lesions. The Marc-145 cells with cytopathic lesions were identified by indirect immunofluorescence. The whole genome sequences of the isolated and purified strains were amplified by RT-PCR and analyzed for homology and genetic evolution. A strain of porcine reproductive and respiratory syndrome virus (PRRSV), named SCMY2023 (GenBank No. PQ179742), was successfully isolated. SCMY2023 has a genome length of 15,321 base pairs (without a poly A tail). Nucleotide and amino acid homology analyses suggest that this strain belongs to Lineage 8, a variant of the highly pathogenic porcine reproductive and respiratory syndrome virus (HP-PRRSV) prevalent in China. Recombination and genetic evolution analyses indicate that this isolate is a PRRSV variant that recombined with HuN-ZZ (Lineage 8, 98.79% homology) on the backbone of the SCSN2020 strain (Lineage 8, 99.35% homology) in the recombination region from 4407 to 13,107 nucleotides (ORF1a to ORF3). In-depth study of the genetic recombination of this isolate can provide a reference for the prevention and control of PRRS.

Treatment of liver hydatidosis:How to treat an asymptomatic carrier?

Liver hydatidosis is the most common clinical presentation of cystic echinococcosis(CE).Ultrasonographic mass surveys have demonstrated the true prevalence,including the asymptomatic characteristic of the majority of cases,providing new insight into the natural history of the *** raises the question of whether to treat or not to treat these patients,due to the high and unsuspected prevalence of *** high rate of liver/lung frequencies of cyst localization,the autopsy findings,and the involution of cysts demonstrated in long time follow-up of asymptomatic carriers contribute to this *** decision to treat an asymptomatic patient by surgery,albendazole,or puncture aspiration injection and reaspiration or to wait and watch,is based on conflicting reports in the literature,the lack of complications in untreated patients over time,and the spontaneous disappearance and involution of *** these points contribute to difficulties of individual clinical *** patients should be informed of the reasons and the risks of watchful/waiting without treatment,the possibility of complications,and the risks of the other *** more information on the natural history of liver hydatidosis is acquired,selection of the best treatment will be come *** this knowledge it would be very difficult to establish definitive rules of *** present,it is possible to manage these patients over time and to wait for the best moment for *** studies must be conducted to achieve this objective.

Comparison of three fluorescence labeling and tracking methods of endothelial progenitor cells in laser-injured retina

AIM： To compare three kinds of fluorescent probes for in vitro labeling and in vivo tracking of endothelial progenitor cells（EPCs） in a mouse model of laser-induced retinal ***： EPCs were isolated from human umbilical cord blood mononuclear cells and labeled with three different fluorescent probes： 5-（and-6）-carboxyfluorescein diacetate succinimidyl ester（CFSE）, 1,1′-dilinoleyl-3,3,3′,3′-tetramethylindo-carbocyanine perchlorate linked acetylated low-density lipoprotein（Di I-Ac LDL）, and green fluorescent protein（GFP）. The fluorescent intensity of EPCs was examined by confocal microscopy. Survival rate of labeled EPCs was calculated with trypan blue staining, and their adhesive capability was assessed. A mouse model of retinal injury was induced by laser, and EPCs were injected into the vitreous cavity. Frozen section and fluorescein angiography on flat-mounted retinal samples was employed to track the labeled EPCs in ***： EPCs labeled with CFSE and Di I-Ac LDL exhibited an intense green and red fluorescence at the beginning; the fluorescence intensity decreased gradually to 20.23% and 49.99% respectively, after 28 d. On the contrary, the florescent intensity of GFP-labeled EPCs increased in a time-dependent manner. All labeled EPCs showed normal morphology and no significant change in survival and adhesive capability. In the mouse model, transplantation of EPCs showed a protective effect against retinal injury. EPCs labeled with CFSE and Di I-Ac LDL were successfully tracked in mice during the development of retinal injury and repair; however, GFP-labeled EPCs were not detected in the laser-injured mouse ***： The three fluorescent markers used in this study have their own set of advantages and disadvantages. CFSE and Di I-Ac LDL are suitable for short-term EPClabeling, while GFP should be used for long-term labeling. The choice of fluorescent markers should be guided by the purpose of the study.

In-depth analysis of the genome of Trypanosoma evansi, an etiologic agent of surra

Trypanosoma evansi is the causative agent of the animal trypanosomiasis surra, a disease with serious economic burden worldwide. The availability of the genome of its closely related parasite Trypanosoma brucei allows us to compare their genetic and evolutionarily shared and distinct biological features. The complete genomic sequence of the T. evansi YNB strain was obtained using a combination of genomic and transcriptomic sequencing, de novo assembly, and bioinformatic analysis. The genome size of the T. evansi YNB strain was 35.2 Mb, showing 96.59% similarity in sequence and 88.97% in scaffold alignment with T. brucei. A total of 8,617 protein-coding genes, accounting for 31% of the genome, were predicted. Approximately 1,641 alternative splicing events of 820 genes were identified, with a majority mediated by intron retention, which represented a major difference in post-transcriptional regulation between T. evansi and T. brucei. Disparities in gene copy number of the variant surface glycoprotein, expression site-associated genes, microRNAs, and RNA-binding protein were clearly observed between the two parasites. The results revealed the genomic determinants of T. evansi, which encoded specific biological characteristics that distinguished them from other related trypanosome species.

Aniba rosaeodora essential oil(RO) has been traditionally used in natural medicine as a substitute for antibiotics due to its notable antidepressant and antibacterial properties. Salmonella, a prevalent pathogen in foodborne illnesses, presents a major challenge to current antibiotic treatments. However, the antibacterial efficacy and mechanisms of action of RO against Salmonella spp. remain underexplored. This study aims to elucidate the chemical composition of RO, evaluate its antibacterial activity and mechanisms against Salmonella in vitro, and further delineate its anti-inflammatory mechanisms in vivo during Salmonella infection. Gas chromatography-mass spectrometry(GC-MS) was utilized to characterize the chemical constituents of RO. The antibacterial activity of RO was assessed using minimal inhibitory concentration(MIC) and time-kill assays. Various biochemical assays were employed to uncover the potential bactericidal mechanisms. Additionally, mouse and chick models of Salmonella infection were established to investigate the prophylactic effects of RO treatment. RO exhibited significant antibacterial activity against both Gram-positive and Gram-negative bacteria, with an MIC of 4 mg·mL^(-1) for Salmonella spp. RO treatment resulted in bacterial damage through the disruption of lipid and purine metabolism. Moreover, RO reduced injury and microbial colonization in infected mice and chicks. RO treatment also modulated the host inflammatory response by inhibiting proinflammatory pathways. In conclusion, our findings demonstrate that RO is effective against Salmonella infection, highlighting its potential as an alternative to antibiotics for antibacterial therapy.

Induction of Apoptosis in the Immature Mouse Testes by a Mixture of Melamine and Cyanuric Acid

The toxicity of melamine has attracted much attention since the recent outbreaks of renal injury in pets and infants. Previous studies indicated that melamine by itself had low toxicity, whereas a mixture of melamine and cyanuric acid （M＋CA） could cause serious renal damage. At present, most researches on the toxicity of M＋CA are focused on the kidney. However, little is known about the adverse effects of this mixture on the reproductive system. In the present study, the toxicity of M＋CA to testes was investigated. Immature male mice were orally dosed with 0, 0.6, 3, and 15 mg kg-1 d-1 of a 1：1 M＋CA for 28 d. Pathological changes occurred in germ cells, such as loose arrangement, reduced numbers and karyopyknosis, indicating that this mixture was toxic to spermatogenesis. Compared with the control group, the TUNEL- positive germ cells increased significantly and the ratio of Bcl-2 to Bax, total antioxidant capacity and superoxide dismutase activity decreased significantly in the 3 and 15 mg kg-1 d- M＋CA treated group, while the activities of caspase-3, caspase- 8 and caspase-9 remained unchanged. The results suggest that M＋CA can induce apoptosis in the mice testes. The downregulation of Bcl-2/Bax and oxidative stress may play a pivotal role in the induction of apoptosis by M＋CA in mice testes.
